The dengue fever epidemic in Guangzhou may have been affected by the Coronavirus Disease 2019 (COVID-19) pandemic. The number of dengue cases dropped drastically in 2020, and there have been only 2 local cases, suggesting that dengue has not become endemic in Guangzhou.

Guangzhou is located on the southeast coast of China and is the country’s third largest city. Since 1978, outbreaks of dengue fever have occurred intermittently in this city. In the past decade, the number of reported dengue cases reached more than 1,000 in 2013, 2014, 2018, and 2019, with 37,385 cases reported in 2014 alone. Therefore, dengue fever is a major public health concern in Guangzhou, and there is a continuing argument that it is endemic in Guangzhou [13].The numbers of dengue cases from 2017 to 2020 are shown in Table 1. In 2020, the total and local case numbers dropped dramatically compared to the previous years. With a high proportion of imported cases (n = 32, 94.12%), the proportion of local cases (n = 2, 5.88%) was considerably low in 2020. All the prevention and control strategies for dengue, including issuing public education messages, preventing further mosquito bites in patients, cleaning vector breeding sites, and using pesticides, were similar during these years. Additionally, dengue, as a mosquito-borne viral infectious disease, is closely related to mosquito density. The mosquito ovitrap index (MOI), which is the proportion of positive mosquito ovitraps, is usually used to indicate mosquito density. The MOI in 2017, 2018, and 2019 was 7.073 ± 1.016, 9.657 ± 1.307, and 8.464 ± 0.961, respectively. The average MOI was 8.398 ± 0.648 from 2017 to 2019 in Guangzhou. The MOI in 2020 was 7.135 ± 0.786, which remained at the median risk level. Therefore, the abnormal decline in dengue cases could not be attributed to the change in mosquito density in 2020.Table 1Numbers and percentages of dengue cases from 2017 to 2019.

Open in a separate windowIn 2020, the 14-day quarantine in a designated hotel for international travelers to curb the spread of Coronavirus Disease 2019 (COVID-19) was an important public health intervention. People had to remain indoors except for medical care needs. All imported dengue cases were identified during their quarantine periods. No secondary case related to the imported cases was reported. This may be because Aedes albopictus, which is the major vector of dengue in Guangzhou, bites aggressively during the day outdoors. The chance of being bitten by A. albopictus was reduced by staying all day indoors. Moreover, some research revealed that viremia occurred 6 to 18 hours before symptoms appeared and lasted as long as 12 days [4]. After the 14-day quarantine, viremia had almost subsided. Therefore, imported dengue cases were unlikely to be transmitted. The impact of imported dengue cases was limited by the quarantine, which provided a rare opportunity to identify the local epidemic.The epidemiology investigation showed that the 2 local cases, who were living in the same building, had no travel history outside Guangzhou in 2020 and had symptoms successively. Two dengue virus serotype 2 (DENV-2) strains were isolated from them. The envelope gene sequences were obtained and deposited in GenBank under accession numbers MW295818 and MW345921. Reference sequences, which were downloaded from GenBank, and sequences of Guangzhou strains identified in the previous years, were used to construct a phylogenetic tree. The 2 isolated strains in 2020 were identical. The tree (Fig 1) shows that the 2 strains belonged to the Malaysia/Indian subcontinent genotype, which was the prevailing genotype in Guangzhou [5]. However, they were neither identical with nor derived from the Guangzhou strains obtained from the previous years. Using the Basic Local Alignment Search Tool in GenBank, the 2 strains were found to be highly similar to those identified in Zhejiang (China), Singapore, and Guangdong (China) in 2017. To obtain bacteria with arsenic accumulation potential that can be used to remove arsenic from contaminated waters, experiments were made to investigate the tolerance and accumulation to arsenic of an indigenous bacterium XZM002 isolated from aquifer sediments of Datong Basin, northern China. The results showed that strain XZM002 belongs to the genus Bacillus and has evolved defense mechanisms to reduce arsenic injury: the change of cellular shape from initial rod to oval and then to round with increment of arsenic toxicity. The effect of arsenate or arsenite on the bacterial growth was also investigated. Results showed that growth of the strain was inhibited under As(III) and high concentration As(V) (over 1200 μg l?1) conditions in the first 2 days and promoted under low concentration As(V) (under 400 μg l?1) condition. Its arsenic bioaccumulation potential was surveyed by monitoring the concentration changes of total arsenic and arsenic speciation in the medium and in the cytoplasm, and those of total arsenic on the membrane. Methylated arsenic species were not detected throughout the experiment. The results indicated that 11.5% of arsenic was removed from liquid medium into the bacterial cells and 9.22% of As(V) in the medium was transformed gradually to As(III) during 4 d of incubation. Approximately 80% of the total accumulated arsenic was adsorbed onto the membrane instead of into cytoplasm; and the arsenic accumulation almost approached saturation after incubation for 72 h.  相似文献

