In a field study, heart rate and motor activity were assessed continuously in 12 male smokers during 2 smoking and 2 abstinence days and in 12 male nonsmokers during 4 days. A circadian analysis revealed earlier activity acrophases in smokers than nonsmokers and earlier heart rate acrophases in abstinent than smoking smokers. Furthermore, heart rate acrophases of smoking smokers significantly anticipated activity acrophases;, whereas in abstinent smokers and nonsmokers the two parameters oscillated in phase. With the use, in smoking smokers, of the individual average smoking interval as a hypothetical ultradian period length, significant periodicities were found for heart rates in 16 and for activity in 15 of 24 observation days. These rhythms were nicotine independent and based on heart rate and activity increases prior to lighting up the cigarettes. Individual frequency spectra for the 16 h after getting up and the 7 h after going to bed did not reveal single dominant frequencies but rather complex frequency distributions. Power spectra of the daytime data revealed no group differences for activity and no heart rate differences between smoking smokers and nonsmokers. In abstinent smokers, however, a significant reduction of heart rate frequencies slower than 1 cycle/135 min and a significant increase of heart rate frequencies faster than 1 cycle/20 min were observed as compared with all other groups. This effect persisted over the 2 abstinence days, suggesting an activity-independent change in the frequency distribution of heart rates after quitting smoking. 相似文献
The inactivation of Bacillus subtilis spores during long-term exposure (up to several months) to extreme dryness (especially vacuum) is strain-dependent, through only to a small degree. During a first phase (lasting about four days) monolayers of spores lose about 20% of their viability, regardless of the strain studied. During this phase loss in viability can be equally attributed both to damages of hydrophobic structures (membranes and proteins) and DNA. During a second phase lasting for the remaining time of experimental observation (weeks, months and years) the loss in viability is slowed. A viability of 55% to 75% (depending on the strain) is attained after a total exposure of 36 days. The loss in viability during the second phase can be correlated with the occurrence of DNA double strand breaks. Also covalent DNA-protein cross-links are formed by vacuum exposure. If the protein moiety of these cross-links is degraded by proteinase K-treatment in vitro additional DNA double strand breaks result. The data are also discussed with respect to survival on Mars and in near Earth orbits. 相似文献
Summary Chronic myelogenous leukemia (CML) patients in chronic phase display compromised lymphokine-activated killer (LAK) cell induction, which is partly restored after therapy with interferon . However, the relative resistance of the leukemic cells from these patients to autologous or allogeneic LAK lysis is not affected by this treatment. In an attempt to render CML cells more susceptible to lysis or cytostasis, they were precultured in serum-free medium with or without recombinant growth factors. In eight patients studied, interleukin-3 (IL-3) significantly enhanced the spontaneous short-term (6-day) proliferation of CML cells, with retention of ability to form colonies in methylcellulose. Culture in either medium alone or IL-3 led to a significant enrichment of CD14+ and CD33+ cells but to a reduction in CD34+ cells. In contrast, culture of the same cells in IL-2 (to generate autologous LAK activity) resulted in a loss of CD14+ and CD33+ as well as CD34+ cells but in a significant increase in CD3+ and CD56+ cells. Despite similarities in their phenotypes, IL-3 cultured cells but not those cultured in medium alone acquired susceptibility to lysis by the IL-2-cultured autologous LAK cells. Somatic embryos of hybrid larch (Larixleptoeuropaea) whichhad been matured for 4 weeks on maturation medium, developednormally on medium supplemented with 60 µM ABA, but abnormallyon medium with no ABA. A comparative structural and histochemicalinvestigation was carried out on these two types of mature embryos.At the light microscope level, differences between both treatmentswere visible only after 23 weeks of maturation. At aroundthis time, abnormal development becomes evident macroscopically:ABA-minus embryos remain rather stubby as opposed to the morecylindrically shaped ABA-plus embryos. Whereas somatic embryosmatured with ABA consist of densely cytoplasmic cells showinga high rate of cell division, ABA-minus embryos are largelymade up of expanded and highly vacuolate cells, indicating thatgrowth in the latter is