Treatment of rats with mirex (40 ppm in diet) caused hypoglycemia, liver enlargement, and inhibition of adrenal corticosteroid-synthesizing enzyme activity. At toxic dosages (20,000 ppm mirex in diet, which has a lethal toxicity-50 [LT-50] of ten days) poisoned female rats showed severe hypoglycemia, fatty liver, adrenal hyperplasia, hypophagia, lipid mobilization, and body weight (bw) loss. A 50 μg/kg intraperitoneal (IP) dose of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in male rats caused similar effects two days posttreatment. Hypoglycemia could be overcome by prednisone (which also inhibited adrenocorticoid-synthesizing enzyme activities) but not by streptozotocin treatment, indicating that hypoglycemia may be related to glucocorticoid deficiency resulting from inhibition of their synthesis and not by direct effects on pancreatic β-cells. Glucocorticoid deficiency could also cause increased release of adrenocorticoid hormone (ACTH), which may enhance fat mobilization caused by hypophagia.  相似文献

Estimates of sediment entrainment are required for models of particle transport in lakes and estuaries but are difficult to make because of the multiplicity of factors affecting cohesiveness of surficial sediments. We present results of sediment resuspension studies performed in an annular flume calibrated with laser-Doppler velocimetry. In our experiments, using sediments collected from two sites in the R. Raisin which flows into L. Erie and from one site in the western basin of L. Erie near the mouth of the R. Raisin, we applied shear stresses at the sediment-water interface in steps from 2 to 12 dyne/cm2. Percent water content at the surface of the sediments was either 77 or 74%, and trials were run with and without oxygenating the water overlying the sediments. Entrainment rates as a function of shear stress at the sediment-water interface were best described by a power-law relationship. All but 14% of the variability in the power law expression was due to shear stress and percent water content; the variability not accounted for was due to differences in particle size distributions, chemical properties, and biological activity in the sediments.  相似文献

Summary A synthetic gene encoding aprotinin (bovine pancreatic trypsin, inhibitor) was fused to theSaccharomyces cerevisiae prepro alpha mating factor leader sequence at the dibasic amino acid processing site.Pichia pastoris strains were developed to'express one or multiple copies of a methanol-inducible expression cassette containing the gene fusion.P. pastoris containing a single copy of the vector secreed approximately 150 mg/l of immunoreactive protein. A construct bearing five copies of the expression cassette secreted 930 mg/l of aprotinin. The purified aprotinin molecule was equipoten with the native molecule in a trypsin inhibition assay. Protein sequence analysis showed that the alpha factor-aprotinin fusion was not processed at the basic amino acid residues Lys-Arg. Instead, recombinant aprotinin had additional N-terminal amino acids derived from prepro alpha factor. The N-terminal extension was variably 11 or 4 amino acids. Inclusion of the spacer DNA sequence encoding Glu and Ala between aprotinin and the Lys-Arg processing site led to the secretion of a biologically active aprotinin containing only a Glu-Ala N-terminal extension.  相似文献

Aim Evidence indicates that species are responding to climate change through distributional range shifts that track suitable climatic conditions. We aim to elucidate the role of meso‐scale dispersal barriers in climate‐tracking responses. Location South coast of England (the English Channel). Methods Historical distributional data of four intertidal invertebrate species were logistically regressed against sea surface temperature (SST) to determine a climate envelope. This envelope was used to estimate the expected climate‐tracking response since 1990 along the coast, which was compared with observed range expansions. A hydrodynamic modelling approach was used to identify dispersal barriers and explore disparities between expected and observed climate tracking. Results Range shifts detected by field survey over the past 20 years were less than those predicted by the changes that have occurred in SST. Hydrodynamic model simulations indicated that physical barriers produced by complex tidal currents have variably restricted dispersal of pelagic larvae amongst the four species. Main conclusions We provide the first evidence that meso‐scale hydrodynamic barriers have limited climate‐induced range shifts and demonstrate that life history traits affect the ability of species to overcome such barriers. This suggests that current forecasts may be flawed, both by overestimating range shifts and by underestimating climatic tolerances of species. This has implications for our understanding of climate change impacts on global biodiversity.  相似文献

Given their trans-synaptic localization, their persistent expression at mature synapses and their distinct biochemical and adhesive properties, cadherins are uniquely poised at the synapse to mediate synaptic plasticity, the ability to change synaptic function thought to underlie learning and memory. For example recent work suggests that cadherins may recruit and stabilize AMPA receptors at the synapse via direct interactions or through complex formation, revealing cross talk between postsynaptic signaling and adhesion. Moreover, the use of small interfering RNA knockdown of cadherin, the availability of N-cadherin-deficient embryonic stem cells and the acute disruption of cadherin function with peptide application in vivo have allowed for more precise dissection of the molecular mechanisms by which cadherins function in both structural and functional plasticity.  相似文献
