Contrary to globular proteins, intrinsically disordered proteins (IDPs) lack a folded structure and they do not lose solubility at elevated temperatures. Although this should also be true at low temperatures, cold stability of IDPs has not been addressed in any scientific work so far. As direct characterization of cold-denaturation is difficult, we approached the problem through a freezing-induced loss-of-function model of globular-disordered functional protein pairs (m-calpain-calpastatin, tubulin-Map2c, Hsp90-ERD14). Our results affirm that in contrast with globular proteins IDPs are resistant to cold treatment. The theoretical and functional aspects of this observation are discussed.  相似文献

Polyploidy is a key factor in the evolution of higher plants and plays an important role in the variation of plant genomes, leading to speciation in some cases. During polyploidisation, different balancing processes take place at the genomic level that can promote variation in nuclear DNA content. We estimated genome size using flow cytometry in 84 populations of 67 Artemisia species and one population of Crossostephium chinense. A total of 73 sequences of nrDNA ITS and 3′‐ETS were newly generated and analysed, together with previously published sequences, to address the evolution of genome size in a phylogenetic framework. Differences in 2C values were detected among some lineages, as well as an increase of genome size heterogeneity in subgenera whose phylogenetic relationships are still unclear. We confirmed that the increase in 2C values in Artemisia polyploids was not proportional to ploidy level, but 1Cx genome size tended to decrease significantly when high ploidy levels were reached. The results lead us to hypothesise that genome size in polyploids tends to a maximum as it follows saturation behaviour, in agreement with the Michaelis–Menten model. We tested different arithmetic functions with our dataset that corroborated a non‐linear relationship of genome size increase in polyploids, allowing us to suggest a theoretical upper limit for the DNA content of this genus.  相似文献

Extremophilic microalgae are unexplored as a source of pharmaceuticals despite the fact that its biomass can be produced at large scale with low risk of contamination. A significant amount of antimicrobial activity was produced by extracts obtained from the eukaryotic acidophilic microalgae Coccomyxa onubensis in non‐polar solvents, such as hexane, diethyl ether, and chloroform or in weakly polar solvents, such as dichloromethane, against Gram‐negative and Gram‐positive bacteria, and also the yeast Candida albicans. The most effective activity was shown by chloroform extract against Escherichia coli S, Salmonella enterica, and Proteus mirabilis; hexane extract against P. mirabilis, Sa. enterica, and Ca. albicans; dichloromethane extract against Sa. enterica or diethyl ether extract against E. coli S and the Gram‐positive Staphylococcus aureus MB. The lowest minimum inhibitory concentration values were recorded against E. coli S (305 μg mL ?1) and P. mirabilis (153 μg mL ?1) (using chloroform extract) and against P. mirabilis (106 μg mL?1) (using hexane extract). Fatty acids, but not carotenoids, seem to be involved in the antimicrobial activity of this microalga. However, further biochemical and biotechnological studies must be conducted in order to characterize and purify the bioactive principles from Co. onubensis for assessing its potential as a pharmaceutical source and feasibility of production.  相似文献

Past climatic shifts have played a major role in generating and shaping biodiversity. Quaternary glacial cycles are the better known examples of dramatic climatic changes endured by ecosystems in temperate regions. Although still a matter of debate, some authors suggest that glaciations promoted speciation. Here we investigate the effect of past climatic changes on the diversification of the ground‐dwelling spider genus Harpactocrates, distributed across the major mountain ranges of the western Mediterranean. Concatenated and species‐tree analyses of multiple mitochondrial and nuclear loci, combined with the use of fossil and biogeographic calibration points, reveal a Miocene origin of most nominal species, but also unravel several cryptic lineages tracing back to the Pleistocene. We hypothesize that the Miocene Climatic Transition triggered major extinction events in the genus but also promoted its subsequent diversification. Under this scenario, the Iberian mountains acted as an island‐like system, providing shelter to Harpactocrates lineages during the climate shifts and favouring isolation between mountain ranges. Quaternary glacial cycles contributed further to the diversification of the group by isolating lineages in peripheral refugia within mountain ranges. In addition, we recovered some unique biogeographic patterns, such as the colonization of the Alps and the Apennines from the Iberian Peninsula.  相似文献

Summary Immune cells producing antibodies to chicken photoreceptor membranes were fused with myeloma cells and supernatants of the resulting hybridoma cells were used to test various types of photoreceptor cells in the chicken retina by means of immunocytochemistry. A polyclonal antibody raised against the protein component of bovine rhodopsin was also used.Outer segments of various photoreceptor cells were labelled by the following antibodies: rods were positive with the anti-rhodopsin antibody, both members of the double cones were stained by supernatant A1, while one type of single cones (designated as type A) was specifically labelled by supernatants A5, B3 and D6. The other type of single cones (type B) reacted with anti-rhodopsin and supernatant A1. The results indicate that there are distinct differences in the molecular structure of various photoreceptor outer segments.  相似文献

Recombinant baculoviruses could be used as biological insecticides through the introduction and expression of exogenous genes (such as those coding for proteins) that interfere with metabolism, metamorphosis (toxins, hormones, and enzymes), and immune system of the insects. The CrV1 secreted protein of Cotesia rubecula polydnavirus (PDV) is responsible for the actin depolymerisation in haemocytes and the abolishment of immune functions such as phagocytosis and cell spreading, thus allowing the successful embryonic development of the parasitoid wasp. CrV1 cDNA was cloned into C6 strain of Autographa californica multiple nucleopolyhedrovirus (AcMNPV-C6-CrV1) under p10 promoter to construct a recombinant virus. The recombinant virus was then tested against the insect pest Spodoptera exigua. The recombinant virus expressing CrV1 protein showed significantly lower LC50 and shorter LT50 as compared with the AcMNPV-C6 wild-type virus. The potential of recombinant baculoviruses expressing PDV genes in relation to their virulence is discussed.  相似文献
