A face-centered central composite design was applied to optimize a cultivation condition for improved beta-carotene production by Rhodotorula glutinis DM28 in a stirred tank reactor using 30 g/l total soluble solid of fermented radish brine as a sole substrate. The experiments were performed with regression models, where temperature, pH and dissolved oxygen were considered as variables. Results showed that an optimum condition for beta-carotene production of the yeast was at 30 degrees C, pH 6 and 80% dissolved oxygen. Under this condition, the yeast yielded 2.7 g/l biomass and the maximum beta-carotene of 201 microg/l after 24-h fermentation indicating approximately 15% higher than those under an initial condition (2.3g/l and 178 microg/l, respectively). 相似文献

Summary Isolation and identification of a thermotolerant feather-degrading bacterial strain from Thai soil as well as purification
and properties of its keratinase were investigated. The thermotolerant bacterium was identified as Bacillus licheniformis. The keratinase was purified to homogeneity by three-step chromatography. The purified enzyme exhibited a high specific activity
(218 U mg−1) with 86-fold purification and 25% yield. The enzyme was monomeric and had a molecular mass of 35 kDa. The optimum pH and
temperature for the enzyme were 8.5 and 60 °C, respectively. The enzyme activity was significantly inhibited by PMSF and partly
inhibited by EDTA and iodoacetamide, but was stimulated by metal ions. It hydrolysed soluble proteins with a relative activity
of 4–100% and insoluble proteins, including keratins, with a relative activity of 3–35%. Therefore, the enzyme could improve
the nutritional value of meat- and poultry-processing wastes containing keratins, collagen and gelatin. 相似文献

Thirty-eight l(+)-lactic acid-overproducing mutants were isolated by mutagenizing a parent strain of Rhizopus oryzae NRRL 395 with u.v. and/or N -methyl- N '-nitro- N -nitrosoguanidine. Three mutants, 1N1, 3N4 and 3N6, were found to yield significantly more l(+)-lactic acid in a rice medium than the parent strain. Of the mutants examined, only mutant 3N4 produced more glucoamylase than the parent strain. 相似文献

Enumeration of Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus is a priority due to their importance in yogurt production. Capillary electrophoresis (CE) of both bacteria could be achieved in 7.2 min with a resolution of 3.2 in the background electrolyte (BGE) containing 4.5 mM Tris(hydroxymethyl) amminomethane (TRIS)–4.5 mM boric acid–0.1 mM ethylenediamine tetraacetate (EDTA) (TBE) buffer (pH 8.4) and 0.05% (v/v) polyethylene oxide (PEO), using a capillary of 47.5 cm (effective length) × 100 μm i.d., injection of 50 mbar × 3 s followed by ?5 kV × 120 s, a voltage and temperature of 20 kV and 25 °C, respectively. Appropriate amounts of PEO in the BGE, sample preparation (i.e. vortex) and introduction were key factors for their separation. A short hydrodynamic injection followed by applying reversed polarity voltage could compress the bacteria into narrow zones, which were detected as separated single peaks. Method linearity (r2 > 0.99), precision (%RSDs < 9.3%), recovery (%R = 91.7–106.7%) and limit of quantitation (1.0 × 106 colony forming unit per mL (CFU/mL)) were satisfactory. Results from the CE analysis of both bacteria in yogurt were not statistically different from those of the plate count method (P > 0.05). The CE method can be used as an alternative for quantitation of L. delbrueckii subsp. bulgaricus and S. thermophilus in yogurt since it was reliable, simple, cost and labor effective and rapid, allowing the analysis of 3 samples/h (comparing to 2d/sample by plate count method). 相似文献

An acid proteinase from Monascus purpureus No. 3403, MpuAP, was previously purified and some characterized in our laboratory (Agric Biol Chem 48:1637–1639, 1984). However, further information about this enzyme is lacking. In this study, we investigated MpuAP’s comprehensive substrate
specificity, storage stability, and prospects for reducing antigenicity of whey proteins for application in the food industry.
MpuAP hydrolyzed primarily five peptide bonds, Gln4–His5, His10–Leu11, Ala14–Leu15, Gly23–Phe24 and Phe24–Phe25 in the oxidized insulin B-chain. The lyophilized form of the enzyme was well preserved at 30–40°C for 7 days without stabilizers.
To investigate the possibility of reducing the antigenicity of the milk whey protein, enzymatic hydrolysates of the whey protein
were evaluated by inhibition ELISA. Out of the three main components of whey protein, casein and α-lactalbumin were efficiently
degraded by MpuAP. The sequential reaction of MpuAP and trypsin against the whey protein successfully degraded casein, α-lactalbumin
and β-lactoglobulin with the highest degree of hydrolysis. As a result, the hydrolysates obtained by using the MpuAP–trypsin
combination showed the lowest antigenicity compared with the single application of pepsin, trypsin or pepsin–trypsin combination.
Therefore, the overall result suggested that the storage-stable MpuAP and trypsin combination will be a productive approach
for making hypoallergic bovine milk whey protein hydrolysates. 相似文献

