In vitro enzyme activities of glycolysis, pentose-phosphate pathway and dark CO2 fixation were assayed in batch cultures of heterotrophic Catharanthus roseus cells under various gassing rates and partial pressures of carbon dioxide. Detrimental effects of low pCO2 culture conditions on the growth characteristics could be linked to marked changes in levels of enzymes of primary metabolism during growth. The enzyme levels observed during the early stages of growth were found to be more stable when a constant pCO2 (20 mbar) was maintained and enabled exponential growth to be reached more rapidly.The importance of carbon dioxide as a conditioning factor of the culture medium is discussed.  相似文献

Summary A search was carried out for mutants, defective in the conversion of stearic acid to oleic acid in effort to improve the quality of lipids produced by Cryptococcus curvatus ATCC, 20509. Mutants were selected as unsaturated fatty acid (Ufa) auxotrophs. After treatment of parent organism with Ethyl methanesulfonate (EMS), 11 oleate-requiring auxotrophs were isolated. Only 3 of them were real unsaturated fatty acid (Ufa) mutants, while the other 8 were designated as fatty acid synthetase (Fas) mutants. The amount of saturated fatty acid (SFA) was about 65.2 % in the lipids extracted from an Ufa mutant named UfaM3 and it was significantly higher than that of the wild-type (WT) (46.6 %) and similar to that of cocoa butter (60.4 %).  相似文献

Summary The effect of culture conditions, especially nitrogen/carbon (N/C) ratio and temperature, on acyl lipid profiles in the oleaginous yeast Rhodotorula glutinis NRRL Y 1091 have been investigated. Cultivation of the microorganism under N-limited conditions (N/C < 0.14 g/g) resulted in enhanced fatty acid (FA) cell content but a reduced relative amount of polyunsaturated fatty acids (PUFA). The maximal FA productivity was obtained for a 0.025 N/C ratio resulting from the arrangement between the specific rate of FA synthesis and the concentration of lipid-free biomass. Under nitrogen-non-limiting conditions, cells grown at lower temperatures had a higher PUFA content and the maximal productivity of -linolenic acid was obtained by shifting the temperature of the culture from 30° C to 25° C. Azospirillum lipoferum crt1 was grown in batch cultures under standard conditions at 85% saturation of dissolved oxygen (DO) and 30-g/liter glucose concentrations. Kinetic studies revealed nutritional limitations of growth and the presence of an initial lag phase prior to consumption of glucose. The influences of various gaseous environments and shear stress on growth, i.e., various conditions of agitation-aeration, were characterized. Faster growth in the first stages of the culture and shorter duration of the lag phase were observed at DO concentrations of <30% saturation. The possible influences of dissolved CO(2) concentration or shear stress or both were discounted, and we confirmed the detrimental effect of high DO levels (up to 80% saturation) and the favorable influence of low DO concentrations (lower than 30% saturation) on growth. It was concluded that the gaseous environment, i.e., the DO concentration, needs to be considered as an operating parameter and be accurately controlled to ensure optimal growth of Azospirillum cells.  相似文献

Summary The manner in which copolymer poly(HB-co-HV) production was influenced by different methods of limiting cell proliferation during the production phase was examined. Polymer production was significantly improved in fermentation strategies in which some growth was maintained, either by linear or exponential nitrogen source feeding as compared to cultures in which nitrogen supply was totally interrupted. Improved volumetric productivities were obtained in cultures fed with NH4OH and the proportion of 3HV incorporated was approximately twofold higher in these cultures. These performance improvements were due to higher specific rates of glucose and propionate consumption in cultures in which true growth capacity was maintained.  相似文献

The limitation of growth of Catharanthus roseus cells was investigated with a view to their entrapment in a Ca-alginate matrix. An examination of the effects of lowered 2,4-D and phosphate concentrations on cell viability and indole alkaloid biosynthesis enabled a growth limiting and product formation stimulating medium to be designed. Entrapped cells showed a retention of both respiratory activity and biosynthetic capacity over an extended period of time compared with free cells. Summary Aerobic batch cultures of Apple fruit cells were carried out using sucrose as the sole carbon source. The determination of the growth parameters was used in order to estimate the maintenance coefficient m and the maximum growth yield YG; their values at 25°C are respectively 0.32. 10-2 g sugar/g cell dry weight.h and 0.57 g cell dry weight/g sugar. Then, a mathematical expression of the cell growth is discussed, with respect to the specific growth rate evolution during cultivation.  相似文献

Survival of alginate-entrapped cells of Azospirillum lipoferum was studied during dehydration using a dry air stream and during prolonged storage at various constant water activity values (aw). During the drying operation, the viability loss remained almost constant from the initial water content to 0.35 g water/g dry weight (DW) corresponding to a 98.5% water removal, strongly increased until a water content of 0.25 g/g DW and then stopped until the end of the drying operational (final aw 0.18). A water content of 0.25 g/g DW (aw=0.55) corresponded to the critical point of the moisture sorption isotherm curve from which water became restricted to the dry material. A high drying rate (5 g/g DW per hour) was shown to be more detrimental for cell viability than a low drying rate (1.18 g/g DW per hour). When the product was stored in a closed chamber with a regulated aw (0.23), the number of living cells decreased during a short period (less than 15 days) corresponding to the product aw stabilization, and then remained constant for more than 150 days. In addition, cell survival during storage was not affected by aw values in the range 0–0.55. Above aw=0.55, the higher the aw and the storage duration, the lower the residual survival percentage. The influence of the drying and storage conditions on the cell death rate are discussed with regard to both the mechanisms generally involved in viability loss and the hydration properties of water. Summary Candida lipolytica is shown to produce an extracellular polymer with emulsifying properties when grown on n-tetradecane or a mixture of linear hydrocarbons. A device for biosurfactant isolation is presented. The polymers recovered from the fermentation broth were found to be complex molecules with a protein, a lipid and a carbohydrate moiety. Their surface active properties suggest a possible role in hydrocarbon uptake by cells. Effectively, the addition of crude polymer resulted in an enhancement of respiration rate which was dependent on n-alkane concentration with glucose grown cells. Likewise in batch culture, maximum growth rate, cell productivity and yield were increased by the presence of the biosurfactant.  相似文献
