Liver plasma membranes contain a morphologically distinct protein complex which serves as a substrate for the plasma membrane-associated transglutaminase. The complex, which appears as a two-dimensional sheet, is insoluble in sodium dodecyl sulfate and reducing agents and has been named SITS for sodium dodecyl sulfate-insoluble transglutaminase substrate (Tyrrell, D. J., Sale, W. S., and Slife, C. W. (1988) J. Biol. Chem. 263, 1946-1951). Polyclonal antibodies raised against SITS were used to probe for soluble constituents of the matrix. Immunoblots showed that proteins of 230, 35, and 32 kDa reacted with the anti-SITS antiserum when the soluble fraction from a liver homogenate was examined. The 230-kDa protein was identified as fibronectin after observing cross-reactivity of anti-SITS antiserum with authentic fibronectin and cross-reactivity of anti-fibronectin antiserum with the 230-kDa cytosolic protein and purified SITS. Preincubating anti-SITS antiserum with purified fibronectin decreased immunostaining of the 230-kDa cytosolic protein and authentic fibronectin. Immunoblots of the plasma membrane fraction using anti-SITS and anti-fibronectin antisera showed that both antisera reacted with proteins at the top of the stacking gel (SITS) and of 230 kDa. In addition, the anti-SITS antiserum reacted with proteins of 85, 35, and 32 kDa. Immunofluorescence microscopy revealed that the anti-SITS and anti-fibronectin antisera both react with isolated SITS and with the same filamentous structures associated with intact plasma membranes. These studies show that fibronectin is a component of the plasma membrane matrix, SITS. This finding is consistent with the proposed role of this matrix which is to mediate cell-cell adhesion between hepatocytes in the tissue.  相似文献

Fractionation of rat liver by homogenization and differential centrifugation revealed that only about 83% of the transglutaminase activity in the tissue is in a soluble form, and that the remainder is associated with the particulate fraction. This latter activity remained with the membranes even after they were extensively washed to remove 99% of such soluble enzymes as lactate dehydrogenase and aldolase. Subsequent fractionation of the membranes by isopycnic density gradient centrifugation in sucrose resulted in a single band of transglutaminase activity at a density of 1.194 g/cm3. This activity was coincident with the major band of plasma membranes, which was identified by its content of 5'-nucleotidase, alkaline phosphodiesterase I, alkaline phosphatase and leucine aminopeptidase activities. After treatment with digitonin and fractionation on sucrose gradients, the transglutaminase activity and the plasma membrane marker enzyme activities were found at a new density of 1.210 g/cm3, while the enzyme markers for the other membrane fractions remained unchanged. From these data, we conclude that approximately 17% of the transglutaminase activity in rat liver is specifically associated with the plasma membranes.  相似文献

Long-chain (sphingoid) bases may serve as another category of "lipid second messenger" because they inhibit protein kinase C and affect multiple cellular functions. Free sphingosine has been found in rat liver (Merrill, A. H., Jr., Wang, E., Mullins, R. E., Jamison, W. C. L., Nimkar, S., and Liotta, D. C. (1988) Anal. Biochem. 171, 373-381); hence, this study determined if liver plasma membranes contain free long-chain bases and have the ability to form them from endogenous enzymes and substrates. Isolated plasma membranes contained 0.45 nmol of sphingosine/mg of protein which, based on the recovery of the membranes, was equivalent to 3.5 +/- 1.2 nmol/g of liver and at least half of the total free sphingosine in liver. When the membranes were incubated at 37 degrees C, the amount increased at an initial rate of 5-25 pmol/min/mg, resulting in a 2-3-fold increase over an hour. Sphingosine formation required divalent cations, was optimal at neutral to alkaline pH, and was temperature-dependent. Activities with these characteristics were not identified in microsomes or lysosomes (lysosomal activities with acidic pH optima were detected, however); hence, they appear to reflect a separate plasma membrane system. Sphingosine formation was stimulated by ceramides either added exogenously or formed endogenously by treating the membranes with sphingomyelinase (but not endoglycoceramidase). Sphingomyelin hydrolysis to ceramide was also observed during incubation of the plasma membranes alone. Some of the properties of this system resembled the neutral sphingomyelinase and ceramidase activities of liver. While the physiological significance of this endogenous sphingosine is not known, this system has the appropriate subcellular location to provide sphingosine as a participant in signal transduction.  相似文献

