Summary The idea has been tested here that the aberration in amino acid controlled regulation of RNA synthesis in a mutant strain ofE. coli might reflect a major breakdown in the specificity of transfer of amino acids to S-RNA. For this purpose, S-RNA and amino acid activating enzymes were extracted from bacteria carrying either the normalRC st or the aberrantRC rel allele of the RNA control gene. The purified S-RNA preparations were first charged enzymatically with one or more of the 20 standard amino acids, then oxidized with periodate, and finally reisolated and retested for their residual capacity to accept an amino acid that was absent from the preliminary charging mixture. If preliminary charging transferred an amino acid to a non-cognate S-RNA species belonging to an absent amino acid, then the acceptor capacity for the missing amino acid would survive periodate oxidation and reveal its presence on recharging with that amino acid after post-periodate reisolation of the S-RNA. The results presented here show that there does not appear to exist any such major breakdown of transfer specificity in eitherRC st orRC rel bacteria: preliminary charging of the S-RNA fromRC rel bacteria with 19 of the 20 standard amino acids by use of the homologous amino acid activating enzymes does not afford protection against periodate oxidation for any appreciable fraction of the acceptor capacity for the absent 20th amino acid (when that amino acid is either methionine or arginine). It is unlikely, therefore, that thecatholic inducer, postulated to explain the continued RNA synthesis ofRC rel amino acid auxotrophs in the absence of their growth requirement, is one of the 20 standard amino acids.This investigation was supported by Public Health Service Research Grant CA 02129, from the National Cancer Institute.  相似文献

Objective: To see whether a fat‐rich (50%) evening meal promoted fat oxidation and a different spontaneous food intake on the following day at breakfast than a meal with a lower fat content (20%) in 10 prepubertal obese girls. Research Methods and Procedures: The postabsorptive and postprandial (10.5 hours) energy expenditure after a low‐fat (LF) (20% fat, 68% carbohydrate, 12% protein) and an isocaloric (2.1 MJ) and isoproteic high‐fat (HF; 50% fat, 38% carbohydrate, 12% protein) meal were measured by in direct calorimetry. Results: Fat oxidation was not significantly different after the two meals [LF, 31 ± 9 vs. HF, 35 ± 9 g/10.5 hours, p = not significant (NS)]. The girls oxidized 1.8 ± 0.9 times more fat than that ingested (11.1 grams) with the LF meal vs. 0.3 ± 0.3 times more fat than that ingested (27.1 grams) with the HF meal (p < 0.001). Carbohydrate oxidation was significantly higher after an LF than an HF meal (39 ± 12 vs. 29 ± 9 g/10.5 hours, p < 0, 05). At breakfast, the girls spontaneously ingested a similar amount of energy (1.5 ± 0.7 vs. 1.5 ± 0.6 MJ, p = NS) and macronutrient proportions (fat, 23% vs. 26%, p = NS; protein, 9% vs. 10%; carbohydrate, 68% vs. 64%,) independently of their having eaten an HF or an LF dinner. Discussion: An HF dinner did not stimulate fat oxidation, and no compensatory effect in spontaneous food intake was observed during breakfast the following morning. Cumulated total fat oxidation after dinner was higher than total fat ingested at dinner, but a much larger negative fat balance was observed after the LF meal. Spontaneous energy and nutrient intakes at breakfast were similar after LF and HF isocaloric, isoproteic dinners. This study points out the lack of sensitivity of short‐term fat balance to subsequently readjust fat intake and emphasizes the importance of an LF meal to avoid transient positive fat imbalance.  相似文献

The chemical composition of the surface layers of synthetic biomaterials used for human medical devices and in biotechnology plays a key role in determining interfacial interactions between biological media (such as protein solutions, cells, tissue) and the synthetic material. Accordingly, considerable research efforts focus on improving the 'biocompatibility' of biomaterials by applying various surface modification and thin film coating approaches. Here we focus on the patterning of surface chemistries, often designed to exercise spatial control over events such as cell attachment and spreading. Secondly, we review recent developments in chemical characterisation of biomaterials surfaces, which is essential both for verifying the success of intended surface modification strategies and for reliable interpretation of observed biological responses. Biomaterials surface analysis by imaging ToF-SIMS and XPS and compositional depth profiling are discussed, as is the emerging complementary technique of Metastable Induced Electron Spectroscopy.  相似文献

Most large mammals have constantly been exposed to anthropogenic influence over decades or even centuries. Because of their long generation times and lack of sampling material, inferences of past population genetic dynamics, including anthropogenic impacts, have only relied on the analysis of the structure of extant populations. Here, we investigate for the first time the change in the genetic constitution of a natural red deer population over two centuries, using up to 200‐year‐old antlers (30 generations) stored in trophy collections. To the best of our knowledge, this is the oldest DNA source ever used for microsatellite population genetic analyses. We demonstrate that government policy and hunting laws may have strong impacts on populations that can lead to unexpectedly rapid changes in the genetic constitution of a large mammal population. A high ancestral individual polymorphism seen in an outbreeding population (1813–1861) was strongly reduced in descendants (1923–1940) during the mid‐19th and early 20th century by genetic bottlenecks. Today (2011), individual polymorphism and variance among individuals is increasing in a constant‐sized (managed) population. Differentiation was high among periods (FST > ***); consequently, assignment tests assigned individuals to their own period with >85% probability. In contrast to the high variance observed at nuclear microsatellite loci, mtDNA (D‐loop) was monomorphic through time, suggesting that male immigration dominates the genetic evolution in this population.  相似文献

A dissected-leaved form of Ranunculus repens L. occurs in the temporary limestone lakes (turloughs) across the west of Ireland. Turloughs fill with groundwater for up to 8 months of the year. Under experimental conditions, these turlough populations demonstrated a higher rate of aerial and submerged photosynthesis than populations of the more typical broad-leaved ruderal form. The turlough populations also had higher rates of stomatal conductance and exhibited a higher stomatal index on the upper leaf surface and a lower index on the lower leaf surface than the ruderal populations. Neither population could utilize bicarbonate to any great extent, with rates of photosynthesis under submerged conditions being only 5 % of aerial rates. Respiration under submerged conditions was significantly higher in the turlough populations than in ruderal populations, and it is hypothesized that the more dissected leaf shape of the turlough population may have a thinner boundary layer and thus enhance gas exchange in submerged conditions.  相似文献

Oithona similis is an abundant but poorly studied cyclopoidcopepod in the brackish Central Baltic Sea. We describe thespatio-temporal distribution of O. similis in a Central Balticdeep basin (Bornholm Basin) during spring and summer 1999. Usingvertically resolving sampling in parallel with hydrographicmeasurements, we found the copepod to dwell in the permanenthalocline characteristic of a Central Baltic deep basin. Thehabitat of O. similis is thus limited from above by low salinityand from below by low oxygen conditions, both characteristicfor the area. Horizontally resolving sampling yielded abundancesurfaces which were compared by analysis of variance showingsimilar patterns among sampling dates. Comparison with flowfields from a three-dimensional hydrodynamic model suggeststhat the horizontal distribution is primarily the result ofcirculation in the dwelling depth. The study shows how the physicalenvironment in the area determines the spatial distributionwhich might affect abundance and production of this copepod.  相似文献
