Postnatal development of sympathetic and sensory innervation of the rhesus monkey ovary. 总被引:1,自引:0,他引:1
We have used immunofluorescence to study the postnatal development of the sympathetic and sensory innervation to the rhesus monkey (Macaca mulatta) ovary. Sympathetic nerves were identified as adrenergic by their content of tyrosine hydroxylase (TH)-like immunoreactivity and as peptidergic by the presence of neuropeptide Y (NPY). Fibers containing substance P (SP) or calcitonin gene-related peptide (CGRP)-like immunoreactivity were considered as sensory, whereas vasoactive intestinal peptide (VIP)-positive fibers were only defined as peptidergic because VIP may be present in both sympathetic and sensory nerves. Ovaries from neonatal (2-mo-old), juvenile (9-18-mo-old), peripubertal (3-3.5-yr-old), adult (9-14-yr-old), and senescent (20-27-yr-old) monkeys were studied. At all ages, with the exception of senescence, TH-, NPY-, and VIP-containing fibers were associated with follicles in different developmental stages. In peripubertal and adult animals, some primordial follicles were found to be selectively innervated by VIPergic fibers that almost completely encircled each follicle. Both sympathetic and VIP fibers were also detected in the interstitial tissue and associated with the ovarian vasculature at all ages. The number of sympathetic and VIP fibers increased significantly (p < 0.01) between 2 mo and 9-18 mo of age, and again increased (p < 0.01) around the age of puberty (approximately 3 yr of age). After this time, the number of NPY and TH fibers remained constant. Post-fire sprouting of dormant buds in resprouter plants is facilitated by stored carbohydrate reserves, with starch being the critical reserve. Starch is mainly stored in xylem parenchyma ray tissue of woody underground organs, such as burls, lignotubers, and roots. We carried out a comparative analysis of the pattern of starch storage and the proportion of parenchymatic ray tissue in the upper root or cotyledonary region of seedlings from seeder and resprouter forms within two Cape Erica (Ericaceae) species: E. coccinea L. and E. calycina L., which were raised in the greenhouse under controlled irrigation. We also explored the root-to-shoot allocation patterns of seeder and resprouter seedlings in these two species. Resprouter seedlings of both species showed higher relative amounts of upper-root starch and upper-root storage tissue as well as a higher root-to-shoot allocation than their seeder counterparts. Pronounced swelling of the upper root region suggests ontogenetic development of a lignotuber in the resprouter forms of the two Erica species. The distinct allocation of starch in roots seems to be genetically determined and would account for the apparent differences in the root-to-shoot allocation patterns between both regeneration forms from the early seedling stage. 相似文献

