A photomixotrophic tissue culture system for Nicotiana plumbaginifolia and N. tabacum has been developed in which a primary symptom (bleching) of the inhibition of photosynthetic electron transport by herbicides can be observed. Photomixotrophic cultures were initiated and maintained in the light on medium containing 0.2–0.3% sucrose or glucose (low-sugar medium) as sole source of respirable carbohydrate. The usual medium for growing heterotrophic cultures contains 2–3% sucrose or glucose (high-sugar medium). Callus grown on low-sugar medium achieved a fresh weight three to four times greater in the light than in the dark and reached about half that of callus grown on high-sugar medium. Carbon-dioxide fixation rates were an order of magnitude higher in cultures grown on low-sugar medium in the light than in those grown on high-sugar medium or in any of the dark-grown cultures. The lightdependent growth and CO2-fixation rates of cultures grown on low-sugar medium indicated that a major proportion of the weight increase resulted from photosynthesis. Under these photomixotrophic conditions it was found that a number of photosystem-II herbicides, at concentrations which inhibit photosynthetic electron transport, also inhibited the light-dependent component of callus growth, and caused bleaching. An in vitro 3D model was developed utilizing a synthetic microgravity environment to facilitate studying the cell interactions. 2D monolayer cell culture models have been successfully used to understand various cellular reactions that occur in vivo. There are some limitations to the 2D model that are apparent when compared to cells grown in a 3D matrix. For example, some proteins that are not expressed in a 2D model are found up-regulated in the 3D matrix. In this paper, we discuss techniques used to develop the first known large, free-floating 3D tissue model used to establish tumor spheroids. The bioreactor system known as the High Aspect Ratio Vessel (HARVs) was used to provide a microgravity environment. The HARVs promoted aggregation of keratinocytes (HaCaT) that formed a construct that served as scaffolding for the growth of mouse melanoma. Although there is an emphasis on building a 3D model with the proper extracellular matrix and stroma, we were able to develop a model that excluded the use of matrigel. Immunohistochemistry and apoptosis assays provided evidence that this 3D model supports B16.F10 cell growth, proliferation, and synthesis of extracellular matrix. Immunofluorescence showed that melanoma cells interact with one another displaying observable cellular morphological changes. The goal of engineering a 3D tissue model is to collect new information about cancer development and develop new potential treatment regimens that can be translated to in vivo models while reducing the use of laboratory animals.  相似文献

EDWARDS'S test for seasonality is extended to multiple peaks and troughs. It is shown how the extended statistic may be adjusted for the population at risk and for unequal lengths of time intervals in the cycle of seasons. A simulation study shows that the extended test statistic is, for sample sizes N ≧ 100, very specific in detecting the number of peaks and troughs for which it is intended. The associated method of parameter estimation is also assessed; for N ≦ 100, the amplitude of a possibly adequate simple harmonic model is estimated well, but the initial value of the phase angle is not. Estimated percentage points for the extended test statistic are tabulated, and some recommendations are offered regarding usage of this method.  相似文献

FSP27 [cell death-inducing DFFA-like effector c (CIDEC) in humans] is a protein associated with lipid droplets that downregulates the fatty acid oxidation (FAO) rate when it is overexpressed. However, little is known about its physiological role in liver. Here, we show that fasting regulates liver expression of Fsp27 in a time-dependent manner. Thus, during the initial stages of fasting, a maximal induction of 800-fold was achieved, whereas during the later phase of fasting, Fsp27 expression decreased. The early response to fasting can be explained by a canonical PKA-CREB-CRTC2 signaling pathway because: i) CIDEC expression was induced by forskolin, ii) Fsp27 promoter activity was increased by CREB, and iii) Fsp27 expression was upregulated in the liver of Sirt1 knockout animals. Interestingly, pharmacological (etomoxir) or genetic (Hmgcs2 interference) inhibition of the FAO rate increases the in vivo expression of Fsp27 during fasting. Similarly, CIDEC expression was upregulated in HepG2 cells by either etomoxir or HMGCS2 interference. Our data indicate that there is a kinetic mechanism of autoregulation between short- and long-term fasting, by which free FAs delivered to the liver during early fasting are accumulated/exported by FSP27/CIDEC, whereas over longer periods of fasting, they are degraded in the mitochondria through the carnitine palmitoyl transferase system.  相似文献

Eupatorin is a naturally occurring flavone that inhibits cell proliferation in human tumor cells. Here we demonstrate that eupatorin arrests cells at the G2-M phase of the cell cycle and induces apoptotic cell death involving activation of multiple caspases, mitochondrial release of cytochrome c and poly(ADP-ribose) polymerase cleavage in human leukemia cells. This flavonoid induced the phosphorylation of members of the mitogen-activated protein kinases and cell death was attenuated by inhibition of c-jun N-terminal kinases/stress activated protein kinases. Eupatorin-induced cell death is mediated by both the extrinsic and the intrinsic apoptotic pathways and through a mechanism dependent on reactive oxygen species generation.  相似文献

Prostaglandin E2 (PGE2), PGF2alfa and PGD2 were synthetized from [1-14C]-arachidonic acid by rat kidney medulla microsomal fraction. The formation of each prostaglandin was significantly less in female animals than in males. The rate of inactivation of [3H]-PGF2alfa by kidney cortex cytosol was almost linear with the time of incubation during the first 30 min. The production of PGF2alfa metabolite (13,14-dihydro-15-keto PGF2alfa) was higher in male rats than in females.  相似文献

The salt-sensitive humid tropical biodiesel crop, Jatropha curcas, was subjected to a 28-day exposure to salinity (0, 50, 100, and 200 mM NaCl), and activities of antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT), and peroxidase (POX), the rate of lipid peroxidation, stomatal conductance, mineral contents, and chlorophyll (Chl) content were compared to corresponding characteristics of J. cinerea, a related wild species of saline-dry areas. Biomass production decreased under the influence of 50 mM NaCl in both species, and the reduction was larger in J. curcas than in J. cinerea at the higher NaCl concentrations. In both species, stomatal conductance and transpiration decreased, leaf temperature and Na+ concentration increased under salt treatment; salinity effect was stronger in J. curcas. Chl degradation was enhanced only in J. curcas. In both Jatropha species, SOD, CAT, and POX activities increased with salinity. J. curcas showed the higher antioxidant activity than J. cinerea. Lipid peroxidation was observed only in J. curcas at concentrations above 100 mM NaCl, partially due to a greater reduction in stomatal conductance and/or the poor ROS-scavenging system. Thus, J. cinerea had more favorable characteristics to adapt to saline environments, and young J. curcas plants could adapt to salt-affected land if soil salinity was moderate (about 50 mM NaCl in solution).  相似文献

Biological data from 1125 female Atlantic wolf‐fish Anarhichas lupus were collected during 2002–2006 at their main spawning and fishing grounds in Iceland. The results demonstrated substantial annual variation in growth and maturity of female A. lupus. The fast growing females mature earlier than the slow growing ones. In addition, females mature at a larger size and greater age in warmer temperatures than colder ones. There was a strong negative relationship between temperature and growth, which may indicate that the sea temperature west of Iceland has risen above the optimum for growth of female A. lupus and thereby reduced the reproductive potential of the species.  相似文献

A library of benzofurans was prepared by solid-phase synthesis methods, and several analogues were identified as potent ligands for the estrogen receptors ER-alpha and ER-beta, with some compounds having selectivity for ER-alpha. Analogues designed to more closely mimic Raloxifene were less effective. Certain benzofurans were effective in a bone pit assay, but were characterized as agonists in a MCF-7 breast tumor cell proliferation assay.  相似文献
