Over the past year, several significant developments have been made in the field of anion binding. The fundamental principles of molecular recognition are increasingly being better understood, and of particular interest are reports of several synthetic anion receptors able to perform their task in complex natural media. Cloned, protein antigen-specific, Ia-restricted T cell lines frequently (approximately 20%) also respond strongly to stimulator cells from strains expressing stimulatory alleles at the chromosome 1-encoded Mls-locus. Furthermore, such responses are blocked by monoclonal antibodies specific for Ia antigens expressed by the stimulator rather than the responder cells. However, such responses show no specificity for polymorphic determinants on Ia molecules, although in such responses, as in primary and secondary T cell responses to stimulating Mls-locus alleles, I-E molecules appear to play a central role. These results, combined with the unique immunobiology of the primary T cell proliferative response to Mls-locus-disparate stimulator cells, suggest to us that this response involves the interaction of the receptor on T cells for antigen:self Ia with a relatively nonpolymorphic region of Ia glycoproteins. This hypothesis is supported by the observation that a monoclonal antibody to the T cell receptor will inhibit both responses, although the response to Mls-locus-disparate stimulators appears to be more sensitive to these antibodies. We propose that the interaction of the T cell receptor with Ia is stabilized by a cell interaction molecule encoded or regulated by the Mls-locus gene product permitting the T cell receptor:Ia glycoprotein interaction to lead to T cell activation. 相似文献

Concentration factor and biological half-life of 54Mn were determined in three species representing an ecologically and economically important food chain. Green algae (Chlorella spp.), Daphnia magna and yellow perch (Perca flavescens) were exposed to 54Mn in water and assayed for 54Mn uptake. Steady state concentration factors computed from the laboratory data for algae, Daphnia and perch were 4230, 17 000 and 11, respectively. The ultrastructure of the calcareous skeleton is described in twenty–one species of recent tubuliporine cyclostome bryozoans, using field emission SEM. The succession of skeletal fabrics in interior walls may be classified into four different fabric suites. The first–formed part of the calcitic skeleton in all species for which it has been observed is a precursory fabric of tiny, wedge–shaped crystallites. This is succeeded in about half of the species studied by a fabric of transverse fibres, followed by foliated fabric and often semi–nacre (fabric suite 1). Most of the remaining species lack transverse fibres and have interior walls largely comprising semi–nacre (fabric suite 2). A few species have skeletons consisting of predominantly distally–oriented, irregularly or regularly foliated fabric (fabric suite 3). A single species has a skeleton of proximally–oriented foliated fabric (fabric suite 4). Basal exterior walls in all species have a precursory fabric of tiny wedge–shaped crystallites without a strong preferred orientation, deposited directly upon the organic cuticle, followed by a layer of planar spherulitic structure, which in turn is succeeded by a similar fabric to that developed in the interior wall of the species concerned. Outermost layers of frontal exterior walls exhibit one of the following combinations of three fabrics: an outer layer of (1) finely granular or wedge–shaped crystallites; a thin dense granular layer followed by (2) distally accreting planar spherulitic fabric., or (3) obliquely accreting planar spherulitic fabric growing partly towards the midline of the frontal wall. Terminal diaphragms usually have outer layers dominated by planar spherulitic ultrastructure with centripetal growth directions. The fabric suites present in tubuliporines encompass most known fabrics found in the other cyclostome suborders and support the notion that this species–rich suborder occupies a central position in cyclostome evolution. 相似文献

3H-Lysergic acid diethylamide (3H-LSD) is irreversibly incorporated into bovine caudate membranes during ultraviolet light illumination. The incorporated radioligand apparently forms a covalent bond with a sub-population of the membrane proteins. Although the photolabeling pattern differs significantly from the Coomassie blue staining pattern on SDS gels, the photolabeling is apparently not specific for LSD binding sites associated with neurotransmitter receptors. 3H-LSD photolabeling can occur during prolonged exposure of membrane samples to room lighting and thus may introduce artifacts into receptor binding assays. 相似文献

Abstract Permeabilized cells of Haemophilus influenzae incorporate wall precursors into murein material in an ampicillin-sensitive reaction. In resistant transformants that contain the low antibiotic affinity penicillin-binding proteins (PBPs) 4 and 5, the sensitivity of this incorporation reaction to ampicillin is proportionally lower, suggesting a catalytic role for these proteins in wall synthesis. We conclude that, analogous to the reaction in Escherichia coli , PBPs 4 and 5 of H. influenzae have transpeptidase activity. 相似文献