In this study, we used a two-dimensional (2D) mechanistic mathematical model in order to evaluate the relative contribution of different microbial reactions to organic matter removal (in terms of COD) in horizontal subsurface-flow constructed wetlands that treated urban wastewater. We also used the model to analyse the effect of increasing or decreasing the organic loading rate (changing the hydraulic loading rate (HLR) at a constant influent organic matter concentration, or changing the organic matter concentration at a constant HLR) on both the removal efficiency and the relative importance of the microbial reactions. The model is based on the code RetrasoCodeBright, which we modified to include the main microbial processes related to organic matter and nitrogen transformations in the wetlands: hydrolysis, aerobic respiration, nitrification, denitrification, sulphate reduction and methanogenesis. The model was calibrated and validated with data from two wetlands (each with a surface area of 55 m2) located in a pilot plant near Barcelona (Spain). According to the simulations, anaerobic processes (methanogenesis and sulphate reduction) are more widespread in the wetlands and contribute to a higher COD removal rate (60–70%) than anoxic (denitrification) and aerobic reactions do. These model results are confirmed by experimental observations. In all the cases tested, the reaction that most contributed to COD removal was methanogenesis (33–52%). According to our simulations, decreasing the HLR (for example, from 40 to 25 mm/d) while maintaining a constant COD influent concentration has a clear positive impact on COD removal efficiency (which increases from 65% to 89%). Changing influent COD concentration (for example, from 290 to 190 mg/L) while maintaining a constant HLR has a smaller impact, causing efficiency to increase from 79% to 84%. Changes in influent COD concentration (at a constant HLR) affect the relative contribution of the microbial reactions to organic matter removal. IL‐15 is a proinflammatory cytokine that acts early in the inflammatory response and has been associated with several autoimmune diseases including rheumatoid arthritis, where it had been proposed as a therapeutic target. We recently reported an IL‐15 antagonist peptide corresponding to sequence 36–45 of IL‐15 (KVTAMKCFLL) named P8, which specifically binds to IL‐15Rα and inhibits IL‐15 biological activity with a half maximal inhibitory concentration (IC50) of 130 µ m in CTLL‐2 proliferation assay. In order to improve binding of peptide P8 to the receptor IL‐15Rα, we used an Ala scan strategy to study contribution of each individual amino acid to the peptide's antagonist effect. Here, we found that Phe and Cys are important for peptide binding to IL‐15Rα. We also investigated other single site mutations and replaced the second Lys in the sequence by the polar non‐charged amino acid threonine. The resulting peptide [K6T]P8 exhibited a higher activity than P8 with an IC50 of 24 µm . We also found that this peptide was more active than peptide P8 in the inhibition of TNFα secretion by synovial cells from rheumatoid arthritis patients. The peptide [K6T]P8 described in this work is a new type of IL‐15 antagonist and constitutes a potential therapeutic agent for rheumatoid arthritis. In rodents, the formation of ovarian follicles occurs after birth. In recent years, several factors required for follicular assembly and the growth of the newly formed follicles have been identified. We now describe a novel gene, Fxna, identified by differential display in the neonatal rat ovary. Fxna encodes an mRNA of 5.4 kb, and a protein of 898 amino acids. Fxna is a transmembrane metallopeptidase from family M28, localized to the endoplasmic reticulum. In the ovary, Fxna mRNA is expressed in granulosa cells; its abundance is maximal 48 hours after birth, i.e. during the initiation of follicular assembly. Reducing Fxna mRNA levels via lentiviral-mediated delivery of short hairpin RNAs to neonatal ovaries resulted in substantial loss of primordial, primary and secondary follicles, and structural disorganization of the ovary, with many abnormal follicles containing more than one oocyte and clusters of somatic cells not associated with any oocytes. These abnormalities were not attributable to either increased apoptosis or decreased proliferation of granulosa cells. The results indicate that Fxna is required for the organization of somatic cells and oocytes into discrete follicular structures. As an endoplasmic reticulum-bound peptidase, Fxna may facilitate follicular organization by processing precursor proteins required for intraovarian cell-to-cell communication. 相似文献

Microbial mats from two ponds with different salinities from the saltern of Guerrero Negro (Mexico) points toward millimeter‐scale coherent variations in trace metal (Me) concentrations (Cd, Co, Cu, Fe, Mn, Ni, Pb, Zn). Total, HCl‐leachable and pyrite‐associated Me showed a trend of increasing concentrations with increasing depth suggesting gradual addition of reactive Me probably as a result of metal sulfide precipitation at depth. The trends in Me profiles can be ascribed to the establishment and maintenance of microzones that promote geochemical processes, bacterial population distributions, and differential mass transport within the mats. Degrees of trace metal pyritization (1 ± 1% for Zn to 24 ± 7% for Cd) as well as metals associated with the pyrite fraction (<1.4–36 ± 18 nmol g?1 for Zn and Mn, respectively) were low, as expected from a reactive Fe‐limited system like Guerrero Negro. Calculated enrichment factors showed that Ni (2.6 ± 2.1), Co (5.5 ± 4.0), Pb (9.4 ± 7.4), and Cd (57 ± 39) were, on average, enriched in the microbial mats of Guerrero Negro. Natural enrichments of Cd, Pb, and Co in sediments along the coast of Baja California and metabolical requirements of Co and Ni by the predominant cyanobacteria in the Guerrero Negro mats may explain these enrichments. Metal characteristics in microbial mats could be advantageously used as biosignatures to identify their presence in the geological record or in other planetary systems. 相似文献