Various cytokinins and purines were ineffective in reversing glyphosate (0.25 millimolar)-induced growth inhibition of carrot (Daucus carota L.) cell suspension cultures. Aspartate was particularly effective in reversing glyphosate inhibition, but asparagine and various combinations of lysine, methionine, threonine, and homoserine (eventual products of aspartate metabolism) were not effective. When organic acids of the tricarboxylic acid cycle were added to the medium, particularly good reversal of inhibition could be obtained with α-ketoglutarate, succinate, and malate. Citrate provided only moderate reversal but the reversal given by glutamate was comparable to that of aspartate and the more effective tricarboxylic acid cycle intermediates. Pyruvate was somewhat toxic to cells when added early in the cell cycle but was most effective at reversing glyphosate inhibition when added at this time. If pyruvate addition was delayed, it was less toxic but was also a less effective reversing agent for glyphosate inhibition.  相似文献

Fumonisin is a recently identified mycotoxin that has been shown to be the cause of pulmonary edema disease in swine and leukoencephalomalacia in horses. Mystery Swine Disease (MSD), is an economically devastating disease complex of unknown etiology that has been reported to have occurred in several swine producing states since 1988. To determine the relationship between MSD and fumonisin, a case-control study was carried out in Illinois in mid-1990. Feed samples collected from 12 case and 9 control farms were analyzed for fumonisin. Sera from swine on all farms was screened for titers against encephalomyocarditis (EMC) virus and concentrations of alpha-1 acid glycoprotein (an acute phase reactive protein). Fumonisin concentrations 20 ppm were found on 1 control farm (1/9) and 8 case farms (8/12). Titers against EMC virus ( 116) were found on 5 control farms (5/9) and on 6 case farms (6/12). Farms with 20 ppm fumonisin in the feed were at significantly increased risk (OR= 11.2, Fisher's exact test p=0.037) for MSD. Furthermore, the 2 test for trend was significant (p= 0.017), meaning that as the level of fumonisin in the feed increased, the risk of MSD also increased. The presence of EMC virus titers in the sow herd was not a significant risk for MSD (OR =1.25, Fisher's exact test p=0.75). Alpha-1 acid glycoprotein concentrations obtained from 2-week old nursing pigs differed significantly (p=0.0005) between MSD case and control herds.  相似文献

Rat liver plasma membranes contain transglutaminase activity and a large molecular weight protein complex which serves as a substrate for this enzyme. When plasma membranes were solubilized in sodium dodecyl sulfate and disulfide-reducing agents the transglutaminase substrate was recovered in the detergent-insoluble fraction. The insolubility of the complex suggested that it might be further studied by adsorbing membranes onto glass slides, then extracting with the detergent and reducing agent. After extraction, dark field light microscopy revealed numerous flattened sheets which varied in size from 4 to 12 micrometers. To confirm that these structures were the large molecular weight transglutaminase substrate, the plasma membranes were solubilized in sodium dodecyl sulfate and dithiothreitol and sedimented through a sucrose gradient containing the agent. The large molecular weight substrate was the only material found at the 1.11/1.23 g/cm3 interface. Microscopic examination showed the same structures previously observed on the glass slides. We conclude that the large molecular weight transglutaminase substrate is a sodium dodecyl sulfate-insoluble, morphologically distinct, protein complex. Due to its considerable size, nondissociable nature, and association with the lateral membrane, the sodium dodecyl sulfate-insoluble transglutaminase substrate may serve as a type of skeleton or scaffolding for this plasma membrane domain.  相似文献

Rat liver plasma membranes contain transglutaminase activity and a large molecular weight protein aggregate that serves as a substrate for this enzyme (Slife, C.W., Dorsett, M.D., Bouquett, G.T., Register, A., Taylor, E., and Conroy, S. (1985) Arch. Biochem. Biophys. 241, 329-336; Slife, C.W., Dorsett, M.D., and Tillotson, M.L. (1986) J. Biol. Chem. 261, 3451-3456). When purified plasma membranes were sonicated and the different plasma membrane domains were separated by sedimentation through a linear sucrose gradient, virtually all of the transglutaminase activity and the large molecular weight transglutaminase substrate were associated with membrane fragments which migrated to a very dense region of the gradient (1.18 g/cm3). The bile canalicular markers, 5'-nucleotidase and HA-4 antigen, were predominantly found at 1.11 g/cm3, while most of the sinusoidal/lateral marker, CE-9 antigen, was detected at 1.14 g/cm3. Smooth membrane vesicles were observed chiefly at the lighter densities upon morphological analysis, while many filament-bearing, plasma membrane segments and junctional complexes were contained in the heavy transglutaminase fractions. These data show that the plasma membrane transglutaminase and the large molecular weight transglutaminase substrate are associated with a distinct region of the plasma membrane.  相似文献