Myopia is a refractive error of the eye that is prevalent worldwide. The most extreme form, high myopia, is usually associated with other ocular disorders such as retinal detachment, macular degeneration, cataract, and glaucoma, and is one of leading causes of blindness. The etiology is complex and has not been fully elucidated. In this study, we identified a novel missense variant of the CCDC111 gene (NM_152683.2: c.265T > G; p.Y89D) in a high myopia family by exome sequencing. The variant was identified in 4 patients from an additional 270 sporadic high myopia patients, but not found in 270 controls. The amino acid is highly conserved across species, and variants giving rise to amino acid substitutions are predicted to be functionally damaging. The CCDC111 gene was ubiquitously expressed in primary cell cultures from human eye tissue, including corneal epithelial cells, choroidal melanoma cells, scleral fibroblasts, retinal epithelial cells, retinal Müller cells, and lens capsule epithelial cells. In summary, our results suggested that the CCDC111 may be a susceptibility gene for high myopia.  相似文献

To explore the disassembly mechanism of tobacco mosaic virus (TMV), a model system for virus study, during infection, we have used single-molecule force spectroscopy to mimic and follow the process of RNA disassembly from the protein coat of TMV by the replisome (molecular motor) in vivo, under different pH and Ca2+ concentrations. Dynamic force spectroscopy revealed the unbinding free-energy landscapes as that at pH 4.7 the disassembly process is dominated by one free-energy barrier, whereas at pH 7.0 the process is dominated by one barrier and that there exists a second barrier. The additional free-energy barrier at longer distance has been attributed to the hindrance of disordered loops within the inner channel of TMV, and the biological function of those protein loops was discussed. The combination of pH increase and Ca2+ concentration drop could weaken RNA-protein interactions so much that the molecular motor replisome would be able to pull and disassemble the rest of the genetic RNA from the protein coat in vivo. All these facts provide supporting evidence at the single-molecule level, to our knowledge for the first time, for the cotranslational disassembly mechanism during TMV infection under physiological conditions.  相似文献

In this study, we prepared nano-hydroxyapatite/polyamide 66/glass fibre (n-HA/PA66/GF) bioactive bone screws. The microstructure, morphology and coating of the screws were characterised, and the adhesion, proliferation and viability of MC3T3-E1 cells on n-HA/PA66/GF scaffolds were determined using scanning electron microscope, CCK-8 assays and cellular immunofluorescence analysis. The results confirmed that n-HA/PA66/GF scaffolds were biocompatible and had no negative effect on MC3T3-E1 cells in vitro. To investigate the in vivo biocompatibility, internal fixation properties and osteogenesis of the bioactive screws, both n-HA/PA66/GF screws and metallic screws were used to repair intercondylar femur fractures in dogs. General photography, CT examination, micro-CT examination, histological staining and biomechanical assays were performed at 4, 8, 12 and 24 weeks after operation. The n-HA/PA66/GF screws exhibited good biocompatibility, high mechanical strength and extensive osteogenesis in the host bone. Moreover, 24 weeks after implantation, the maximum push-out load of the bioactive screws was greater than that of the metallic screws. As shown by their good cytocompatibility, excellent biomechanical strength and fast formation and ingrowth of new bone, n-HA/PA66/GF screws are thus suitable for orthopaedic clinical applications.  相似文献

Background

Macrophage death in advanced lesion has been confirmed to play an important role in plaque instability. However, the mechanism underlying lesion macrophage death still remains largely unknown.

