Foraging patterns of large herbivores may give important clues as to why their life history varies depending on population density. In this landscape-scale experiment, domestic sheep Ovis aries were kept at high (80 sheep km−2 ) and low (25 sheep km−2 ) population densities during summer in high mountain pastures in Hol, Norway. We predicted an increasing use of less preferred plant species or habitat types with increasing sheep population density. Foraging behaviour was investigated by direct observations of individually marked sheep on different spatial scales, and diet composition was also assessed with microhistological analysis of faecal samples from known individuals. We found that the effects of density on foraging behaviour depended on scale and were only detected at the scale of diet choice. Use of the common grass species Deschampsia flexuosa , which provided the bulk forage (10–65% of the diet), remained constant throughout the season at low densities, but increased significantly over time at high densities. On a coarser spatial scale, neither within vegetation type nor between vegetation types, selection was affected by density, but vegetation type selection differed depending on whether the sheep were grazing or resting. The incidence of colorectal cancer (CRC) increases with age and early onset indicates an increased likelihood for genetic predisposition for this disease. The somatic genetics of tumor development in relation to patient age remains mostly unknown. We have examined the mutation status of five known cancer critical genes in relation to age at diagnosis, and compared the genomic complexity of tumors from young patients without known CRC syndromes with those from elderly patients. Among 181 CRC patients, stratified by microsatellite instability status, DNA sequence changes were identified in KRAS (32%), BRAF (16%), PIK3CA (4%), PTEN (14%) and TP53 (51%). In patients younger than 50 years (n = 45), PIK3CA mutations were not observed and TP53 mutations were more frequent than in the older age groups. The total gene mutation index was lowest in tumors from the youngest patients. In contrast, the genome complexity, assessed as copy number aberrations, was highest in tumors from the youngest patients. A comparable number of tumors from young (<50 years) and old patients (>70 years) was quadruple negative for the four predictive gene markers (KRAS-BRAF-PIK3CA-PTEN); however, 16% of young versus only 1% of the old patients had tumor mutations in PTEN/PIK3CA exclusively. This implies that mutation testing for prediction of EGFR treatment response may be restricted to KRAS and BRAF in elderly (>70 years) patients. Distinct genetic differences found in tumors from young and elderly patients, whom are comparable for known clinical and pathological variables, indicate that young patients have a different genetic risk profile for CRC development than older patients. 相似文献

Cytokines are released during T cell activation, including the potentially anti-leukemic interferon-γ (IFNγ), but also the
hematopoietic growth factor granulocyte-macrophage colony-stimulating factor (GM-CSF) that enhance proliferation and inhibit
apoptosis of acute myelogenous leukemia (AML) cells. In the present study we investigated the release of IFNγ and GM-CSF by
circulating T cells in AML patients with chemotherapy-induced cytopenia. T cells were activated with anti-CD3 plus anti-CD28
in a whole-blood assay in the presence of their natural cytokine network. We examined 63 samples derived from 16 AML patients
during 28 chemotherapy cycles. Activated T cells showed a broad cytokine release profile, but IFNγ and GM-CSF levels showed
a significant correlation and were generally higher than the other cytokine levels. Higher IFNγ and GM-CSF responses were
associated with a low CD4:CD8 ratio, older patient age and no ongoing chemotherapy indicating potential utility of T cell
activation regimes for the older AML patient. The cytokine levels could be further increased by the novel protein kinase C
agonist PEP005, which also induced significant production of IL2 and TNFα which could contribute to anti-tumor effects in
AML patients. We conclude that remaining T cells after intensive AML therapy show a broad cytokine release profile including
high and significantly correlated levels of potentially anti-leukemic IFNγ and the AML growth factor GM-CSF. The final outcome
of an AML-initiated T cell cytokine response will thus depend on the functional characteristics of the AML cells, in particular

Intra- and intersexual variation in 16 skull dimensions and total body length of 233 aged Hooded seals caught in the north-west Atlantic were investigated. Absolute growth was described by asymptotic growth curves applied to single dimensions, as well as to scores on the first principal component of logarithmically transformed cranial data, which are believed to reflect the multivariate nature of growth. All dimensions were found to be fully developed later in males than in females. The growth of the male skulls was found to continue for more than 20 years, while the females approach final size about–7 years earlier than males, according to the scores on the first principal component. Female seals were found to reach 86 % of final total body length at the time of maturation, which is a generalized pinniped pattern. In both sexes, a yearling skull was characterized by a large brain-case, which decreased relatively with growth. The male skulls were further characterized by an increase in zygomatic width and orbital width in relation to basal length, a pattern which was not found in females.

1. Wolves Canis lupus L. recolonized the boreal forests in the southern part of the Scandinavian peninsula during the late 1990s, but so far there has been little attention to its effect on ecosystem functioning. Wolf predation increases the availability of carcasses of large prey, especially moose Alces alces L., which may lead in turn to a diet switch in facultative scavengers such as the wolverine Gulo gulo L. 2. Using 459 wolverine scats collected during winter-spring 2001-04 for DNA identity and dietary contents, we compared diet inside and outside wolf territories while controlling for potential confounding factors, such as prey density. We tested the hypothesis that wolverine diet shifted towards moose in the presence of wolves, while taking into account possible sexual segregation between the sexes. Occurrence of reindeer, moose and small prey was modelled against explanatory covariates using logistic mixed-effects models. Furthermore, we compared diet composition and breadth among habitats and sexes. 3. Occurrence of reindeer, moose and small prey in the diet varied with prey availability and habitat. As expected, diet contained more moose and less reindeer and small prey in the presence of wolves. Their diet in tundra consisted of 40% reindeer Rangifer tarandus L., 39% moose and 9% rodents. In forest with wolf, their diet shifted to 76% moose, 18% reindeer and 5% rodents; compared to 42% moose, 32% reindeer and 15% rodents in forest without wolf. This diet switch could not be explained by higher moose density in wolf territories. Female diet consisted of more small prey than for males, but there was a tendency for females to use the highly available moose carrion opportunistically and to hunt less on small prey within wolf territories. 4. Our study highlights how wolves increase scavenging opportunities for wolverines, and how sexual differences in diet may also apply to large scavengers. Due to their more restricted home range, female wolverines are forced to rely more on hunting small prey. The relatively high occurrence of wolf kills, however, forms an important food source to wolverines in this area. The recolonization of wolves may therefore have contributed to the consequent recolonization of wolverines into the same area. 相似文献

