The sex-ratio trait described in several Drosophila species is a type of naturally occurring X-linked meiotic drive that causes males bearing a sex-ratio X chromosome to produce progenies with a large excess of females. We have previously reported the occurrence of sex-ratio X chromosomes in Drosophila simulans. In this species, because of the co-occurrence of drive suppressors, the natural populations and the derived laboratory strains show an equal sex-ratio even when sex-ratio X chromosomes are present at a high frequency. The presence of sex-ratio X chromosomes is established via crosses with a standard strain that is devoid of drive suppressors. In this article, we show first that the sex-ratio trait in D. simulans results from the action of several X-linked loci. Second we describe drive suppressors on each major autosome as well as on the Y chromosome. The Y-linked factors suppress the drive partially whereas the autosomal suppression can be complete.  相似文献

The sex-ratio trait we describe here in Drosophila simulans results from X-linked meiotic drive. Males bearing a driving X chromosome can produce a large excess of females (about 90%) in their progeny. This is, however, rarely the case in the wild, where resistance factors, including autosomal suppressors and insensitive Y chromosomes, prevent the expression of the driver. In this study, we searched for drive and resistance factors in strains of Drosophila simulans collected all over the world. Driving X chromosomes were found in all populations whenever a good sample size was available. Their frequency may reach up to 60%. However, the presence of driving X chromosomes never results in an excess of females, due to the systematic co-occurrence of resistance factors. The highest frequencies of driving X chromosomes were observed in islands, while populations from East and Central Africa (the supposed center of origin of the species) showed the highest level of resistance. The geographical pattern of drive and resistance factors, as well as the results of crosses between strains from different geographical areas, suggest that the sex-ratio system described here has a unique and ancient origin in the species.  相似文献

Aeromonas hydrophila, an opportunist human pathogen of low virulence, was shown to display a high degree of sensitivity upon exposure to hydrogen peroxide. As with other species, Aer. hydrophila is able to develop the capacity to resist loss of viability induced by such oxidative stress. Development of stress resistance follows the archetypal profile where pre-exposure of a population to sub-lethal levels of H2O2 stimulates onset of tolerance to further exposure. Acquisition of tolerance critically requires nascent protein synthesis. Further analysis demonstrated population growth phase influences the degree of sensitivity of the organism. Late stationary phase cultures demonstrate a decreased sensitivity compared with younger populations. Significantly, it was also determined that stock culture age influenced the level of sensitivity of the derived experimental culture, where an increased stock culture age corresponded with enhanced resistance to H2O2. These data show that Aer. hydrophila population phenotype is influenced by the phenotype of the donor stock culture.  相似文献

Summary In the leaves (but not corms) of the submerged aquatic Isoetes storkii malic acid concentration fluctuated from 22 eg g FW-1 in the evening to 171 eg g FW-1 in the morning. Associated with this was a change in titratable acidity of 152 eg g FW-1 between morning and evening. 14C carbon was fixed in both the light and the dark, though the amount of carbon fixed in the light was more than that fixed in the dark. Autoradiographs show 88% of 14CO2 fixed in the dark is recovered after 1 h, in malic acid and the remainder in one other unidentified product, whereas these two products contain less than 15% of the 14C fixed after 1 h exposure to 14CO2 in the light. It is suggested that CAM metabolism in this aquatic species may be related to the low availability of CO2 for photosynthesis during the day in its aquatic environment and that this metabolic pathway may prove common in the genus Isoetes.  相似文献

The motile mesophilic Aeromonas are ubiquitous to a wide variety of aquatic environments including drinking water distribution systems. Concern over the presence of mesophilic Aeromonas in public drinking water supplies has been expressed in recent years as it has been regarded as a pathogenic organism of importance in gastroenteritis. A major drinking water distribution system in north-east Scotland was monitored over a 12 month period to determine the prevalence of mesophilic Aeromonas. These data were examined in relation to chlorine concentration, pH, temperature, rainfall and the standard bacteriological indicators of water quality. Aeromonas were isolated to varying degrees from 21 of the 31 reservoirs investigated. The maximum recovery observed during the study was 605 cfu in 300 ml. The probability of isolation generally decreased with increasing levels of chlorination, although this oxidant was found to be ineffective in many reservoirs. Certain reservoirs with poor chlorination profiles yielded very few isolates, whereas some highly chlorinated sites liberated Aeromonas frequently and in relatively high numbers. A seasonal pattern in the incidence of Aeromonas emerged with infrequent isolation during the winter period increasing to a peak during the summer, with most isolates recovered when water temperature was >12 °C. An association was demonstrated between the pattern of Aeromonas isolations and that of rainfall. No relationship was apparent between incidence of Aeromonas and total heterotrophic plate counts.  相似文献

