We developed 15 microsatellite loci in the poplar rust fungus, Melampsora larici‐populina, using two enrichment protocols. Polymorphism of each locus was assessed on a panel of 30 isolates, comprising three subpanels (world, regional and local scales). Thirteen loci were polymorphic with three to eight alleles detected. The 15 loci were also tested on five related Melampsora species, M. allii‐populina, M. medusae f. sp. deltoidae, M. larici‐tremulae, M. rostrupii and M. pinitorqua, and partial or global cross‐amplification events were detected. 相似文献

Species delimitation, an issue central to systematics and biodiversity studies, is addressed in the epunctifera group of the stem borer genus Sesamia Guenée (Lepidoptera: Noctuidae). This group is composed of four sub‐Saharan species: Sesamia poephaga Tams & Bowden; Sesamia epunctifera Hampson; Sesamia penniseti Tams & Bowden; and Sesamia poebora Tams & Bowden, the taxonomic status of which was unclear. The first species was considered a possible synonym of the second, and the third species was considered a possible synonym of the fourth. An analysis combining morphological, ecological and molecular data enables us to conclude that S. epunctifera and S. poephaga are different species, and that S. poebora is a synonym of S. penniseti. Two new species were discovered: Sesamia firmata sp.n. and Sesamia veronica sp.n. Sesamia firmata sp.n. has atypical genitalic morphology, suggesting a strong selection resulting in a reinforcement of pre‐zygotic isolation. Some specimens previously identified as S. penniseti on the basis of morphology are sisters to S. epunctifera on the mitochondrial tree, and are connected to S. penniseti on the nuclear tree. The mitochondrial distance from S. penniseti and S. epunctifera is 7.6% and 3.9%, respectively, suggesting an ancient mitochondrial introgression from S. epunctifera into S. penniseti. The possible causes of the reinforcement and introgressive hybridization are discussed. This case of mitochondrial introgression, uncommon in Lepidoptera, in which females are the heterogametic sex, may be an exception to Haldane's rule. Ontogenetic and then heterochronic approaches are used here to analyze sexual differentiation within two well-known Jurassic dimorphic species. This analysis compares two ways of determining the relative biological age of ammonites, one using size (diameter) and the other the number of septa as a proxy of age. The shape standard is established from factor analysis of morphological and growth parameters. Size-age-shape relationships are analyzed on the basis of a new heterochronic representation. When diameter is used as a proxy of age, microconch morphs are globally considered to be progenetic compared with macroconch morphs. When size and age are determined separately and shape is included, (1) Ebrayiceras jactatum (microconch morph) has a shorter period of growth (progenesis) with acceleration of shape compared with its macroconch morph ( Morphoceras macrescens ), (2)( Cardioceras cordatum (microconch morph) has a shorter period of growth (progenesis) coupled with a slower rhythm of growth (dwarfism) compared with its macroconch morph. These findings emphasize the complex relationship between supposed sexual dimorphs in ammonites when size and age standards can be separated, as well as the need for an ontogenetic approach. 相似文献

Methionine (Met) in proteins can be oxidized to two diastereoisomers of methionine sulfoxide, Met‐S‐O and Met‐R‐O, which are reduced back to Met by two types of methionine sulfoxide reductases (MSRs), A and B, respectively. MSRs are generally supplied with reducing power by thioredoxins. Plants are characterized by a large number of thioredoxin isoforms, but those providing electrons to MSRs in vivo are not known. Three MSR isoforms, MSRA4, MSRB1 and MSRB2, are present in Arabidopsis thaliana chloroplasts. Under conditions of high light and long photoperiod, plants knockdown for each plastidial MSR type or for both display reduced growth. In contrast, overexpression of plastidial MSRBs is not associated with beneficial effects in terms of growth under high light. To identify the physiological reductants for plastidial MSRs, we analyzed a series of mutants deficient for thioredoxins f, m, x or y. We show that mutant lines lacking both thioredoxins y1 and y2 or only thioredoxin y2 specifically display a significantly reduced leaf MSR capacity (–25%) and growth characteristics under high light, related to those of plants lacking plastidial MSRs. We propose that thioredoxin y2 plays a physiological function in protein repair mechanisms as an electron donor to plastidial MSRs in photosynthetic organs. 相似文献