T lymphocyte secretion of interleukin-13 (IL-13) in response to different activation signals was characterized in vitro. IL-13
release was investigated when virus transformed B lymphocytes or acute myelogenous leukaemia (AML) blasts were used as accessory
cells during T cell activation. First, a majority of both CD4+ and CD8+ TCRαβ+ T lymphocyte clones, derived from normal individuals and bone marrow transplant recipients, secreted IL-13 in response to
a standardized mitogenic activation signal (phytohaemagglutinin+IL-2+ B lymphocyte accessory cells). The CD4+ cells showed significantly higher IL-13 levels than the CD8+ subsets. Second, when leukaemic accessory cells (more than 95% AML blasts) were used during T cell activation, IL-13 was
released both during alloactivation of normal T lymphocytes and during mitogen activation of posttransplant T cells. Third,
when normal T lymphocytes were stimulated with allogeneic AML blasts, addition of IL-13-neutralizing monoclonal antibodies
decreased interferon γ levels. Although addition of IL-13-neutralizing antibodies did not alter granulocyte-colony-stimulating
factor secretion by allostimulating AML blasts, altered blast proliferation was detected for certain patients. Thus, most
T cell clones can release IL-13, and IL-13 can modulate cytokine responses during T cell recognition of allogeneic AML cells.

Non-random association of females has been reported in many social species, with kinship being the main mechanism in shaping these associations. Understanding the patterns of association during rut is particularly important in polygynous animals, as this may have implications for inbreeding vulnerability and genetic structuring of populations. However, these patterns are highly dynamic and may be influenced by the population sex ratio and male age structure. We examined females' association in reindeer (Rangifer tarandus), a highly polygynous ungulate, to determine whether genetically related females associate more often than non-kin during rut and whether this association was influenced by the aforementioned factors. During three rutting seasons the male age structure and the sex ratio of the herd was manipulated: two extremely female biased sex ratios, one with only adult males and one with only young males, whereas the third being closer to a more even sex ratio and male age structure. We found that genetic relatedness influenced female association in the mixed male age group, but not in the adult and young male age groups. Highly related females (relatedness ??0.5) associated more closely than unrelated females. Further, period of the rut as well as male age structure and sex ratio had a significant effect on female association. In northern Fennoscandia, the spatial and temporal grazing practices of semi-domesticated reindeer (Rangifer tarandus tarandus) vary, which implies different grazing effects dependent on natural conditions as well as management regime (i.e., timing
and intensity of grazing). We compared density and biomass of main plant groups in semi-dry mountain birch forests exposed
to either long-term summer or winter grazing in three reindeer herding districts in the northernmost Finland. Percent plant
cover, height, and biomass of reindeer lichens (Cladonia spp.) and dwarf shrubs (Vaccinium uliginosum, Calluna vulgaris, and Betula nana) were lower on summer ranges compared with winter ranges. The biomass of other dwarf shrubs (Vaccinium myrtillus and V. vitis-idaea), and graminoids and herbs, and the % cover of non-vegetated bare soil and litter were, however, higher on summer ranges
than on winter ranges. Young mountain birch shoots (Betula pubescens ssp. czerepanovii) were less frequent on summer ranges than on winter ranges. The total leaf biomass under the browsing height of reindeer
(<1.5 m) was also lower on summer ranges compared with winter ranges. Especially in drier and nutrient poor mountain birch
forests, intensive summer grazing reduces the quantity of lichens and total plant biomass which affects the ecological state
and productivity of these forests and also reduces especially their winter grazing value for reindeer. Therefore, in addition
to regulating the maximum sustained numbers of reindeer, pasture rotation systems that effectively protect dry and nutrient

Accurate estimates of trait evolvabilities are central to predicting the short‐term evolutionary potential of populations, and hence their ability to adapt to changing environments. We quantify and evaluate the evolvability of herkogamy, the spatial separation of male and female structures in flowers, a key floral trait associated with variation in mating systems. We compiled genetic‐variance estimates for herkogamy and related floral traits, computed evolvabilities, and compared these among trait groups and among species differing in their mating systems. When measured in percentage of its own size, the median evolvability of herkogamy was an order of magnitude greater than the evolvability of other floral size measurements, and was generally not strongly constrained by genetic covariance between its components (pistil and stamen lengths). Median evolvabilities were similar across mating systems, with only a tendency toward reduction in highly selfing taxa. We conclude that herkogamy has the potential to evolve rapidly in response to changing environments. This suggests that the extensive variation in herkogamy commonly observed among closely related populations and species may result from rapid adaptive tracking of fitness optima determined by variation in pollinator communities or other selective factors. 相似文献
