An improved method of cell fractionation allowed the extraction of soluble (sNR) and membrane-associated (mNR) forms of nitrate reductase (NR) from a dinoflagellate, even though in previous studies only mNR had been found in these algae. Both activities were assayed in cell-free extracts of Peridinium gatunense from Lake Kinneret, Israel, after disruption of the cells and differential centrifugation. In the cultures used, sNR showed much higher NO3 − -reducing activity. Only a low proportion, 2.5–3% of NR activity, was found to be associated with mNR. Moreover, mNR comprised two forms as indicated by protein solubilization: a tightly membrane-bound and a more weakly attached NR. Ascorbate inhibited all NR activities, but that of mNR recovered after its removal. Polyvinyl pyrrolidone (PVP) and DTT also diminished sNR and mNR activities. For both enzymes, pH optima (7.65) and temperature optima (13–25°C) were similar, and agreed with those for optimum growth of P. gatunense both in culture and in the lake. The most efficient electron donor was NADH, though NADPH sustained low NR activities. Carboxylic anions such as succinate and malate did not support any reduction of NO3 − , nor did they cause any stimulation of sNR or mNR activities. Both forms of NR showed a high affinity for their substrates: K m was c. 10 μM for NO3 − and c. 5 μM for NADH. The high efficiency of NO3 − assimilation by Peridinium seems to be limited mainly by energy under otherwise optimal nutritional conditions and, at low nitrate concentrations, the low K m may be one of the main reasons for the high competitivity of this alga in Lake Kinneret. 相似文献

One potential approach to combat the impacts of climate change is the expansion of renewable energy installations, leading to an increase in the number of wave‐powered marine renewable energy installations (MREIs). The consequences of increased use of these devices for birds are unknown. Here we describe the wave‐powered energy‐generating devices currently either operational or in development and review the potential threats and benefits of these to marine birds, their habitats and prey. Direct negative effects include risk of collision, disturbance, displacement and redirection during construction, operation and decommissioning. Above‐water collision is a particular concern with wind‐powered devices, but, because of their low profiles, the collision risk associated with wave‐powered devices is likely to be much lower. Conversely, wave devices also pose the novel threat of underwater collision. Wave‐energy‐generating devices may indirectly impact marine birds by altering oceanographic processes and food availability, with implications for trophic cascades. Through appropriate mitigation, wave‐powered MREIs offer the potential to enhance habitats. Direct positive effects may include provision of roosting sites, and indirect positive effects may include prey aggregation due to suitable substrates for sessile organisms or because they act as de facto protected areas. The cumulative effect of these could be the improvement and protection of foraging opportunities for marine birds. Recent studies have been critical of the methods used in the assessment of wind‐powered MREI impacts, which lack sufficient sample sizes, controls or pre‐development comparisons. Here we suggest solutions for the design of future studies into the effects of MREIs. Wave‐powered MREIs are certain to become part of the marine environment, but with appropriate planning, mitigation and monitoring they have the potential to offer benefits to marine birds in the future. 相似文献

Uptake of a number of suiphonarnides and p-aminobenzoic andsuiphanilic acids can be represented by equations derived fromPick's Law of diffusion. The diffusion constants so derivedhave a Q10 of the order of 1.4 or 1.7, indicating that the processis primarily physical. The Fick's Law equations provide a measureof the permeability of the system and of the size of the effectivevolume into which the compound is diffusing. Assuming that thebasic process is diffusion from water outside the tissue intowater inside the tissue, the size of the effective volume isconditioned by the absorption of the compound from the aqueousphase of the free space. With sulphanilaniide the effectivevolume is finite while with p-amino benzoic acid the effectivevolume is infinite within the limits of the experiment and concentrationsup to three times that of the treating solution may be attained.A proportion of the suiphanilamide will diffuse out of shootsimmersed in water but p-aminobenzoic acid is held irreversiblyby the tissue. 相似文献

A laser beam of 6.5-1 joule is directed for 1 msec at dorsalsurfaces of adult female Araneus diadematus Cl. spiders. Thelesion is located by means of histological methods, and relationshipof its position to behavioral consequencesas measuredby changes in the web-patternare studied. Twelve animalswith lesions in the area of the supra-esophageal ganglion showdifferent degrees of extensive web-pattern disturbances consistingof smaller and rounder webs with less detail and irregular spacingof threads. In M. braunii, the uptake of NO3− and NO2− is blue-light-dependent and is associated with alkalinization of the medium. In unbuffered cell suspensions irradiated with red light under a CO2-free atmosphere, the pH started to rise 10s after the exposure to blue light. When the cellular NO3− and NO2− reductases were active, the pH increased to values of around 10, since the NH4+ generated was released to the medium. When the blue light was switched off, the pH stopped increasing within 60 to 90s and remained unchanged under background red illumination. Titration with H2SO4 of NO3− or NO2− uptake and reduction showed that two protons were consumed for every one NH4+ released. The uptake of Cl− was also triggered by blue light with a similar 10 s time response. However, the Cl− -dependent alkalinization ceased after about 3 min of blue light irradiation. When the blue light was turned off, the pH immediately (15 to 30 s) started to decline to the pre-adjusted value, indicating that the protons (and presumably the Cl−) taken up by the cells were released to the medium. When the cells lacked NO3− and NO2− reductases, the shape of the alkalinization traces in the presence of NO3− and NO2− was similar to that in the presence of Cl−, suggesting that NO3− or NO2− was also released to the medium. Both the NO3− and Cl−-dependent rates of alkalinization were independent of mono- and divalent cations. 相似文献

A large number of proposed biofuel crops share the same traits as known invasive plant species, many of which are already present in Africa and include species such as Prosopis glandulosa Torrey (Mimosaceae), P. juliflora (Sw.) DC, Leucaena leucocephala (Lam.) de Wit (Mimosaceae), Azadirachta indica A. Juss. (Meliaceae), and others. In this paper, we mainly assess the impacts of invasive Prosopis species in Africa, particularly in Kenya and South Africa. Introduced Prosopis species have invaded over 4 million hectares in Africa, threatening crop and pasture production, reducing underground water reserves, and displacing native plant and animal species. This has major implications for millions of people who depend on natural resources for their survival. It is therefore suggested that known invasive or potentially invasive plant species not be introduced to countries or regions for biofuel production. If (after a stringent cost–benefit analysis) the introduction of a potentially invasive species is deemed critical for economic development and the benefits clearly outweigh the potential costs, countries should endeavour to abide by the Principles and Criteria for Sustainable Biofuel Production developed by the Roundtable on Sustainable Biofuels. 相似文献