The genus Pseudobulweria includes four extant species, three being classified as critically endangered, while the fourth (Tahiti Petrel P . rostrata ) is near threatened. Information on the breeding biology of any species of the genus is scarce, even for the Tahiti Petrel. We monitored two Tahiti Petrel nests in New Caledonia and provide the first data on the breeding biology for the genus. Incubation lasts about 55 days, and young fledge at 110 days. Breeding in the southern lagoon of New Caledonia extends from September to April. Very strong interspecific competition for nest cavities occurs with Wedge-tailed Shearwaters Puffinus pacificus . We also document geographical variation in both biometry and breeding phenology over the Pacific breeding range. 相似文献

Ten microsatellites were isolated from the barley stem gall midge, Mayetiola hordei. Polymorphism at each locus was tested on 40 individual midges, among which 34 were collected on barley and six on wheat crops in Tunisia. Six loci were polymorphic with the number of alleles ranging from two to seven. The observed heterozygosity varied between 0.025 and 0.2. These microsatellite loci revealed a strong effect of host plant on the population genetic structure of M. hordei. 相似文献

The major pest of maize in Mediterranean Europe, the stem borer Sesamia nonagrioides (Lefèbvre) (Lepidoptera: Noctuidae), has a fragmented distribution, north and south of the Sahara. The present study aimed: (1) to clarify the uncertain taxonomic status of the Palearctic and sub‐Saharan populations which were first considered as different species and later on as subspecies (Sesamia nonagrioides nonagrioides and Sesamia nonagrioides botanephaga) and (2) to investigate the origin of the Palearctic population which extends from Spain to Iran, outside what is considered typical for this mainly tropical genus. We reconstructed the evolutionary history of both populations using one nuclear and two mitochondrial genes. The sub‐Saharan taxon was fragmented in two isolated populations (West and East) whose mitochondrial genes were distant by 2.3%. The Palearctic population was included in the East African clade and its genes were close or identical to those of a population from Central Ethiopia, where the species was discovered for the first time. Similarly, in Africa, the alleles of the nuclear gene were distributed mainly in two West and East clades, whereas some Palearctic alleles belonged to the West clade. The Palearctic population originated therefore from East and West Africa and is the progeny of the cross between these two African populations. The main species concepts were in agreement, leading to the conclusion that the three populations are still conspecific. In the surveyed regions, the species therefore does not include two subspecies but three isolated populations. The Palearctic population suffered from severe bottlenecks that resulted in the fixation of one East African mitochondrial genome and the large reduction in its genetic diversity compared to the African populations. The data suggest that natural colonization of the Palearctic region was more plausible than human introduction. The allelic distribution of the Palearctic population was similar to that of species that survived the last glaciation. It is concluded that the African populations expanded during the last interglacial, crossed the Sahara and mixed in North Africa where fixation of the East mitochondrial genome occurred. The species then colonized Europe westward through only one eastern entrance. Introgression arising from crop-to-wild gene flow provides novel sources of genetic variation in plant species complexes. Hybridization within the Beta vulgaris species complex is of immediate concern; crop lineages ( B . vulgaris ssp. vulgaris ) hybridize easily with their wild relatives ( B . vulgaris ssp. maritima ) thereby threatening wild beet gene diversity with genetic swamping. Hybridization 'hotspots' occur in European seed production areas because inland ruderal wild beets occur and reproduce in sympatry with cultivated beets. We studied gene flow occurring between seed-producing cultivars and ruderal wild B . vulgaris in southwestern France to determine whether feral beets, arising from unharvested cultivated seed, represent an opportunity for crop-to-wild gene flow. We surveyed 42 inland ruderal beet populations located near seed production fields for nucleo-cytoplasmic variation and used a cytoplasmic marker diagnostic of cultivated lines. Occurrence of cultivated-type cytoplasm within ruderal populations clearly reflected events of crop seed escape. However, we found no genetic signatures of nuclear cultivated gene introgression, which suggests past introgression of cultivated cytoplasm into a wild nuclear background through seed escape rather than recent direct pollen flow. Overall, patterns of genetic structure suggested that inland ruderal wild beet populations act as a metapopulation, with founding events involving a few sib groups, followed by low rates of seed or pollen gene flow after populations are established. Altogether, our results indicate that a long-lived seed bank plays a key role in maintaining cultivated-type cytoplasm in the wild and highlight the need for careful management of seed production areas where wild and cultivated relatives co-occur. 相似文献