Plant diversity patterns in the Aljibe Mountains (S. Spain): a comprehensive account 总被引:6,自引:0,他引:6
The Aljibe Mountains are located in the southern tip of the Iberian Peninsula and have a remarkable biogeographical interest. The complete plant species list (trees, climbers, shrubs, perennial and annual herbs, ferns, lichens, bryophytes and macroscopic algae) was recorded in four 0.1 ha plots from each of the most representative community types (Quercus suber woodland, Q. canariensis forest, open heathland and Q. coccifera shrubland). Up to 119 plant species were found in total in the Q. suber woodland plot. The diversity of woody plants was analysed from 44 samples of cover (100 m line), and the herbaceous layer was explored in 200 quadrats (of 0.5 × 0.5 m). Three biodiversity components (species richness, endemism, and taxonomic singularity) were evaluated in both shrub and herbaceous layers. Open heathlands showed the highest richness of endemic species, both woody and herbaceous. The highest number of woody species was found in the evergreen Q. suber woodland, and of herbaceous species in the semi-deciduous Q. canariensis woodland. Taxonomic singularity was higher in Q. canariensis woodlands and Q. coccifera shrublands for woody species, but there were no significant differences in the herbaceous layer. Local species diversity of heathlands in this region resembles that of South African heathlands (fynbos), despite the obvious geographic and floristic distance, and contrasts with the low diversity of biogeographically closer, European temperate heathlands. The Aljibe Mountains show high diversity values for different life forms (from trees to mosses) and spatial scales (from community to region), and are rich in endemic species. Thus, this area should be recognised as a relevant unit within the Mediterranean plant diversity hot spots. 相似文献

A brief literature review shows that ionizing radiation in biological membranes and in pure lipid membranes causes malondialdehyde formation, indicating lipid peroxidation processes. With respect to membrane fluidization by ionizing radiation, in pure lipid membranes rigidization effects are always reported, whereas contradictory results exist for biological membranes. Starting from the assumption that membrane proteins at least partly compensate for radiation effects leading to a rigidization of membrane lipid regions, pig liver microsomes, as a representative protein-rich intracellular membrane system, were irradiated with X-rays or UV-C with doses up to 120 Gy at a dose rate of 0.67 Gy min–1 and up to 0.73 J cm–2 at an exposure rate of 16.2 mJ cm–2 min–1, respectively. For both irradiation types a weak but significant positive correlation between malondialdehyde formation and membrane fluidity is revealed throughout the applied dose ranges. We conclude that the membraneous protein lipid interface increases its fluidity under radiation conditions. Also, thymocyte ghosts showed an increased fluidity after X-ray irradiation. The resistance to experimental, highly frequent disturbance has been analysed in three congeneric, strong-resprouter species
(Erica australis, E. scoparia and E. arborea) that co-occur in heath-dominated communities of the northern side of the Strait of Gibraltar, southern Spain. To do so,
mature individuals of the three species from a long undisturbed location were clipped at the ground level every sixth month
during two years. The relationship between the resprouted biomass dry weight (as indicative of the resprouting vigour) and
the upper surface area of the lignotuber along the experiment was established separately for each species at each clipping
event by means of linear regressions analysis. The resprouting vigour of the three species was compared by means of independent
one-way ANOVAs within each clipping event. Resprouting vigour decreased after recurrent clippings in the three species. Nevertheless,
significant differences between species in this loss of resprouting vigour were detected, being E. scoparia the most resistant to the experimental, highly frequent clipping. It is concluded that experimental levels of recurrent disturbance
may help to find out differences in resilience within similar (taxonomically, morfologically and/or ecologically), strong-resprouter
plant species. Considering the history of forestry management in the nothern side of the Strait of Gibraltar, differences
in this regard between the three Erica species may contribute to explain their somewhat segregated ecological distribution in this region. 相似文献

Environmental heterogeneity shapes the traits of natural populations, including space use and burrowing behavior. We studied the red viscacha rat (Tympanoctomys barrerae), a semifossorial rodent with patchy distribution in drylands of west-central Argentina, to provide ecological data on habitat and burrow features of this specialist species. We collected data on 165 burrows and 55 individuals from 2017–2018 and observed correlations between different vegetation types, vegetation diversity, and physical attributes (e.g., soil hardness). A high number of individual red viscacha rats was negatively associated with herbs. The number of burrow entrances was positively associated with harder soils and negatively associated with vegetation cover. Our results suggest that the species' patchy distribution is strongly associated with environmental heterogeneity of these arid environments, and consequently provide an essential step towards conserving their populations across their geographic range. 相似文献
