This work examines the inhibitory effect of TCEP on the in vitro fibrillation of hen lysozyme at pH 2. We demonstrate that the inhibition of hen lysozyme fibrillation by TCEP follows a dose-dependent manner. Our data show that the addition of TCEP prevents α-to-β transition and promoted unfolding of lysozyme. Moreover, our findings suggested that the TCEP-induced attenuated fibrillation is associated with disulfide disruption and structural unfolding of HEWL.  相似文献

During the development of the globular embryos via heart-shaped, torpedo-shaped, and cotyledonary embryos into plantlets, contents of chlorophyll (Chl) a and b and carotenoids, and activity of ribulose-1,5-bisphosphate carboxylase (RuBPC, EC 4.1.1.39) were investigated. In the solid cultivation (SC) the contents of Chl a, Chl b, Chl a/b, and total pigments increased up to plantlet stage. In the liquid cultivation (LC), contents of Chl a, Chl b, and total pigments increased till the torpedo-shaped stage, but decreased with the further development up to plantlets stage. During SC, RuBPC activity increased up to the torpedo-shaped embryo stage, but in the LC RuPBC activity increased continuously with the progress in the developmental stages. The correlations between Chl a and RuBPC activity on the SC and LC were negative, r = −0.26 and −0.56, respectively.  相似文献

The herbicides diuron, fluridone, or sulcotrione differently reduced chlorophyll (Chl) and carotenoid (Car) contents. Four days after herbicide treatment, application of sulcotrione resulted in a Chl/Car ratio of 5.88, similar as in untreated controls; diuron resulted in ratio of 5.24, while fluridone induced a greater diminution in Car contents and yielded a final ratio of 7.02. Sulcotrione induced a more rapid decrease than fluridone did in the quantum yield of photosystem 2 (PS2) as monitored by Chl fluorescence. Measurements of DPIP reduction with isolated thylakoids indicated that sulcotrione was a more effective inhibitor of the Hill reaction in cucumber, a herbicide sensitive species, than in maize, a herbicide-insensitive species. These results are consistent with the view that inhibition of electron transport via reduction in plastoquinone contents in plants leads to the major herbicidal effect of sulcotrione in mature green tissues.  相似文献

The effects of passive integrated transponder (PIT) tagging on the growth and survival of kelp grouper Epinephelus bruneus was examined over 1 year. The overall average mass gain for the 1 year period of the abdominal cavity tagged group was higher than the dorsal muscle tagged group. No significant differences were found in overall specific growth rate and food conversion between control and treatment groups.  相似文献

Transposition studies of the transposon, En/Spm, have dealt with general aspects of the timing of the excision event with regard to DNA replication and plant development, but without describing details of the process. By following the excision events of an En transposon inserted at the a1 locus [a1-m(Au)], several features of this process can be elucidated. In progenies from reciprocal crosses between the a1-m(Au) allele containing an En insert, and a nonautonomous En allele, [a1-m(r) is a deficiency derivative of En], several features of the En at the a1-m(Au) allele can be observed taking place during ear development and during microsporogenesis. First, it has long been known that the distribution of mutant kernel phenotypes on an ear indicates that En transposes late in most of the events during ear development. Second, the phase change of En (presence and absence of activity) is observed during cob development. Third, discordant kernel phenotypes of two ears, reported herein, resulting from a reciprocal cross with the parental phenotype can be deduced to arise from the transposition of En during microsporogenesis and subsequent fertilization, leading to a discordant genotype between endosperm and embryo. The phase change and discordance lead us to conclude that these events can arise from transposition after host DNA replication. It can also be concluded that the activity of the En inserted in this a1-m(Au) allele is not limited to a specific stage or timing during plant development. Multicompetitive reactions catalyzed by lipases in organic media were used for the determination of lipase specificity towards alcohols. The competitive factors (, defined as the ratio of the kinetic powers, kcat/Km, for two substrates in competition for the enzyme active site) were estimated in a one-step experiment and a scale of specificity was easily deduced. The specificity towards the alcohol chain length and degree of substitution of seventeen commercial lipase preparations was investigated. The results show that, like fatty acids, alcohols greatly influence the reaction rates of lipase catalyzed reactions in organic solvents. Five groups of alcohol specificity are proposed after using the statistical method of principal component analysis.  相似文献