Metal pollution, in combination with other environmental stressors such as acid deposition and climate change, may disturb metal biogeochemical cycles. To investigate the influence of dissolved organic carbon, acidity and seasonality on metal geochemistry, this study has described concentrations of 19 metals as they pass through an acidified forested catchment on the Precambrian Shield in south-central Ontario, Canada. Metal, dissolved organic carbon (DOC) and sulphate (SO4 2−) concentrations fluctuate throughout the catchment compartments as the water passes through and interacts with vegetation, soils and bedrock. Relationships among metals, DOC and SO4 2− are most pronounced in compartments where DOC and SO4 2− exhibit high variability, namely in the throughfall, organic horizon soil water, and wetland-draining stream. Metal, DOC and SO4 2− concentrations varied seasonally in the streams, and temporal coherence occurred among metal, DOC and SO4 2− concentrations in the organic horizon soil water and the wetland-draining stream (PC1). In the wetland-draining stream, the highest DOC, Cr, Cu, Fe, Pb, and V concentrations occur in the summer, whereas concentrations of SO4 2− and most other metals peak in the fall after a period of drought. Despite the rural location, provincial water quality objectives for surface water were exceeded for many metals when the peak fall values occurred.  相似文献

The Colilert defined substrate technology system allows specific, one-step detection of both coliforms and Escherichia coli while claiming to suppress the influence of non-coliform heterotrophs. The Colilert assay was examined in order to determine whether organisms from the genus Aeromonas could interfere and cause production of a false-positive coliform result as aquatic Aeromonas are known to constitute a fraction of the heterotrophic population found in drinking water. Results obtained clearly demonstrate that Aeromonas sp. can elicit a positive coliform type reaction at very low densities. Cell suspensions as low as 1 × 101 cells 10 ml−1 were observed to yield a positive reaction using Colilert reagent 4 weeks short of shelf-life expiry. Use of aged Colilert for monitoring water quality could lead to over-estimation of coliforms as Aeromonas have been identified in many treated drinking water supplies.  相似文献

The intracellular glutathione levels of two human tumor lines and seven murine tumor lines were determined in order to investigate the role of oxidant injury in tumor cell sensitivity to human rTNF (rhTNF). Correlations were found between high intracellular glutathione levels and in vivo tumor resistance to rhTNF, and on the other hand, low glutathione levels and rhTNF sensitivity. The transplantable murine fibrosarcoma, Meth A, a TNF-sensitive line in vivo, was less sensitive to rhTNF and host toxicity was reduced when the hosts were pretreated with uric acid, a major reactive oxygen scavenger in humans and certain other primates. Conversely, pretreatment of the tumor-bearing hosts with DL-buthionine-(S,R)-sulfoximine, an inhibitor of GSH biosynthesis, resulted in an increased sensitivity of Meth A to rhTNF. This effect was not limited to tumor-bearing mice, as rats pretreated with diethyl maleate, a compound which irreversibly binds glutathione, were more sensitive to rhTNF toxicity than control rats. On the other hand, pretreatment with N-acetyl cysteine, an oxidant scavenger, reduced the toxicity of rhTNF treatment in rats. The data are consistent with the hypothesis that tumor cell sensitivity to rhTNF in vivo is dependent on its capacity to buffer oxidative attack. In addition, host toxicity is also related to the production of reactive oxygen species. Activated effector cells such as granulocytes and macrophages are hypothesized to produce most of this damage by their respiratory burst and oxidant release, although the direct action of rhTNF may also contribute to oxidative injury in vivo.  相似文献
