We have previously demonstrated that introgression of PcINO1 gene from Porteresia coarctata (Roxb.) Tateoka, coding for a novel salt-tolerant L-myo-inositol 1-phosphate synthase (MIPS) protein, confers salt tolerance to transgenic tobacco plants (Majee, M., Maitra, S., Dastidar, K.G., Pattnaik, S., Chatterjee, A., Hait, N.C., Das, K.P. and Majumder, A.L. (2004) A novel salt-tolerant L-myo-inositol-1-phosphate synthase from Porteresia coarctata (Roxb.) Tateoka, a halophytic wild rice: molecular cloning, bacterial overexpression, characterization, and functional introgression into tobacco-conferring salt-tolerance phenotype. J. Biol. Chem. 279, 28539-28552). In this communication we have shown that functional introgression of the PcINO1 gene confers salt-tolerance to evolutionary diverse organisms from prokaryotes to eukaryotes including crop plants albeit to a variable extent. A direct correlation between unabated increased synthesis of inositol under salinity stress by the PcINO1 gene product and salt tolerance has been demonstrated for all the systems pointing towards the universality of the application across evolutionary divergent taxa. 相似文献

In an unusual reaction of [RuIII(acac)2(CH3CN)2](ClO4) ([1], acac = acetylacetonate) and aniline (Ph-NH2), resulted in the formation of ortho-semidine due to dimerisation of aniline via oxidative ortho-Carom-N bond formation reaction. This oxidation reaction is associated with stepwise chlorination of coordinated acac ligands at the γ-carbon atom resulting in the formation of [RuIII(acac)2L−] [2a], [RuIII(Cl-acac)(acac)L−] [2b], [RuIII(acac)(Cl-acac)L−] [2c] and [RuIII(Cl-acac)2L−] [2d] (L− = N-phenyl-ortho-semiquinonediimine) complexes, respectively. These have been characterized by 1H NMR, UV-Vis-NIR, ESI-MS and cyclic voltammetry studies. Single crystal X-ray structures of 2c and 2d are reported. Crystallographic structural bond parameters of 2c and 2d revealed bond length equalization of C-C, C-O and M-O bonds. It has been shown that perchlorate () counter anion, present in the starting ruthenium complex, acts as the oxidizing agent in bringing about oxidation of Ph-NH2 to ortho-semidine. The chloronium ions, produced in situ, chlorinate the coordinated acac ligands at the γ-carbon atom. Such electrophilic substitution of coordinated acac ligands indicates that the Ru-acac metallacycles in the reference compounds are aromatic. The complexes showed an intense and featureless band centered near 520 nm, and a structured band near 275 nm. These displayed one reversible cathodic response in the range, −1.1 to −0.8 V and one reversible anodic response between 0.4 and 0.6 V versus the Saturated Calomel reference Electrode, SCE. The response at the anodic potential is due to oxidation of the coordinated ligand L, while the reversible response at cathodic potential is due to reduction of the metal center. 相似文献

The role of methanolic extract and n-butanol fraction of A. macrophylla leaves was investigated on the forward motility of goat spermatozoa. The methanol extract (600 micro/g/ml) and one n-butanol fraction (Fraction A; 100 microg/ml) showed marked inhibition of sperm forward motility, tested by microscopic and spectrophotometric methods. Approximately, 50-60% of the spermatozoa lost their motility when treated with 600 microg/ml of methanol extract or 100 microg/ml of Fraction A. The Fraction A at 400 microg/ml concentration showed complete inhibition of sperm forward motility at 0 min. The inhibitory activity increased with the increasing concentrations of the fraction. The motility inhibitory activity of the Fraction A was stable to heat treatment at 100 degrees C for 2 min. The compound showed high inhibitory effect in the pH range 6.7-7.6. Fraction A also showed high efficacy for inhibiting human sperm motility, assessed by the microscopic method. The phytochemical analysis of methanolic extract of A. macrophylla leaves revealed the presence of sterols, triterpene, flavonoid, alkaloid, tannin and reducing sugar, while the Fraction A contains beta-sitosterol, a common phytosterol. The results demonstrate that Fraction A (beta-sitosterol) is a potent inhibitor of sperm motility and thus it has the potential to serve as a vaginal contraceptive. 相似文献

