人鼻咽癌细胞基因组中瘤基因Ha－ras－1的Dnase Ⅰ超敏感位点的初步研究

ＤＮａｓｅⅠ超敏感位点的研究能够发现潜在的调控基因转录活化的位点,比较正常人外周血有核细胞,淋巴瘤细胞株Ｐ３ＨＲ１和人鼻咽癌低分化磷癌细胞株ＨＯｎＥ１和ＨＮＥ２中Ｈａ－ｒａｓ－１瘤基因的ＤＮａｓｅⅠ超敏感位点发现,只有ＨＯＮＥ１和ＨＮＥ２细胞基因组中存在一个ＤＮａｓｅⅠ超敏感位点,位于第一个外显子上游０．３７ｋｂ处,上述结果提示正常白细胞和Ｐ３ＨＲ１细胞中Ｈａ－ｒａｓ－１基因处于失活状态,而在鼻咽癌细胞基因组中则处于活化状态,它的活化可能与０．３７ｋｂ处的ＤＮＡ序列有密切的关系。     

Haemodynamic and renin responses to prostacyclin infusion in Na depleted and Na restricted sheep

The haemodynamic and renin responses to prostacyclin (PGI2) infusion were examined in sheep during sodium depletion and dietary sodium restriction. The haemodynamic effects of PGI2 infusion in sodium depleted and sodium restricted sheep were similar to those obtained in the sodium replete animal. The renin proportionate response to PGI2 was not altered by sodium restriction but blunted by sodium depletion, compatible with the hypothesis that endogenous PGI2 is high in Na depletion.     

DLGCN:基于图卷积网络的药物-lncRNA[]关联预测

为实现高通量识别新的药物-长链非编码RNA(Long non-coding RNA, lncRNA)关联,本文提出了一种基于图卷积网络模型来识别潜在药物-lncRNA关联的方法DLGCN(Drug-LncRNA graph convolution network)。首先,基于药物的结构信息和lncRNA的序列信息分别构建了药物-药物和lncRNA-lncRNA相似性网络,并整合实验证实的药物-lncRNA关联构建了药物-lncRNA异质性网络。然后,将注意力机制和图卷积运算应用于该网络中,学习药物和lncRNA的低维特征,基于整合的低维特征预测新的药物-lncRNA关联。通过效能评估,DLGCN的受试者工作特性曲线下面积(Area under receiver operating characteristic, AUROC)达到0.843 1,优于经典的机器学习方法和常见的深度学习方法。此外,DLGCN预测到姜黄素能够调控lncRNA MALAT1的表达,已被最近的研究证实。DLGCN能够有效预测药物-lncRNA关联,为肿瘤治疗新靶点的识别和抗癌药物的筛选提供了重要参考。     

抗菌肽VIP在毕赤酵母中的高效表达及鉴定

基于人抗菌肽VIP(Vasoactive intestinal peptide)基因序列,按照毕赤酵母密码子偏好性设计引物;用SOE-PCR法扩增目的基因;然后将目的基因克隆至毕赤酵母分泌型表达载体pPICZαA上,构建VIP分泌表达菌株GS115-p PICZαA-vip。用甲醇诱导96 h收集上清,用质谱进行鉴定,结果显示分泌表达产物与人抗菌肽VIP理论值(3 326.82 Da)完全一致,表明人抗菌肽VIP成功得到分泌表达。琼脂糖凝胶扩散法实验结果显示,重组VIP对大肠杆菌Escherichia coli ATCC25922和金黄色葡萄球菌Staphylococcus aureus ATCC25923都有很强的抗菌活性,MIC(Minimal inhibitory concentration)分别为8 mmol/L和16 mmol/L。进一步细胞毒性和溶血性实验结果显示,重组VIP对正常细胞NCM460和IPEC-J2没有毒性,其对SD大鼠红细胞不具有溶血活性。通过透射电镜观察了VIP的抗菌机制,结果显示VIP主要通过破坏细胞膜的方式抑杀细菌。本研究为人抗菌肽VIP的开发应用和大量生产奠定了基础。     

Diallyl trisulfide induces apoptosis and inhibits proliferation of A549 cells in vitro and in vivo

Lung cancer is the leading cause of cancer-related mortality all over the world. In recent years, pulmonary adenocarcinoma has surpassed squamous cell carcinoma in frequency and is the predominant form of lung cancer in many countries. Epidemiological investigations have shown an inverse relationship between garlic (Allium sativum) consumption and death rate from many cancers. Diallyl trisulfide (DATS) is one of the garlic-derived compounds (also known as: organosulfer compounds, OSC). DATS can induce apoptosis and inhibit the growth of many cancer cell lines. Our study demonstrated that the apoptotic incidents induced by DATS were a mitochondria-dependent caspase cascade through a significant decrease of the anti-apoptotic Bcl-2 that resulted in up-regulation of the ratio of Bax/Bcl-2 and the activity of caspase-3, -8, and -9. Eventually, DATS induced the apoptosis and inhibited the proliferation in a concentration- and time-dependent manner. Furthermore, by establishing an animal model of female BALB/c nude mice with A549 xenografts, we found that oral gavage of DATS significantly retarded growth of A549 xenografts in nude mice without causing weight loss or any other side effects compared with the control group. All the evidence both in vitro and in vivo suggested that DATS could be an ideal anti-cancer drug.     

Recent developments in membrane-protein structural genomics

Recent work has identified the topology of almost all the inner membrane proteins in Escherichia coli, and advances in nuclear magnetic resonance spectroscopy now allow the determination of α-helical membrane protein structures at high resolution. Together these developments will help overcome the current limitations of high-throughput determination of membrane protein structures.     

Comparative Effects of Hispidulin,Genistein, and Icariin with Estrogen on Bone Tissue in Ovariectomized Rats

Icariin, Genistein, and Hispidulin have been proven to have estrogen-like and antiosteoporotic activity and can be potentially used for the treatment of osteoporosis. The present study found that Icariin, Genistein, and Hispidulin treatments, emulating estrogen, significantly contributed to bone density. Comparative effects of Icariin, Genistein, and Hispidulin with estrogen on in ovariectomized rats were investigated. Our results showed that genistein was found to have superior bone protective effects against osteoporosis among genistein, Icariin, and Hispidulin.     

不同冷季型牧草品种的产草量及品质比较分析

采用随机区组试验设计,对引进的8个冷季型牧草品种进行产草量和营养成分的比较分析.结果表明:4次刈割的总鲜重和总干重在品种间差异极显著,且以蓝天堂黑麦草(7号)最高.品种间的粗蛋白含量差异显著,其中第1次刈割以奇可利(3号)最高,蓝天堂黑麦草(7号)居第2位;第2次仍以奇可利(3号)最高,特高黑麦草(1号)和蓝天堂黑麦草(7号)居其次;第3次以紫花苜蓿(8号)最高,其次是多年生黑麦草(4号);第4次以奇可利(3号)最高,多年生黑麦草(4号)居第2位;粗纤维、粗脂肪和粗灰分在品种间的差别较小.综合产草量和品质两方面考虑,蓝天堂黑麦草(7号)可作为本地首选推广的冷季型牧草品种,奇可利(3号)和多年生黑麦草(4号)可作为备选推广的牧草品种.
