Pulse radiolytically generated halogenated alkylperoxyl radicals, namely CCl3OO*, CBr3OO*, CHCl2OO*, CHBr2OO*, etc. have been employed for a study of the oxidation of beta-carotene in a quaternary microemulsion. All these halogenated alkylperoxyl radicals produce a radical cation (absorption maximum at 840 nm), either via the initial formation of an adduct (absorption maximum at 740 nm), or by direct reaction. There was a considerable blue shift of both absorption peaks in the present system as compared to those earlier reported in non-polar as well as in micellar media. An additional intense absorption peak at approximately 345 nm was noted at a longer time scale and was stable up to 5 ms. On irradiation with a large number of electron pulses, a stable product with an absorption maximum at 315 nm appeared. On excitation at 315 nm, this product gave a fluorescence spectrum with an emission maximum at 525 nm. The absorption and fluorescence spectra of the stable product compared with those of retinol; this was further confirmed by HPLC analysis. A suitable mechanism has been proposed. 相似文献

Thermostable direct hemolysin (TDH) is a ~19 kDa, hemolytic pore-forming toxin from the gram-negative marine bacterium Vibrio parahaemolyticus, one of the causative agents of seafood-borne acute gastroenteritis and septicemia. Previous studies have established that TDH exists as a tetrameric assembly in physiological state; however, there is limited knowledge regarding the molecular arrangement of its disordered N-terminal region (NTR)—the absence of which has been shown to compromise TDH's hemolytic and cytotoxic abilities. In our current study, we have employed single-particle cryo-electron microscopy to resolve the solution-state structures of wild-type TDH and a TDH construct with deletion of the NTR (NTD), in order to investigate structural aspects of NTR on the overall tetrameric architecture. We observed that both TDH and NTD electron density maps, resolved at global resolutions of 4.5 and 4.2 Å, respectively, showed good correlation in their respective oligomeric architecture. Additionally, we were able to locate extra densities near the pore opening of TDH which might correspond to the disordered NTR. Surprisingly, under cryogenic conditions, we were also able to observe novel supramolecular assemblies of TDH tetramers, which we were able to resolve to 4.3 Å. We further investigated the tetrameric and inter-tetrameric interaction interfaces to elaborate upon the key residues involved in both TDH tetramers and TDH super assemblies. Our current structural study will aid in understanding the mechanistic aspects of this pore-forming toxin and the role of its disordered NTR in membrane interaction. 相似文献

Simple predator-prey models with a prey-dependent functional response predict that enrichment (increased carrying capacity) destabilizes community dynamics: this is the 'paradox of enrichment'. However, the energy value of prey is very important in this context. The intraspecific chemical composition of prey species determines its energy value as a food for the potential predator. Theoretical and experimental studies establish that variable chemical composition of prey affects the predator-prey dynamics. Recently, experimental and theoretical approaches have been made to incorporate explicitly the stoichiometric heterogeneity of simple predator-prey systems. Following the results of the previous experimental and theoretical advances, in this article we propose a simple phenomenological formulation of the variation of energy value at increased level of carrying capacity. Results of our study demonstrate that coupling the parameters representing the phenomenological energy value and carrying capacity in a realistic way, may avoid destabilization of community dynamics following enrichment. Additionally, under such coupling the producer-grazer system persists for only an intermediate zone of production--a result consistent with recent studies. We suggest that, while addressing the issue of enrichment in a general predator-prey model, the phenomenological relationship that we propose here might be applicable to avoid Rosenzweig's paradox. 相似文献

We explore the mutual dependencies and interactions among different groups of species of the plankton population, based on
an analysis of the long-term field observations carried out by our group in the North–West coast of the Bay of Bengal. The
plankton community is structured into three groups of species, namely, non-toxic phytoplankton (NTP), toxic phytoplankton
(TPP) and zooplankton. To find the pair-wise dependencies among the three groups of plankton, Pearson and partial correlation
coefficients are calculated. To explore the simultaneous interaction among all the three groups, a time series analysis is
performed. Following an Expectation Maximization (E-M) algorithm, those data points which are missing due to irregularities
in sampling are estimated, and with the completed data set a Vector Auto-Regressive (VAR) model is analyzed. The overall analysis
demonstrates that toxin-producing phytoplankton play two distinct roles: the inhibition on consumption of toxic substances
reduces the abundance of zooplankton, and the toxic materials released by TPP significantly compensate for the competitive
disadvantages among phytoplankton species. Our study suggests that the presence of TPP might be a possible cause for the generation
of a complex interaction among the large number of phytoplankton and zooplankton species that might be responsible for the
prolonged coexistence of the plankton species in a fluctuating biomass. 相似文献
