Regulation of collagenase and collagenase mRNA production in early- and late-passage human diploid fibroblasts 总被引:6,自引:0,他引:6
The levels of collagenase and collagenase mRNA produced by early-passage (less than 40% of lifespan completed) and late-passage (greater than 80% of lifespan completed) cultures of human fibroblasts were analyzed. The constitutive levels of collagenase and collagenase mRNA produced by the late-passage cultures were 10-30 x greater than the levels observed in similarly treated early-passage cultures. Immunofluorescence analysis established that the percentage of collagenase-positive cells was also greater (77% vs. 4%) in the late-passage cultures. To determine whether the difference in collagenase production resulted from cell-derived regulatory factors, collagenase production was examined in cultures plated onto substrates coated with fibroblast extracellular matrix (ECM). Collagenase and collagenase mRNA production was enhanced in both types of cultures, although amounts produced by ECM-induced early-passage cultures was significantly less than that produced by similarly treated late-passage cultures. A pour-plate method is described for the detection of coagulase production by Staphylococcus aureus. Either Brain Heart Infusion agar or yeast extract-Trypticase soy agar containing swine plasma was the best medium for the detection of coagulase by this method. The advantages of this method and its potential utilization in the clinical laboratory, in food microbiology, and in specialized studies with S. aureus are discussed. 相似文献

Summary Microsomal and soluble fractions of Pleurotus pulmonarius exhibited a reduced carbon monoxide difference spectrum with P450 maxima at 448nm and 450–452nm respectively. Substrate induced Type I spectra were observed on addition of benzo(a)pyrene to both fractions. Benzo(a)pyrene hydroxylation was measured using the aryl hydrocarbon hydroxylase assay and was observed to be P450 dependent as indicated by carbon monoxide inhibition together with the substrate binding characteristics. The activity of the fractions were observed to give Km of 200mM and 660mM and Vmax of 1.25 nmol/min/nmol P450 and 0.57 nmol/min/nmol P450 for the microsomal and cytosolic fractions respectively. 相似文献

Recent publications have suggested the existence of germ stem cells in the mouse at postnatal stages. The mechanism of de
novo oocyte formation is proposed to involve a contribution from the bone marrow to the germ cell pool, via the bloodstream.
Critical examination of the data underpinning these contentious claims is under way from a reproductive biology perspective
but little has been said about the nature of this elusive bone marrow population with germ cell potential. Furthermore, whereas
the prospect of marrow-derived germ cells may appear propitious for fertility applications, its wider impact on transplantation
medicine remains to be considered. This paper examines the evidence leading to the current debate and considers the implications
of such findings for the field of bone marrow transplantation.

A preliminary examination of the spermatodesms of Orthoptera Tettigonioidea revealed a structure that is similar in individuals of the same sex but very different in specimens of opposite sex. This reorganization would seem to take place inside the spermatophore during transit from the male to female genital tracts. The results of incubating spermatodesms with the secretions of glandular extract (GE) obtained from male accessory glands, known to be involved in forming the spermatophore wall, revealed changes in the spermatodesm 'cap' that are comparable to those occurring in vivo. Moreover, incubation of spermatodesms with the extracts obtained separately from tubules of the 1st and 2nd orders (GE1, GE2) established that GE2 alone modifies the spermatodesms, thus excluding the possible implication of 1st order tubules in the rearrangement process. In conclusion, data from incubations of spermatodesms with the single fractions obtained by submitting GE2 to gel-filtration FPLC show that only the peak 4 maintains intact the biological activity of GE2, SDS-PAGE analysis of the fraction corresponding to peak 4 revealed a greater protein content of 29 kD, which also appears to a lesser degree in fraction 3. This material is responsible for a partial dismantling of the 'cap' in the incubated spermatodesms. 相似文献

Human embryonic stem cells (hESCs) are thought to be susceptible to chromosomal rearrangements as a consequence of single cell dissociation. Compared in this study are two methods of dissociation that do not generate single cell suspensions (collagenase and EDTA) with an enzymatic procedure using trypsin combined with the calcium-specific chelator EGTA (TEG), that does generate a single cell suspension, over 10 passages. Cells passaged by single cell dissociation using TEG retained a normal karyotype. However, cells passaged using EDTA, without trypsin, acquired an isochromosome p7 in three replicates of one experiment. In all of the TEG, collagenase and EDTA-treated cultures, cells retained consistent telomere length and potentiality, demonstrating that single cell dissociation can be used to maintain karyotypically and phenotypically normal hESCs. However, competitive genomic hybridization revealed that subkaryotypic deletions and amplifications could accumulate over time, reinforcing that present culture regimes remain suboptimal. In all cultures the cell surface marker CD30, reportedly expressed on embryonal carcinoma but not karyoptically normal ESCs, was expressed on hESCs with both normal and abnormal karyotype, but was upregulated on the latter. 相似文献

Nero Siciliano (Sicilian Black, SB) is a local pig breed generally of uniform black color. In addition to this officially recognized breed, there are animals showing morphological characteristics resembling the SB but with gray hair (Sicilian Grey, SG). The SG, compared with the SB, also shows a more compact structure with greater transverse diameters, higher average daily gains and lower thickness of the back fat. In this study, using the Illumina PorcineSNP60 BeadChip, we run genome-wide analyses to identify regions that may explain the phenotypic differences between SB (n = 21) and SG (n = 27) individuals. Combining the results of the two case–control approaches (GWAS and FST), we identified two significant regions, one on SSC5 (95 401 083 bp) and one on SSC15 (55 051 435 bp), which contains several candidate genes related to growth traits in pig. The results of the Bayesian population differentiation approach identified a marker near the MGAT4C, a gene associated with average daily gain in pigs. Finally, scanning the genome for runs of homozygosity islands, we found that the two groups have different runs of homozygosity islands, with several candidate genes involved in coat color (in SG) or related to different pig performance traits (in SB). In summary, the two analyzed groups differed for several phenotypic traits, and genes involved in these traits (growth, meat traits and coat color) were detected.