The effect of loop length on quadruplex stability has been studied when the G-rich strand is present along with its complementary C-rich strand, thereby resulting in competition between quadruplex and duplex structures. Using model sequences with loop lengths varying from T to T5, we carried out extensive FRET to discover the influence of loop length on the quadruplex-Watson Crick duplex competition. The binding data show an increase in the binding affinity of quadruplexes towards their complementary strands upon increasing the loop length. Our kinetic data reveal that unfolding of the quadruplex in presence of a complementary strand involves a contribution from a predominant slow and a small population of fast opening conformer. The contribution from the fast opening conformer increases upon increasing the loop length leading to faster duplex formation. FCS data show an increase in the interconversion between the quadruplex conformers in presence of the complementary strand, which shifts the equilibrium towards the fast opening conformer with an increase in loop length. The relative free-energy difference (Delta DeltaG(o)) between the duplex and quadruplex indicates that an increase in loop length favors duplex formation and out competes the quadruplex. 相似文献

An assemblage of Phlycticeras Hyatt from a precisely dated zone of Middle Callovian in Kutch, India has been analyzed. Systematic study reveals that this sample can be divided into two size groups. The larger set shows different adult modifications leading to ornamental polymorphism. Polymorphs are very similar, if not identical, to different chronospecies of Europe, which range between Middle to Upper Callovian. They are grouped under well-known Phlycticeras polygonium (Zieten). The group of smaller specimens on the other hand, strongly resembles a species, which has been previously described as Phlycticeras schaumburgi (Waagen) from the younger Upper Callovian horizons in Kutch. It has a peculiar ‘rooster’-like septicarinate keel in the venter near adult aperture. Phlycticeras-Oecoptychius have been considered as a possible dimorphic pair since long, but here shown to have many inconsistencies to support these views. Instead, sexual dimorphism is explored within Phlycticeras and P. polygonium-P. schaumburgi is proposed as possible antidimorphs. A similar association is found in different stratigraphic assemblages of Europe. Interestingly these two morphs show parallel evolutionary changes within the Phlycticeras genus. 相似文献

Akt, a serine-threonine protein kinase, exists as three isoforms. The Akt signaling pathway controls multiple cellular functions in the cardiovascular system, and the atheroprotective endothelial cell-dependent role of Akt1 has been recently demonstrated. The role of Akt3 isoform in cardiovascular pathophysiology is not known. We explored the role of Akt3 in atherosclerosis using mice with a genetic ablation of the Akt3 gene. Using hyperlipidemic ApoE(-/-) mice, we demonstrated a macrophage-dependent, atheroprotective role for Akt3. In vitro experiments demonstrated differential subcellular localization of Akt1 and Akt3 in macrophages and showed that Akt3 specifically inhibits macrophage cholesteryl ester accumulation and foam cell formation, a critical early event in atherogenesis. Mechanistically, Akt3 suppresses foam cell formation by reducing lipoprotein uptake and promoting ACAT-1 degradation via the ubiquitin-proteasome pathway. These studies demonstrate the nonredundant atheroprotective role for Akt3 exerted via the previously unknown link between the Akt signaling pathway and cholesterol metabolism. 相似文献

Metals can have a number of detrimental or beneficial effects in the cell, but first they must get in. Organisms have evolved transport mechanisms to get metals that are required, or essential into the cell. Nonessential metals often enter the cell through use of the machinery provided for essential metals. Much work has been done to advance our understanding of how these metals are transported across plasma and organelle membranes. This review provides an overview of essential and nonessential metal transport and homeostatic processes. 相似文献

World health organization has called for academic research and development of new chemotherapeutic strategies to overcome the emerging resistance and side effects exhibited by the drugs currently used against leishmaniasis. Diospyrin, a bis-naphthoquinone isolated from Diospyros montana Roxb., and its semi-synthetic derivatives, were reported for inhibitory activity against protozoan parasites including Leishmania. Presently, we have investigated the antileishmanial effect of a di-epoxide derivative of diospyrin (D17), both in vitro and in vivo. Further, the safety profile of D17 was established by testing its toxicity against normal macrophage cells (IC50 ∼ 20.7 μM), and also against normal BALB/c mice in vivo. The compound showed enhanced activity (IC50 ∼ 7.2 μM) as compared to diospyrin (IC50 ∼ 12.6 μM) against Leishmania donovani promastigotes. Again, D17 was tested on L. donovani BHU1216 isolated from a sodium stibogluconate-unresponsive patient, and exhibited selective inhibition of the intracellular amastigotes (IC50 ∼ 0.18 μM). Also, treatment of infected BALB/c mice with D17 at 2 mg/kg/day reduced the hepatic parasite load by about 38%. Subsequently, computational docking studies were undertaken on selected enzymes of trypanothione metabolism, viz. trypanothione reductase (TryR) and ornithine decarboxylase (ODC), followed by the enzyme kinetics, where D17 demonstrated non-competitive inhibition of the L. donovani ODC, but could not inhibit TryR. 相似文献
