In a high mammary tumor strain of SHN virgin mice, the percent increase in the number of palpable mammary tumors during 3 weeks after the 1st tumor appearance was significantly retarded by ovariectomy and this retardation was prevented by pituitary grafting associated with the decrease and the increase in the circulating prolactin levels, respectively. A low mammary tumor strain of SLN virgin mice grafted with a single pituitary each at 2 and 5 months of ages developed mammary tumors 4 and 2 months after grafting (6 and 7 months of ages), respectively. However, there was no difference between groups in mammary tumor incidence at any month after mice in each group developed tumors first. Mammary tumor incidences of both groups were significantly higher than that of the untreated control at all ages examined. These findings have demonstrated that prolactin plays a key role in the initial progression of spontaneous mammary tumors of mice besides its prerequisite role in tumor development. They also suggest that there is no intrinsically age-related difference in prolactin effect on mammary tumorigenesis.  相似文献

Summary To breed industrially useful strains of a slow-growing, red-pigment-producing strain ofMonascus anka, protoplasts ofM. anka MAK1 (arg) andAspergillus oryzae AOK1 (met, thr) were fused. A mixture of protoplasts prepared from mycelia ofM. anka MAK1 treated with 2% Usukizyme and ofA. oryzae AOK1 treated with 2% Usukizyme and 0.2% NovoZym 234 was incubated with 30% (w/v) polyethylene glycol no. 6000. Heterokaryon fusants complementing the auxotrophies of both mutants were isolated on minimal medium, but segregated into red (MAK1) and white (AOK1) sectors after being cultured on a complete medium. After irradiation with UV light, the fusants gave stable heterozygous diploids that formed long white hyphae. These diploids, which had twice as much DNA in the nucleus as their parents, grew more rapidly than the parent strain YZT1, and produced ethanol earlier than the parents. Production of amylase, protease, and kojic acid by the fusants was intermediate in amount between that of the two parents.  相似文献

How floral organ number is specified is an interesting subject and has been intensively studied in Arabidopsis thaliana. In rice (Oryza sativa L.), mutations associated with floral organ number have been identified. In three mutants of rice, floral organ number 1 (fon1) and the two alleles, floral organ number 2-1 (fon2-1) and floral organ number 2-2 (fon2-2), the floral organs were increased in number centripetally. Lodicules, homologous to petals, were rarely affected, and stamens were frequently increased from six to seven or eight. Of all the floral organs the number of pistils was the most frequently increased. Among the mutants, fon1 showed a different spectrum of organ number from fon2 -1 and fon2 -2. Lodicules were the most frequently affected in fon1, but pistils of more than half of fon1 flowers were unaffected; in contrast, the pistils of most flowers were increased in fon2 -1 and fon2-2. Homeotic conversion of organ identity was also detected at a low frequency in ectopically formed lodicules and stamens. Lodicules and stamens were partially converted into anthers and stigmas, respectively. Concomitant with the increased number of floral organs, each mutant had an enlarged apical meristem. Although meristem size was comparable among the three mutants and wild type in the early phase of flower development, a significant difference became apparent after the lemma primordium had differentiated. In these mutants, the size of the shoot apical meristem in the embryo and in the vegetative phase was not affected, and no phenotypic abnormalities were detected. These results do not coincide with those for Arabidopsis in which clavatal affects the sizes of both shoot and floral meristems, leading to abnormal phyllotaxis, inflorescence fasciation and increased floral organs. Production of water-insoluble glucan (ISG) from sucrose by cell-free Streptococcus mutans AHT glucosyltransferase (GTF) first rapidly increased, and then sharply declined, as the amounts of water-soluble Dextrans T20 approximately T500 present, were increased. The decline of ISG synthesis was accompanied by an increased synthesis of the water-soluble fraction (SG). Prolonged incubation, however, induced enhanced synthesis of ISG even at higher dextran concentrations. The concentration of dextran required to stimulate or suppress ISG synthesis depended on the amounts of GTF used, but the extent of the stimulation was almost identical for the same GTF/dextran ratio. Thus, ISG synthesis is stimulated by the presence of dextrans at relatively low concentrations, but retarded at higher concentrations by being shifted to SG synthesis. ISG produced in the presence of dextrans contained higher proportions of alpha-1,6 glucosidic linkage and lower molecular size fractions, and possessed lower viscosity. These ISG products did not exhibit the coalescence of two component fibrils as observed with control ISG. These changes combined may contribute to the reduction of ISG-dependent adherence to glass of S. mutans cells by the presence of soluble dextrans, irrespective of their molecular size and concentration.  相似文献

Approximately 14 days after SHN, SLN and C3H/He strains of mice gave birth, their pups were removed. After 14--16 hours away from their pups, the dam was anesthetized, injected with oxytocin and milked. The milking device consisted of a rubber-stoppered centrifuge tube containing a stainless steel catheter and polyvinyl chloride tube. It was operated by vacuum.  相似文献

The application of enzymatic transformations is attracting increasing attention. Until recently, such an approach has generally been confined to producing fine chemicals difficult to obtain through conventional chemical methods. Microbial nitrile hydratase (NHase) has now been applied to the industrial, kiloton-scale production of the important chemical commodity acrylamide. Recent progress in understanding microbial nitrile metabolism at both the gene and protein levels is permitting improvement of the acrylamide production process.  相似文献

Peptide inhibitors of angiotensin I-converting enzyme (peptidyldipeptide hydrolase, EC 3.4.15.1) were produced by digesting gelatin with bacterial collagenase. The inhibitors were isolated from the digests with a combination of alcohol fractionation, treatment with Amberlite CG-50 column, gel filtration through Sephadex G-25, and Dowex 50 column and paper chromatography. Nine peptide fractions were purified to apparent homogeneity judging by thin-layer and ion-exchange column chromatography, and amino acid composition. Amino acid sequences of the peptides were determined: 2 were found to be mixtures of peptides and the sequence of another was only partially determined. Six of the peptides were potent inhibitors of the converting enzyme, while the other three were less active. 6 peptides were substrates for the enzyme. The enzyme released a dipeptide, Ala-Hyp from one peptide and was strongly inhibited by this dipeptide. The remainder of the parent peptides was a less effective inhibitor.  相似文献

We have investigated the limited proteolysis of the third component of complement, C3, by a human leukocyte protease, cathepsin G, by using a chemically modified C3, which was prepared by treatment of C3 with methylamine and a fluorescent thiol reagent, N-(dimethylamino-4-methylcoumarinyl)-maleimide (DACM) and was thus named DACM-C3me. Although native C3 was hardly cleaved by cathepsin G, DACM-C3me was cleaved by cathepsin G into three major fragments, which were termed C3c-G (150,000 daltons, 150 kd), C3d-G (25 kd), and C3a-G (10 kd). C3c-G was composed of four disulfide-linked polypeptide chains of 75 kd, 35 kd, and two 25 kd. C3d-G and C3a-G were single-chain fragments derived from the alpha chain. The N-terminal sequence of C3d-G was determined as Thr-Glu-Asp-Ala-Val-, suggesting that cathepsin G released C3d-G by cleaving a Met-Thr peptide bond which is located at 19 residues toward the N-terminal from the cysteinyl residue forming an internal thiolester linkage in native C3. C3d-G, like C3d-K (a C3d fragment produced by the action of plasma kallikrein), was found to have bioactivities such as leukocytosis-inducing and immunosuppressive activities.  相似文献
