Improved and efficient methods were developed for isolating high quality DNA and RNA from different sources of Iranian Yew
(Taxus baccata L.). The methods were based on CTAB extraction buffer added with high levels of polyvinylpyrrolidone (PVP) and β-mercaptoethanol to properly remove polysaccharides and prevent oxidation of phenolics. The pellets obtained by ethanol precipitation
were washed only with Chloroform: isoamyl alcohol (24:1). So, we could successfully eliminate the dangerous phenol/chloroform
extraction steps from the isolation procedure. Both spectrophotometric (A260/A280 and A260/A230 ratios) and agarose electrophoresis analysis of isolated nucleic acids (DNA and RNA) indicated good results. DNA with the
average yield of 100–300 μg/g leaf and stem tissue and total RNA with an average yield of 20–30 μg/g cell culture and 80–100 μg/g
leaf and stem tissue of Iranian yew could be obtained. Successful amplification of pam and pds by PCR and RT-PCR, showed the integrity of isolated DNA and RNA, respectively. 相似文献

Male Pattern Baldness (MPB) or androgenetic alopecia is a common form of hair loss with androgens and genetics having etiological
significance. Androgens are thought to pathophysiologically power on cascades of chronically dramatic alterations in genetically
susceptible scalp dermal papillas, specialized cells in hair follicles in which androgens react, and finally resulting in
a patterned alopecia. However, the exact mechanisms through which androgens, positive regulators of growth and anabolism in
most body sites, paradoxically exert their effects on balding hair follicles, are not yet known. The role of microRNAs, a
recently discovered class of non-coding RNAs, with a wide range of regulatory functions, has been documented in hair follicle
formation and their deregulation in cancer of prostate, a target organ of androgens has also been delineated. Yet, there is a lack of knowledge in agreement with microRNAs’ contribution in pathophysiology of MPB. To investigate the
role of microRNAs in pathogenesis of MPB, we selected seven microRNAs, predicted bioinformatically on a reverse engineering
basis, from previously published microarray gene expression data and analyzed their expression in balding relative to non-balding
dermal papillas. We found for the first time upregulation of four microRNAs (miR-221, miR-125b, miR-106b and miR-410) that
could participate in pathogenesis of MPB. Regarding microRNAs’ therapeutic potential and accessibility of hair follicles for
gene therapy, these microRNAs can be considered as good candidates for a new revolutionized generation of treatments. 相似文献

Summary The bioconversion of hydrocortisone by a locally isolated strain of cyanobacterium Fischerella ambigua PTCC 1635 was investigated. Fischerella ambigua had not been previously examined for this potential. The fermentation led to production of 11β,17α, 20β, 21-tetrahydroxypregn-4-en-3-one and 11β-hydroxyandrost-4-en-3,17-dione. The metabolites were isolated and purified by chromatographic methods and identified using

Knowledge of pistachio genetic diversity is necessary for the formulation of appropriate management strategies for the conservation
of these species. We analysed amplified fragment length polymorphisms in a total of 216 pistachio accessions, which included
seven populations from three wild species (Pistacia vera, Pistacia khinjuk and Pistacia atlantica subsp. kurdica) and most of the important cultivars from Iran, together with some foreign cultivars. High levels of genetic diversity were
detected within the Iranian cultivars, and they showed a clear separation from foreign cultivars, as revealed by unweighted
pair group method with arithmetic averaging and supported by analysis of molecular variance. The lowest amount of polymorphism
was observed in P. atlantica subsp. kurdica, which showed the lowest number of total bands as compared to the other species. This revealed strong genetic erosion of
P. atlantica subsp. kurdica, which reflected a severe decline in habitat and over-exploitation. Based on these findings, strategies are proposed for
the genetic conservation and management of pistachio species and cultivars. 相似文献

Summary One hundred and sixty-five strains of microorganisms with the ability to grow in a medium containing uric acid as a major
source of nitrogen were isolated from soil samples during a screening program. Among them, a zygomycete fungus with well-developed
columellae was recognized to produce high levels of the enzyme in a short time. Classification of the isolated fungus was
carried out according to the morphological and culture characteristics of the organism, and it was identified as Mucor hiemalis. The fungus was able to produce an intracellular urate oxidase in a fermentation medium mainly containing uric acid. Optimized
composition of the medium consisted of (l−1 of distilled water) uric acid, 7.0 g; maltose, 6.0 g; Vogel stock solution, 20 and 1 ml of 0.5 M copper sulphate. The optimum
pH and temperature for uricase production in the optimized medium were pH 6 and 30 °C, respectively. 相似文献

Lead tri‐iodide methylammonium (MAPbI3) perovskite polycrystalline materials show complex optoelectronic behavior, largely because their 3D semiconducting inorganic framework is strongly perturbed by the organic cations and ubiquitous structural or chemical inhomogeneities. Here, a newly developed time‐dependent density functional theory‐based theoretical formalism is taken advantage of. It treats electron–hole and electron–nuclei interactions on the same footing to assess the many‐body excited states of MAPbI3 perovskites in their pristine state and in the presence of point chemical defects. It is shown that lead and iodine vacancies yield deep trap states that can be healed by dynamic effects, namely rotation of the methylammonium cations in response to point charges, or through slight changes in chemical composition, namely by introducing a tiny amount of chlorine dopants in the defective MAPbI3. The theoretical results are supported by photoluminescence experiments on MAPbI3?mClm and pave the way toward the design of defect‐free perovskite materials with optoelectronic performance approaching the theoretical limits. 相似文献
