Summary The stability of allelic gene expression of X-linked phosphoglycerate kinase was studied in seven carriers of a rare genetic variant named PGK München. The enzymatic activities in erythrocytes of five heterozygous females and three hemizygous males were determined repeatedly over a period of 10 years (1975–1984) and shown to remain constant. As the phosphoglycerate kinase activity is lower in cells expressing the PGK München allele, the ratio of the two cell types in all heterozygous females of the PGK München kindred could be calculated from the PGK activity and from the known allozyme activities in erythrocytes of homozygous wild type or hemizygous PGK München carriers. Since the maternal or paternal origin of both alleles is known from the pedigree, the quantitative expression of the maternally derived allozyme in heterozygous women could be determined. In heterozygous carriers the cell pool expressing the maternally inherited allele was significantly increased, independently, of the PGK allele linked to the maternal X chromosome (P<0.001). Our data show that inactivation of one of the two X chromosomes in human female erythropoietic stem cell precursors may be non-random, at least in the kindred and cell populations described here. Uridine phosphorylase activity has been used to detect mycoplasmas in cell cultures by measuring formation of14C-uracil from14C-uridine. In this report we show that all species ofMycoplasma, Acholeplasma, andUreaplasma tested exhibited uridine phorphorylase activity. Among the genusSpiroplasma, serogroups I-1, I-3, I-5, I-7, I-8, IV, XIII, and XIV lacked uridine phosphorylase activity.Present address: Ciba-Geigy, Basel, Switzerland.  相似文献

Summary In order to investigate whether during mitosis of guard cell mother cells the plastids are distributed to the daughter cells at random, a haploid of Trifolium hybridum, a species with only three to four chloroplasts in one diploid guard cell, was searched for and found. As expected, the guard cell mother cells in this plant contained only about two plastids. If distribution to the daughter cells would occur strictly at random, among the guard cell pairs with two chloroplasts the pairs with 1/1 and those with 2/0 chloroplasts should appear in equal amounts. However, 159 pairs of type 1/1 and only 35 pairs of type 2/0 were found, i.e., 18% of type 2/0 (upper limit of 99% confidence interval: 25%), indicating that the plastids have been apportioned to a fairly great degree. The result may be understood by considering that the plastids in guard cell mother cells are not scattered at random throughout the cell space, but are more regularly spread as are the chloroplasts in adult cells.  相似文献

Comparative electron microscope investigations on sections of the lens cortex of the normal, mature rat, rabbit, monkey, and the normal calf reveal similar patterns of intracellular organization. The superficial lens fiber contains a nucleus, mitochondria, endoplasmic reticulum, dense granules, Golgi complex, and a quantity of small structures of low opacity which appear as filamentous and spherical configurations. Variations in number, distribution, and spatial arrangement of cytoplasmic elements in lens fibers are described. These changes in the pattern of cytoplasmic organization are concomitant with development of fibers and their displacement towards the center of the lens. Structural details of the various zones of the lens epithelium and the lens fibers are compared.  相似文献

Summary The hippocampal region of the rat has been studied with the histochemical fluorescence method for demonstration of catecholamines and 5-hydroxytryptamine. Noradrenalin nerve terminals of characteristic appearance were observed throughout the region, but the density of the terminals in the various areas differed considerably. The highest amount of terminals were seen in the hilus of the area dentata of the rat. Otherwise, noradrenalin nerve terminals constitute a small percentage of the afferents to the hippocampal region. Noticeable differences existed between rat and guinea-pig. In both species there was a conspicuous lack of stratification and delineation of areas as compared with the pictures seen with most other methods, which have revealed precise architectonic patterns.Supported by a Research Grant (12X-715-02) from the Swedish Medical Research council and by a Grant from Stiftelsen M. Summary For the mapping of the terminal area of transected axons within the central nervous system, electron microscopy has recently been adopted. A greater accuracy is thereby obtained than with silver impregnation and light microscopy, since it becomes possible to determine the kinds of structure (soma, dendrites, spines) with which the degenerating boutons establish synaptic contact. In the present study this technique was extended by Golgi impregnation of such material with the aim of making possible classification of the postsynaptic neuron. A few days after transection of a pathway (commissural fibres to the hippocampus being used as a model in this study) the brain was fixed by perfusion with phosphate buffered formalin with sucrose. This was followed by immersion in an osmium tetroxide-potassium dichromate mixture (Dalton's fixative without sodium chloride) later replaced by a solution of silver nitrate. Satisfactory Golgi impregnation of nerve cells and processes was obtained. By careful trimming, and reembedding of selected areas, blocks for ultramicrotomy could be obtained which contained only one type of impregnated cell, e.g., hippocampal pyramidal cells.The relation of basal dendrites of such cells to degenerating boutons of commissural fibres was studied. Numerous examples of contact between degenerating boutons and spines belonging to the basal dendrites were seen. Although the Golgi precipitate obscured the postsynaptic substance in the spines, a number of features at the sites of contact were considered strong indication that many of the contacts were synapses and not merely the result of random juxtaposition. Benzothiadiazole (BTH) is a novel chemical activator of disease resistance in tobacco, wheat and other important agricultural plants. In this report, it is shown that BTH works by activating SAR in Arabidopsis thaliana. BTH-treated plants were resistant to infection by turnip crinkle virus, Pseudomonas syringae pv ‘tomato’ DC3000 and Peronospora parasitica. Chemical treatment induced accumulation of mRNAs from the SAR-associated genes, PR-1, PR-2 and PR-5. BTH treatment induced both PR-1 mRNA accumulation and resistance against P. parasitica in the ethylene response mutants, etr1 and ein2, and in the methyl jasmonate-insensitive mutant, jar1, suggesting that BTH action is independent of these plant hormones. BTH treatment also induced both PR-1 mRNA accumulation and P. parasitica resistance in transgenic Arabidopsis plants expressing the nahG gene, suggesting that BTH action does not require salicylic acid accumulation. However, because BTH-treatment failed to induce either PR-1 mRNA accumulation or P. parasitica resistance in the non-inducible immunity mutant, nim1, it appears that BTH activates the SAR signal transduction pathway.  相似文献
