Comparisons of selenite cystine (SC) and TT enrichment broths for detecting salmonellas were made with pure culture suspensions, with samples of naturally or artificially contaminated foods and with poultry feed. Selenite cystine recovered higher numbers of salmonellas from pure cultures and ground beef, while TT broth recovered higher numbers from pork sausage and poultry feed. Differences in the recovery of salmonellas from other food products appeared to be insignificant. The use of both SC and TT is thus recommended for maximal recovery of these organisms.  相似文献

BMAA (β-methylamino-L-alanine), a nonprotein amino acid with neurotoxic properties occurs in the cycad Cycas micronesica Hill in Guam. BMAA may have originally played a role as an antiherbivory compound in the plant, but is now of great interest because of its possible link to ALS-PDC, a neurological disease among the Chamorro people of Guam. Biomagnified cycad neurotoxins may play a role because they accumulate in flying foxes of the genus Pteropus that are eaten during traditional feasts. Since flying foxes feed on the seed sarcotesta it is important to understand the distribution of BMAA in the various tissues of the cycad. Using HPLC techniques, we quantified both BMAA and glutamic acid (GLU) in all cycad tissues. Although GLU is distributed randomly throughout the plant, BMAA is concentrated in cycad reproductive organs, with the highest concentrations being found in the immature staminate sporangium and the outmost layer of the sarcotesta. This finding is consistent with the putative evolutionary role of BMAA as an antiherbivory compound, as well as the biomagnification of the compound in flying foxes that ingest the seed sarcotesta.  © 2003 The Linnean Society of London, Botanical Journal of the Linnean Society , 2003, 143 , 165–168.  相似文献

ABSTRACT Although North American wood ducks (Aix sponsa) are well-studied throughout their range, researchers know little about demographic and environmental factors influencing survival of ducklings and broods, which is necessary information for population management. We studied radiomarked female and duckling wood ducks that used nest boxes and palustrine wetlands at Noxubee National Wildlife Refuge (NNWR) in Mississippi, USA, in 1996–1999, and riverine wetlands of the Tennessee-Tombigbee Rivers and Waterway (TTRW) system in Alabama in 1998–1999. We estimated survival of ducklings and broods and evaluated potentially important predictors of duckling survival, including age and body mass of brood-rearing females, hatch date of ducklings, duckling mass, brood size at nest departure, inter-day travel distance by ducklings, site and habitat use, and daily minimum air temperature and precipitation. At NNWR, survival of 300 radiomarked ducklings ranged from 0.15 (95% CI = 0.04-0.27) to 0.24 (95% CI = 0.13-0.38) and was 0.21 (95% CI = 0.15-0.28) for 1996–1999. Our overall estimate of brood survival was 0.64 (n = 91; 95% CI = 0.54-0.73). At TTRW, survival of 129 radiomarked ducklings was 0.29 in 1998 (95% CI = 0.20-0.41) and 1999 (95% CI = 0.13-0.45) and was 0.29 (95% CI = 0.20-0.40) for 1998–1999. Our overall estimate of brood survival was 0.71 (n = 38; 95% CI = 0.56-0.85). At NNWR, models that included all predictor variables best explained variation in duckling survival. Akaike weight (wi) for the best model was 0.81, suggesting it was superior to other models (<0.01 < wi < 0.18). We detected 4 competing models for duckling survival at TTRW. Inter-day distance traveled by ducklings was important as this variable appeared in all 4 models; duckling survival was positively related to this variable. Patterns of habitat-related survival were similar at both study areas. Ducklings in broods that used scrub-shrub habitats disjunct from wetlands containing aggregations of nest boxes had greater survival probabilities than birds remaining in wetlands with such nest structures. Managers may increase local wood duck recruitment by promoting availability of suitable brood habitats (e.g., scrub-shrub wetlands) without aggregations of nest boxes that may attract predators and by dispersing nest boxes amid or adjacent to these habitats. We did not determine an optimal density of nest boxes relative to local or regional population goals, which remains important research and conservation needs.  相似文献

It is demonstrated, without the use of special assumptions, that red cells are heterogeneous with respect to their resistance to at least certain lysins, that the reaction between the cell components and the lysin is virtually irreversible in some cases but reversible, although to different extents, in others, and that the lysin initiates a process in the cell which is not adequately described by the terms reversible and irreversible, but rather by the term progressive. Progressive reactions, i.e. reactions which cannot be stopped once they are well under way, may be looked for in systems which have structure, and in which local reactions occurring at strategic points lead to disproportionate results.  相似文献

SYNOPSIS. Tetrahymena pyriformis syngen 1, mating type II, (optimal growth temperature ∼ 37 C) ordinarily dies out in 5-14 days at 0-5 C. Dying cells were lumpy, suggesting membrane damage. By supplying crude soy lecithin, survival at 0-5 C was prolonged (after growth in peptone-yeast-dextrin) to at least 22 weeks. Crude soy sterols or sitosterol or stigmasterol, and antioxidant, e.g., Ionox 330 or ascorbylpalmitate, permitted survival of cells in suspension or in growth media for at least 16-22 weeks. These sterols are known to protect against triparanol toxicity, which suggested that triparanol, which blocks cholesterol synthesis in higher animals, might enhance cold-induced injury. Triparanol was more toxic at 0–5 than at 28 C for cell suspensions and cells in growth medium; this toxicity was annulled by crude soy lecithin or β-sitosterol, the only phytosterol tested. The synthetic medium intended as a control on the crude media became toxic at 0–5 C. Protection against cold damage is discussed as a means of elucidating the role of sterols—especially phytosterols—and other lipids in maintaining the integrity of the ciliate cell membrane.  相似文献

SYNOPSIS. In chemically defined media, pH 6.4–6.8, supplementation with glycerol permitted growth equal to that with a carbohydrate supplement. In a pH 5.4 medium described in detail, containing L-glutamic acid 0.25%, supplementation with L-serine (essential for this strain), especially when further supplemented with proline, likewise permitted growth nearly equal to that with carbohydrate. These results are discussed in relation to the question of which components of peptone act as substrates and the resemblances of the Tetrahymena pattern of substrate utilization to that of metazoa.  相似文献

ABSTRACT. The incidence of diapause in Ephestia kuehniella Zeller from an unhealed granary in Scotland was influenced by both photoperiod and temperature. At 25°C, nearly 50% of larvae entered diapause when reared in continuous darkness (DD) and up to 30% did so in short photoperiods. Little diapause was detected around LD 14:10 but a second, smaller peak of about 20% occurred at LD 16:8 and LD 18:6, falling away again to nearly zero in continuous light. More larvae entered diapause when reared continuously at 15 or 20°C than at 25 and 30°C. However, when larvae reared from hatch at 25°C in LD 16:8 were transferred after 1 week to 15°C in LD 9:15, almost twice as many entered diapause as did those reared at 15°C throughout. The sensitive phase for diapause induction occurred near the start of the final instar. The mean duration of diapause was between 2 and 3 months in most photoperiods at 20 and 25°C, and was shorter at 15°C. However, in DD at 25°C, it lasted about 7 months. Termination of diapause was hastened in larvae reared at 25°C in DD by transferring them to LD 14:10, and also by chilling them at 7.5°C for 6 weeks before returning to 25°C in DD. In an unhealed store in southern England, viable adults emerged from May to July and originated from larvae which terminated diapause relatively late. It would appear from the results of transferring larvae back to the laboratory at various times during the winter that some phases of diapause development were completed quite early after exposure to low temperatures, although no further development took place in the store until temperatures rose again in April.  相似文献