mainly due to cell expansion and notdivision. After 4 weeks of maturation, ABA-minus embryos beginto elongate in the hypocotyl region, and precocious germinationwas observed frequently. Again, these morphogenetic events werelargely due to abnormal timing of cell expansion. Histochemically,storage proteins were found only in somatic embryos maturedfor 4 weeks with ABA. This observation is in line with resultsobtained by total protein analysis, yielding significantly lowertotal protein contents in ABA-minus embryos both on a freshweight and a per embryo basis after 45 weeks of maturation.Deposition of starch grains mainly in the cortex tissue of thehypocotyl region was observed within 2 weeks of maturation invarying amounts regardless of ABA supply. Polyphenols, in particularcatechins and proanthocyanidins, were present in all embryosfrom the very onset of development. They were localized preferentiallyin the proximal suspensor cells and the basal region of theembryo. However, accumulation of polyphenols was generally muchmore pronounced in embryos matured without ABA, indicating alack of biochemical regulatory competence in those embryos. During 1988 and 1989, 409 specimens of southern African Anura comprising 50 species in 9 families were checked for opalinids in the cloaca. Cepedea acuta n. sp. was found in six of 19 Tomopterna cryptotis and four of seven T. krugerensis (Ranidae); C. affinis (Nazaretskaja, 1992) in two of four Afrixalus aureus, two of four Hyperolius horstocki, 23 of 42 H. marmoratus, one of seven H. pusillus, two of six H. semidiscus, and eight of 12 H. tuberilinguis (Hyperoliidae); C. magna Metcalf, 1923 in one of 20 Bufo garmani, eight of 33 B. gutturalis and two of nine B. rangeri (all Bufonidae), one of two Heleophryne natalensis (juveniles) (Heleophrynidae), four of six Kassina maculata, three of seven K. senegalensis and two of six Semnodactylus wealii (all Hyperoliidae), three of five Phrynomerus bifasciatus (Microhylidae), three of 12 Phrynobatrachus natalensis, 11 of 19 Tomopterna cryptotis, 13 of 14 T. delalandii, one of seven T. krugerensis and one of six T. natalensis (all Ranidae), and four of five Chiromantis xerampelina (Rhacophoridae); Cepedea vanniekerkae n. sp. in one of 19 Tomopterna cryptotis. It is suggested that the Ranidae (in particular the genus Tomopterna) and the Hyperoliidae (in particular the genus Hyperolius) are among the major carriers of Cepedea in the Afrotropical Region. 相似文献
High performance liquid chromatography (HPLC), a valuable tool for characterization of peptides, is frequently used in combination with sensitive radioimmunoassays (RIA). The shadow phenomenon, representing carry-over of the peptide from previous application of the standard, can appear to result in the presence of endogenous peptide in the test sample when none is actually there. With delta sleep-inducing peptide (DSIP), we found the shadowing to be as high as 10%, although it was only 1% with 125I-Tyr-DSIP. Thus, when HPLC-RIA systems are used for identification of peptides, caution must be used to avoid false positive results. 相似文献
In order to study the effects of temperature enhancement on alpine calcareous grassland species, a warming experiment was carried out in the Berchtesgaden National Park (Southeast Germany, Northern Calcareous Alps) between 2002 and 2004. The study was conducted in stands of the Carex sempervirens and the Carex firma communities; the two most widespread grassland types in the alpine zone of the Northern Calcareous Alps. The temperature of the vegetation stand and the upper soil was passively enhanced using open top chambers (OTCs). The construction of the OTCs was appropriate since temperature was clearly increased while water conditions (humidity, soil water content) were not changed.
By comparing manipulated (temperature enhancement) with non-manipulated plots, the effects of warming on growth and reproduction of selected key species were studied. To test if vegetation response to temperature enhancement is at least partly due to increases in nutrient availability, soil solution concentrations of nitrate and ammonium were analysed.
We found that most of the studied plant species are sensitive to temperature enhancement. Growth and/or reproduction of 12 of the 14 studied species were significantly stimulated by warming. Only two species showed no response; none of the species experienced decreases in growth or reproduction. Dwarf shrubs and graminoids showed a stronger response than herbaceous perennials. A significant effect of warming on nutrient availability could not be detected. The observed response of vegetation is therefore mainly caused by direct and not by indirect temperature effects. 相似文献