Methods and Results

Immunohistochemistry showed that caspase-1 activated in advanced lesion and co-located with macrophages and TUNEL positive reaction. In in-vitro experiments showed that ox-LDL induced caspase-1 activation and this activation was required for ox-LDL induced macrophages lysis, IL-1β and IL-18 production as well as DNA fragmentation. Mechanism experiments showed that CD36 and NLRP3/caspase-1/pathway involved in ox-LDL induced macrophage pyroptosis.

Conclusion

Our study here identified a novel cell death, pyroptosis in ox-LDL induced human macrophage, which may be implicated in lesion macrophages death and play an important role in lesion instability.  相似文献

The initial growth of the nacreous layer is crucial for comprehending the formation of nacreous aragonite. A flat pearl method in the presence of the inner-shell film was conducted to evaluate the role of matrix proteins in the initial stages of nacre biomineralization in vivo. We examined the crystals deposited on a substrate and the expression patterns of the matrix proteins in the mantle facing the substrate. In this study, the aragonite crystals nucleated on the surface at 5 days in the inner-shell film system. In the film-free system, the calcite crystals nucleated at 5 days, a new organic film covered the calcite, and the aragonite nucleated at 10 days. This meant that the nacre lamellae appeared in the inner-shell film system 5 days earlier than that in the film-free system, timing that was consistent with the maximum level of matrix proteins during the first 20 days. In addition, matrix proteins (Nacrein, MSI60, N19, N16 and Pif80) had similar expression patterns in controlling the sequential morphologies of the nacre growth in the inner-film system, while these proteins in the film-free system also had similar patterns of expression. These results suggest that matrix proteins regulate aragonite nucleation and growth with the inner-shell film in vivo.  相似文献

Link Clustering (LC) is a relatively new method for detecting overlapping communities in networks. The basic principle of LC is to derive a transform matrix whose elements are composed of the link similarity of neighbor links based on the Jaccard distance calculation; then it applies hierarchical clustering to the transform matrix and uses a measure of partition density on the resulting dendrogram to determine the cut level for best community detection. However, the original link clustering method does not consider the link similarity of non-neighbor links, and the partition density tends to divide the communities into many small communities. In this paper, an Extended Link Clustering method (ELC) for overlapping community detection is proposed. The improved method employs a new link similarity, Extended Link Similarity (ELS), to produce a denser transform matrix, and uses the maximum value of EQ (an extended measure of quality of modularity) as a means to optimally cut the dendrogram for better partitioning of the original network space. Since ELS uses more link information, the resulting transform matrix provides a superior basis for clustering and analysis. Further, using the EQ value to find the best level for the hierarchical clustering dendrogram division, we obtain communities that are more sensible and reasonable than the ones obtained by the partition density evaluation. Experimentation on five real-world networks and artificially-generated networks shows that the ELC method achieves higher EQ and In-group Proportion (IGP) values. Additionally, communities are more realistic than those generated by either of the original LC method or the classical CPM method.  相似文献

Little is known about the relevance of genetic polymorphisms to arsenic-related bladder cancer. A preliminary case-control study was conducted to explore the association between genetic polymorphisms of GSTT1, p53 codon 72 and bladder cancer in southern Taiwan, a former high arsenic exposure area. Fifty-nine urinary transitional cell carcinoma (TCC) patients from a referral centre in south-western Taiwan and 81 community controls matched on residence were recruited from 1996 to 1999. A questionnaire was administered to obtain arsenic exposure and general health information. Genotypes of p53 codon 72 and GSTT1 were analysed by polymerase chain reaction-restriction fragment length polymerase. The combined variant genotypes (heterozygous or homozygous variant) of p53 codon 72 and GSTT1 null were observed in 29% of cases and in 44% of controls, respectively. In this preliminary study, bladder cancer risk was slightly elevated for subjects carrying the variant genotype of p53 codon 72 or in subjects carrying the GSTT1 null genotype. Variants in p53 codon 72 increased the risk of bladder cancer among smokers. However, the results were not statistically significant and larger confirmatory studies are needed to clarify the role of candidate gene polymorphisms and bladder cancer risk in arsenic exposed populations.  相似文献
