[6-3H]glucose was used for measurements of plasma glucose turnover in adult non-growing mice with sarcoma-induced malnutrition. Plasma glucose turnover was increased in freely-fed tumour-bearing mice but not in starved tumour-bearing mice. Elevated glucose turnover in freely-eating sarcoma-bearing mice was associated with an increased glucose pool, the latter not only being due to an expanded distribution volume of glucose. Plasma lactate was elevated in sarcoma-bearing mice independently of their food intake. The results show that elevated plasma glucose turnover in an anorectic tumour-bearing host is not due to insufficient adaptation to food depression. Increased glucose turnover in tumour-bearing mice was associated with an altered intermediary whole body metabolism of glucose which is not entirely accounted for by the state of malnutrition of the host.  相似文献

Gravitropically-stimulated seedlings show autotropism in weightlessness   总被引:1,自引:0,他引:1  
In a spaceflight experiment, autotropism by oat ( Avena sativa L.) coleoptiles following gravitropic responses was prominent in weightlessness: counter-reactions led to the straightening of the curved coleoptiles. This was not the case during clinorotation on earth. The autotropic reactions appeared to be related to the stimulus received during the stimulus period, i.e. the greater the response the greater the autotropic counter-reaction. Previous models of the gravitropic system which predicted that coleoptiles would not straighten in weightlessness are disproved. A modification to one of the models is proposed which includes the autotropic response observed in spaceflight. The nature of the counter-reactions in the absence of gravitropic stimulation is discussed.  相似文献

Seasonal differences between moose and roe deer in ability to digest browse   总被引:1,自引:0,他引:1  
The digestive capacity of free-living moose Alces alces (L.) and roe deer Capreolus capreolus (L.) for four plant species was studied with in vitro technique. Moose were found to digest all species better than roe deer. Individual variations were also smaller for moose. In moose the level of digestibility was the same both summer and winter, while roe deer shot in winter digested winter samples of bilberry Vaccinium myrtillus L. and to some extent Scots pine Pinus silvestris L. better than did summer roe deer. This was taken as evidence for a seasonal adaptation in the roe deer rumen. Digestibility of the plant material studied showed a strong negative correlation to the fiber content. The results are discussed in relation to winter problems for roe deer and possible food competition between moose and roe deer.  相似文献

Fragments from section 3 of the salivary gland X chromosome of D. melanogaster were dissected with a micromanipulator. The DNA was extracted, cut and ligated to a λ vector in a volume of a few nanoliters in an oil chamber monitored through a microscope. From about 10 pg of DNA we obtained 80 recombinant clones, a sample of which were analysed and shown to contain Drosophila DNA which hybridises in situ to the region of section 3 of the X chromosome. With this technique we can isolate clones from any desired region as small as 200 kb from the euchromatic arms of polytene chromosomes. This paper is dedicated to Professor W. Summary The effects of chlorpromazine (cpz) and tetracaine (tc) on the rapid axonal transport of neurosecretory material (NSM) in the hypothalamo-neurohypophysial system was investigated. Following subarachnoidal injection of these drugs, the incorporation of (35S) cysteine into proteins of the supraoptic nucleus was slightly depressed. The protein-bound radioactivity in the posterior pituitary was markedly lowered in experimental rats which indicates a partial blockage of the rapid axonal transport of NSM along the hypothalamoneurohypophysial tract. Both cpz and tc induced an increase in the number of mitochondria and profiles of granular endoplasmic reticulum. The axons in the infundibulum and neurohypophysis were enlarged by dammed organelles, indicating a blockage of axonal transport. There was an increased number of microvesicles, often arranged in a crystalloid pattern, in the terminals. The number and distribution of neurotubuli and neurofilaments were not changed. A possible stimulatory effect of cpz on the release of NSM from the neural lobe is assumed Possible mechanisms for the action of mitotic inhibitors, transquilizers and local anesthetics are discussed.The present work was supported by grants from Svenska Livforsäkringsbolags Fond, H. Francisella tularensis is a facultative intracellular bacterium that survives and multiplies inside macrophages. Here we constructed a new promoter probe plasmid denoted pKK214 by introduction of a promoter-less chloramphenicol acetyltransferase (cat) gene into the shuttle vector pKK202. A promoter library was created in F. tularensis strain LVS by cloning random chromosomal DNA fragments into pKK214. Approximately 15% of the recombinant bacteria showed chloramphenicol resistance in vitro. The promoter library was also used to infect macrophages in the presence of chloramphenicol and after two cycles of infection the library contained essentially only chloramphenicol resistance clones which shows that pKK214 can be used to monitor F. tularensis genes that are expressed during infection.  相似文献

Chromosome terminal, complex repeats in the dipteran Chironomus pallidivittatus show rapid concerted evolution during which there is remarkably efficient homogenization of the repeat units within and between chromosome ends. It has been shown previously that gene conversion is likely to be an important component during these changes. The sequence evolution could be a result of different processes—exchanges between repeats in the tandem array as well as information transfer between units in different chromosomes—and is therefore difficult to analyze in detail. In this study the concerted evolution of a region present only once per chromosome, at the junction between the telomeric complex repeats and the subtelomeric DNA was therefore investigated in the two sibling species C. pallidivittatus and C. tentans. Material from individual microdissected chromosome ends was used, as well as clones from bulk genomic DNA. On the telomeric side of the border pronounced species-specific sequence differences were observed, the patterns being similar for clones of different origin within each species. Mutations had been transmitted efficiently between chromosomes also when adjoining, more distally localized DNA showed great differences in sequence, suggesting that gene conversion had taken place. The evolving telomeric region bordered proximally to subtelomeric DNA with high evolutionary constancy. More proximally localized, subtelomeric DNA evolved more rapidly and showed heterogeneity between species and chromosomes. Received: 24 September 1997 / Accepted: 24 November 1997  相似文献

The closely related B-subunits of cholera toxin (CTB) and Escherichia coli heat-labile enterotoxin (LTB) both bind strongly to GM1 ganglioside receptors but LTB can also bind to additional glycolipids and glycoproteins. A number of mutant CT B-subunits were generated by substituting CTB amino acids with those at the corresponding positions in LTB. These were used to investigate the influence of specific residues on receptor-binding specificity. A mutated CTB protein containing the first 25 residues of LTB in combination with LTB residues at positions 94 and 95, bound to the same extent as native LTB to both delipidized rabbit intestinal cell membranes, complex glycosphingolipids (polyglycosylceramides) and neolactotetraosylceramide, but not to non-GM1 intestinal glycosphingolipids. In contrast, when LTB amino acid substitutions in the 1–25 region were combined with those in the 75–83 region, a binding as strong as that of LTB to intestinal glycosphingolipids was observed. In addition, a mutant LTB with a single Gly-33→Asp substitution that completely lacked affinity for both GM1 and non-GM1 glycosphingolipids could still bind to receptors in the intestinal cell membranes and to polyglycosylceramides. We conclude that the extra, non-GM1 receptors for LTB consist of both sialylated and non-sialylated glycoconjugates, and that the binding to either class of receptors is influenced by different amino acid residues within the protein.  相似文献
