Background  

The Immunoglobulin heavy chain (IgH) 3' Regulatory Region (3'RR), located at the 3' of the constant alpha gene, plays a crucial role in immunoglobulin production. In humans, there are 2 copies of the 3'RR, each composed of 4 main elements: 3 enhancers and a 20 bp tandem repeat. The single mouse 3'RR differs from the two human ones for the presence of 4 more regulative elements with the double copy of one enhancer at the border of a palindromic region.  相似文献

Injection of tumor cells in mice more than 30 years ago resulted in the discovery of an epithelial antigen, later defined as a cell adhesion molecule (EpCAM). Although EpCAM has since evoked significant interest as a target in cancer therapy, mechanistic insights on the functions of this glycoprotein have been emerging only very recently. This may have been caused by the multitude of functions attributed to the glycoprotein, its localization at different subcellular sites and complex posttranslational modifications. Here, we review how EpCAM modifies cell–cell contact adhesion strength and tissue plasticity, and how it regulates cell proliferation and differentiation. Major knowledge derived from human diseases will be highlighted: Mutant EpCAM that is absent from the cell surface leads to fatal intestinal abnormalities (congenital tufting enteropathy). EpCAM-mediated cell proliferation in cancer may result from signaling (i) via regulated intramembrane proteolysis and/or (ii) the localization and association with binding partners in specialized membrane microdomains. New insight in EpCAM signaling will help to develop optimized cancer therapies and open new avenues in the field of regenerative medicine.  相似文献

The centromere is an essential structure in the chromosomes of all eukariotes and is central to the mechanism that ensures proper segregation during mitosis and meiosis. The comparison of DNA sequence motifs, organization and kinetocore components from yeast to man is beginning to indicate that, although centromeres are highly variable DNA elements, a conserved pattern of sequence arrangement and function is emerging. We have identified and characterized the first satellite DNA (P.k.SAT) from microbat species Pipistrellus kuhli. The presence of mammalian CENP-B box and yeast CDEIII box could indicate the participation of P.k.SAT in centromere organization.  相似文献

The aim of this study was to investigate the productive traits and fatty acid profiles of eight sweet varieties of Lupinus albus, Lupinus angustifolius and Lupinus luteus cultivated in a Mediterranean environment (Sicily, southern Italy). This is being done to identify the most suitable species, from an agronomical and nutritional point of view, for supporting animal husbandry in this area. Seed yield, oil quantity and oil quality were considerably different depending on the genotype. L. albus showed the highest yield (2.0–2.4 t/ha), 100 seed weight (27.0 g, on average), oil content (70.4–75.5 g/kg), and n?3/n?6 polyunsaturated fatty acid ratio (0.39–0.51), L. angustifolius showed the highest content of saturated fatty acids (21.03–26.66 g/100 g) and L. luteus showed the highest levels of n?6 polyunsaturated fatty acids (47.56–50.85 g/100 g), unsaturated/saturated (4.93–5.62) and polyunsaturated/saturated (3.33–3.99) fatty acid ratios and the best atherogenic (0.06) and thrombogenic (0.09–0.11) indices. The data obtained suggests L. albus as the most interesting lupin species for aiding the crop-livestock food chain and L. luteus as a promising crop due to its high nutritive traits for the Mediterranean environment.  相似文献

The major chaperone genes of Helicobacter pylori are negatively regulated by HspR, a homologue of the repressor of the dnaK operon of Streptomyces coelicolor. Using an in vitro selection and amplification approach we identified two new chromosomal binding sites of the HspR protein. Both binding sites were characterized by footprinting analysis with purified HspR protein. Intriguingly, these HspR binding sites are located at the 3prime prime or minute ends of two genes coding for predicted proteins with functions unrelated to those of chaperones. This suggests that H. pylori HspR may regulate the expression of genes encoding proteins with diverse functions. Nucleotide sequence alignment of HspR-binding sites highlights conserved nucleotides extending outside the previously proposed consensus binding sequence with structural features predicting geometry of HspR binding as an oligomer.  相似文献