Cultivation conditions affecting feather degradation by Bacillus sp. FK 46 were investigated. The results showed that feather was almost completely degraded under the following conditions: 1% whole chicken feather as a substrate at the initial medium pH of 9 with 5% bacterial inoculum, at a temperature of 37 degrees C and a shaking speed of 250 rev/min. Glucose, methanol, Tween 80 and Triton X-100, however, had no effect on feather degradation. After feather was degraded, its residue and fermented broth would become a protein feed for animals. 相似文献

Fermentation kinetics of growth and β-carotene production by Rhodotorula glutinis DM28 in batch and continuous cultures using fermented radish brine, a waste generated from fermented vegetable industry, as a cultivation medium were investigated. The suitable brine concentration for β-carotene production by R. glutinis DM28 was 30 g l?1. Its growth and β-carotene production obtained by batch culture in shake flasks were 2.2 g l?1 and 87 μg l?1, respectively, while, in a bioreactor were 2.6 g l?1 and 186 μg l?1, respectively. Furthermore, its maximum growth rate and β-carotene productivity in continuous culture obtained at the dilution rate of 0.24 h?1 were 0.3 g l?1 h?1 and 19 μg l?1 h?1, respectively, which were significantly higher than those in the batch. Therefore, improved growth rate and β-carotene productivity of R. glutinis in fermented radish brine could be accomplished by continuous cultivation. 相似文献

The use of lettuce brine, a by-product of the vegetable fermentation industry, as a medium for yeast cultivation was investigated.
Six strains of yeast, Saccharomyces sp., Pichia sp., Rhodotorula sp., Candida sp., Kluyveromyces sp. and Trichospora sp. grew well in diluted lettuce brine under aerobic conditions. The acid brine becomes neutral after yeast cultivation.
The yeast strains reached the maximum growth after the first day of cultivation. Trichosporon sp. was found to grow best in the brine with the maximum specific growth rate at 0.09 h−1 and growth yield of 67%.

Potato chip processing waste of trimmed potato, potato peel and substandard (low-quality) potato chips, obtained from a potato
chip processing plant, were used as substrates for chitosan production from Rhizopus oryzae. It was cultured on each waste product at 30 ± 2°C and 70% moisture content for 21 days. Fermented potato peel had the highest
yield after 5 days of fermentation. The cultivation condition of chitosan obtained from R. oryzae was optimum for a peel size of less than 6 mesh, 70% moisture content and a pH of 5. Furthermore, the best extraction condition
was using 46% sodium hydroxide at 46°C for 13 h followed by 2% acetic acid at 95°C for 8 h. The maximum chitosan yield obtained
by these conditions was 10.8 g/kg substrate. Fungal chitosan properties were found to be 86–90% degree of deacetylation, molecular
weight of 80–128 kDa and viscosity of 3.1–6.1 mPa s. Therefore, potato peel could be applied as a low cost substrate for chitosan

AIMS: The objective of this investigation was to evaluate the chitosans produced by several species of fungi. METHODS AND RESULTS: Representatives of four species of filamentous fungi, Aspergillus niger, Rhizopus oryzae, Lentinus edodes and Pleurotus sajo-caju, and two yeast strains, Zygosaccharomyces rouxii TISTR5058 and Candida albicans TISTR5239, were investigated for their ability to produce chitosan in complex media. Fungal chitosan was produced at 10-140 mg g-1 cell dry weight, had a degree of deacetylation of 84-90% and a molecular weight of 2.7 x 104-1.9 x 105 Da with a viscosity of 3.1-6.2 centipoises (cP). CONCLUSIONS: Rhizopus oryzae TISTR3189 was found to be the producer of the highest amounts of chitosan. SIGNIFICANCE AND IMPACT OF THE STUDY: Commercial chitosan could be obtained from Rhizopus mycelia and would have potential applications for medical and agricultural uses. 相似文献
