Trehalose is absorbed by two distinct systems-one constitutive, the other induced by turanose and to a lesser extent by nigerose but not by trehalose. The constitutive system is apparently mediated by a surface trehalase; the induced system has the characteristics of a permease. The specificity of the induced system is apparently limited to the alpha glucosyl-glucose or glucosyl-fructose linkage, because absorption of kojibiose, nigerose, maltose, isomaltose, turanose, sucrose, and melezitose, in addition to that of trehalose, was increased. Absorption of beta-linked or of galactose-containing disaccharides was not increased. The constitutive and induced trehalose-absorbing systems differ in their activity, specificity, lability to acid treatment, effects of substrate concentration, and pH optima. Both systems require oxygen, and no marked differential effects of inhibitors were observed. The activity of the induced system is proportional to log turanose concentration (from about 1 to 300 mug/ml), and is an approximate linear function of time of exposure (from about 1 to 50 min). Accumulation of trehalose occurred against a concentration gradient in both systems but particularly in the induced. No leakage was observed. The activity of the induced system declined slowly upon removal of the inducer. Accumulated trehalose is metabolized after activation by azide as are the endogenous trehalose reserves. The accumulated trehalose appears to enter the endogenous trehalose pool found in these spores, although some data suggest it may be more accessible. Respiratory data indicate that absorbed trehalose is available for metabolism while in transit from the external membrane to the internal pool.  相似文献

Urban expansion is a major cause of land use change and presents a significant threat to biodiversity worldwide. Agricultural land is often acquired by local councils and developers to expand urban growth boundaries and establish new housing estates. However, many agricultural landscapes support high biodiversity values, especially farmlands that feature mosaics of native vegetation and keystone habitat such as hollow‐bearing trees. In south‐eastern Australia, many arboreal marsupials including the threatened Squirrel Glider (Petaurus norfolcensis) have populations within peri‐urban zones of expanding rural cities. A key challenge to planners, developers and conservation organisations is the need to maintain habitat for locally rare and threatened species as land undergoes changes in management. Critical to the sustainable development of peri‐urban landscapes is a thorough understanding of the distribution, habitat requirements and resources available to maintain and improve habitat for species dependent on limited resources such as tree cavities. In this management report, we present background information on an integrated research programme designed to evaluate potential impacts of urban development on fauna in the Albury Local Government Area, NSW. We mapped hollow‐bearing trees, erected nest boxes and monitored arboreal marsupials. Information presented in this report provides a blueprint for monitoring arboreal marsupials, including threatened species in other developing regions, and will assist the Albury‐Wodonga local governments in future planning of sustainable living environments.  相似文献

This article critically examines the generation of discourses on Greek identity following an episode that took place in northern Greece (2000/2003) when an Albanian student was elected flag-carrier in a commemorative parade. Three versions of Greek identity emerged in this context: the first was based on civic understandings of identity, promoting the current Europeanist project of citizenship as belonging. The second version drew upon the notion of “culture” as an all-encompassing concept to promote ideas of Greek cultural “purity” that have roots in Greek ethnogenesis. The third version adopted an understanding of the “nation” in terms of racial affiliation, transforming it thus into a natural category. The argument put forward is that in the context of the 2000/2003 episode (a) Greek self-perceptions are affected by the problematic economic and cultural position of Greece within Europe and (b) Greek discourses of identity are a form of resistance to processes of Europeanization that threaten traditional “imagined communities” embedded in history.  相似文献

Epoxyeicosatrienoic acids (EETs) confer vasoactive and cardioprotective functions. Genetic analysis of the contributions of these short-lived mediators to pathophysiology has been confounded to date by the allelic expansion in rodents of the portion of the genome syntenic to human CYP2J2, a gene encoding one of the principle cytochrome P450 epoxygenases responsible for the formation of EETs in humans. Mice have eight potentially functional genes that could direct the synthesis of epoxygenases with properties similar to those of CYP2J2. As an initial step towards understanding the role of the murine Cyp2j locus, we have created mice bearing a 626-kb deletion spanning the entire region syntenic to CYP2J2, using a combination of homologous and site-directed recombination strategies. A mouse strain in which the locus deletion was complemented by transgenic delivery of BAC sequences encoding human CYP2J2 was also created. Systemic and pulmonary hemodynamic measurements did not differ in wild-type, null, and complemented mice at baseline. However, hypoxic pulmonary vasoconstriction (HPV) during left mainstem bronchus occlusion was impaired and associated with reduced systemic oxygenation in null mice, but not in null mice bearing the human transgene. Administration of an epoxygenase inhibitor to wild-type mice also impaired HPV. These findings demonstrate that Cyp2j gene products regulate the pulmonary vascular response to hypoxia.  相似文献
