Re-examination of tissue sections from four Takydromus tachydromoides (Sauria: Lacertidae) naturally infected with Plasmodium sasai found liver parenchymal cells, containing uninucleate parasites which may correspond to the hypnozoite stage of primate malaria parasites, schizonts and segmenters in parenchymal cells, and hepatic macrophages which contained numerous schizonts. Following destaining of the original H&E and prolonged restaining with warm Giemsa stain, encysted schizonts, protected by a hyaline wall, were discovered in the connective tissue or capillary endothelium of lung, liver, brain, heart, pancreas, kidney, intestine wall, testis, and both intra- and intermuscularly in the femoral muscles. Unencysted schizonts in the pulmonary endothelium apparently represent the phanerozoic stages, which, following encystment in the various tissues, are recognized as a new stage in the life cycle of reptilian malarial parasites, the chronozoic schizonts. A hypothesis is presented to describe the life cycle of P. sasai, which may be characteristic of other saurian malaria parasites. It interprets the sequence of pre-erythrocytic stages found as follows: sporozoites enter hepatic parenchymal cells where some may become dormant as hypnozoites, and others form cryptozoic schizonts. The cryptozoites parasitize hepatic macrophages and form metacryptozoic schizonts. Metacryptozoites produce phanerozoic schizonts in the capillary endothelium and connective tissue of the lung and other organs. Phanerozoites and possibly metacryptozoites then invade the erythrocytes to begin the erythrocytic cycle. Some of the phanerozoites in endothelium, connective tissue and skeletal muscle become encysted as chronozoic schizonts, and their progeny, chronozoites, renew the erythrocytic cycle throughout the life of the host and produce seasonal relapses of gametocytemia, in spring, at the end of hibernation by the lizard.  相似文献

Summary A combination of simple membrane theory and statical analysis has been used to determine how stresses are carried in echinoid skeletons. Sutures oriented circumferentially are subject principally to compression. Those forming radial zig-zags are subject to compression near the apex and tension near the ambitus. Radial and circumferential sutures in Eucidaris are equally bound with collagen fibers but in Diadema, Tripneustes, Psammechinus, Arbacia and other regular echinoids, most radial sutures are more heavily bound, and thus stronger in tension. Psammechinus, Tripneustes and several other echinoids have radial sutures thickened by ribs which increase the area of interlocking trabeculae. Ribs also increase flexural stiffness and carry a greater proportion of the stress. Further, ribs effectively draw stress from weaker areas pierced by podial pores, and increase the total load which can be sustained.Allometry indicates that regular echinoids become relatively higher at the apex as size increases, thus reducing ambital stresses. Some spatangoids with very high domes (eg Agassizia) maintain isometry, but others (eg Meoma) become flatter with size. Both holectypoids (Echinoneus) and cassiduloids (Apatopygus) maintain a constant height to diameter relationship. Flattening, and consequently ambital tensile stress, is greatest in the clypeasteroids. In this group the formation of internal buttresses which preferentially carry stress, reaches maximum development. A notable exception, however, is the high domed Clypeaster rosaceus.In this analysis it was assumed that local buckling or bending does not occur. The test of some echinoids (e.g. Diadematoida) have relatively wide sutures swathed in collagen, which allows local deformation. Others (e.g. Arbacia) have rigid sutures with reduced collagen. In Psammechinus and other members of the Order Echinoida, in addition to rib formation, inner and outer surface trabeculae are thickened so that the individual plates are stiffened. Some spatangoids (Meoma, Paleopneustes) have extensive sutural collagen, but the cassiduloid Apatopygus has collagen confined to junctions of sutures, and elsewhere the joints are strengthened and stiffened by fusion of trabeculae. Fusion of surface trabeculae is almost complete in the holectypoid, Echinoneus, and the sutures are obscured.  相似文献

The structure of chromatin containing amplified N-myc in neuroblastoma and retinoblastoma cells was investigated using micrococcal nuclease digestion of isolated nuclei. The size distribution of DNA fragments containing N-myc, produced by micrococcal nuclease digestion of nuclei, was determined and compared to that of DNA containing the structural gene for dihydrofolate reductase. A perturbation of the native structure of chromatin containing N-myc was evident from the association of N-myc with more extensively digested DNA when compared with chromatin containing dihydrofolate reductase.  相似文献

To determine the identity of the tick vector of enzooticBabesia gibsoni in California, two common ixodid ticks were allowed to engorge uponB. gibsoni infected dogs. Sporozoites were observed in the salivary glands of prefed nymphalRhipicephalus sanguineus ticks that fed as larvae onB. gibsoni-infected dogs. A higher proportion (31%) of nymphal ticks that prefed on an uninfected dog for 48 hours contained sporozoites in their salivary glands than did ticks which had fed for 24 hours (13%). Sporozoites were not observed in the salivary glands of prefedR. sanguineus nymphs which were derived from the eggs of adult females that fed on an infected dog, in adults that were fed as nymph on an infected dog, or in the nymphal and adult uninfected controls.Dermacentor variabilis ticks appeared not to become infected. Although attempts to transmitB. gibsoni to susceptible, splenectomized dogs were unsuccessful,R. sanguineus would appear to be the most likely tick vector to maintain this piroplasm in California. The chaetognaths are an extraordinarily homogeneous phylum of animals at the morphological level, with a bauplan that can be traced back to the Cambrian. Despite the attention of zoologists for over two centuries, there is little agreement on classification within the phylum. We have used a molecular biological approach to investigate the phylogeny of extant chaetognaths. A rapidly evolving expansion segment toward the 5′ end of 28S ribosomal DNA (rDNA) was amplified using the polymerase chain reaction (PCR), cloned, and sequenced from 26 chaetognath samples representing 18 species. An unusual finding was the presence of two distinct classes of 28S rDNA gene in chaetognaths; our analyses suggest these arose by a gene (or gene cluster) duplication in a common ancestor of extant chaetognaths. The two classes of chaetognath 28S rDNA have been subject to different rates of molecular evolution; we present evidence that both are expressed and functional. In phylogenetic reconstructions, the two classes of 28S rDNA yield trees that root each other; these clearly demonstrate that the Aphragmophora and Phragmophora are natural groups. Within the Aphragmophora, we find good support for the groupings denoted Solidosagitta, Parasagitta, and Pseudosagitta. The relationships between several well-supported groups within the Aphragmophora are uncertain; we suggest this reflects rapid, recent radiation during chaetognath evolution. Summary A detailed study was conducted of the three-dimensional accuracy of phonotaxis by femaleHyperolius marmoratus. This analysis involved videotape recordings of phonotactic approaches to an elevated loudspeaker through a three-dimensional grid. Females readily resolved the sound source elevation, but the jump error angles describing the precision of approach were considerably greater in this three-dimensional analysis than in the more conventional two-dimensional ground approach analysis. Extensive use was made of visual cues in elevated phonotactic approach and lateral head scanning prior to jumps, often accompanied by vertical changes in head orientation, was frequent.
