Colchicum autumnale L. is a monocotyledonous geophyte with hysteranthous leaves, i.e. flowering and leaf growth occur in different time periods.
Because after the starch, the second prominent storage compound of corm is represented by proteins, we were interested in
nitrogen remobilisation during the annual life cycle of C. autumnale. In this context the content of soluble and insoluble proteins were measured in parallel with determination of some exo-and
endopeptidase activities. Our results indicate that the continual proteolysis occurs in both mother and new daughter corms
during the whole life-cycle of the plant. L-Ala-aminopeptidase and trypsin-like endopeptidase were the most active peptidases
in both mother and daughter corms. As the protein level of mother corm did not change significantly during the development
of the future above-ground part under the soil surface (the first, autumnal developmental stage), the developmental profile
of nitrate reductase activity was estimated followed by evaluation of total nitrogen and amino acid contents. Significant
activity of root nitrate reductase was detected in the roots only in the second, vernal stage. Our results showed that the
stored proteins constituted a relevant nitrogen source partly required by the growth processes of the late autumnal stage,
but mainly by the intensive growth of leaves and reproductive structures during the second, photosynthetically active stage

Exposure of cells to ultraviolet radiation (UVR) is one of the best studied and most used model system for the examination
of the biological effects of DNA damage, its repair and tolerance. The major product after UVR treatment is cyclobutane pyrimidine
dimer (TT, TC, CC). Pyrimidine dimers are repaired by a direct reversal called photoreactivation or by excision of damage
in a process of nucleotide excision repair. Several methods have been developed for the detection and quantification of pyrimidine
dimers in DNA. The technique of Small and Greimann, in which DNA is incubated with the pyrimidine dimer-specific endonuclease,
was used for the analysis of mutant strains with impaired excision repair system of the unicellular green alga Chlamydomonas reinhardtii. Another method is based on the binding of specific monoclonal antibodies to pyrimidine dimers. The aim of our work was to
compare these two techniques with the use of mutant strains of C. reinhardtii — uvsX1 and uvsX2 which are assumed to be deficient in DNA damage recognition. One of their traits was sensitivity to UVR which could be caused
by breakdown of the excision repair pathway. The results suggest that the immuno-approach is suitable for the detection of
DNA damage induced by UVR.

In a previous study, it was demonstrated that N-(phenethyl)succinamic acid (PESA) derivatives form a new category of root-promoting substances which do not exhibit auxin-like activities, such as stem elongation and leaf epinasty (Soejima et al., 2000 [Plant Cell Physiol. 41s: 197]). In this study, N-[2-(3-indolyl)ethyl]succinamic acid (IESA) and N-[2-(1-naphthyl)ethyl]succinamic acid (NESA) were synthesized, and their biological activities were evaluated. In an adzuki root-promoting assay, IESA and NESA exhibited root-promoting activity equivalent to PESA. In adzuki stem elongation assays, elongation activity was not observed in the stem segments soaked in either an IESA or NESA aqueous solution, whereas the stem segments immersed in Indole-3-acetic acid (IAA) or 1-naphthylacetic acid (NAA) aqueous solution were clearly elongated. In an epinastic bending study, IAA and NAA exhibited leaf epinasty, whereas IESA and NESA did not, suggesting that the IESA and NESA derivatives belong to the same category of root-promoting substances as PESA derivatives and are different from auxin-like substances. In addition, eleven kinds of IESA derivatives and nineteen kinds of NESA derivatives were synthesized, and their root-promoting activities were measured. The activities of methyl ester derivatives were approximately three times higher than that of the acid compounds, with exceptions for some compounds. The partition coefficient (P) between 1-octanol and water for each IESA, NESA, and PESA derivative was measured in order to evaluate the hydrophobicity of their molecules and to determine their structure–activity relationship. The results indicate that the root-promoting activity of the acid compounds was significantly correlated with their hydrophobicity, whereas that of ester derivatives was not correlated. 相似文献

This study aims to assess the photoprotective potential of desiccation-induced curling in the light-susceptible old forest
lichen Lobaria pulmonaria by using chlorophyll fluorescence imaging. Naturally curled thalli showed less photoinhibition-induced limitations in primary
processes of photosynthesis than artificially flattened specimens during exposures to 450 μmol m−2 s−1 in the laboratory after both 12- (medium dose treatment) and 62-h duration (high dose treatment). Thallus areas shaded by
curled lobes during light exposure showed unchanged values of measured chlorophyll fluorescence parameters (F
V/F
M, ΦPS II), whereas non-shaded parts of curled thalli, as well as the mean for the entire flattened thalli, showed photoinhibitory
limitation after light treatments. Furthermore, the chlorophyll fluorescence imaging showed that the typical small-scale reticulated
ridges on the upper side of L. pulmonaria caused a spatial, small-scale reduction in damage due to minor shading. Severe dry-state photoinhibition readily occurred
in flattened and light-treated L. pulmonaria, although the mechanisms for such damage in a desiccated and inactive stage are not well known. Natural curling is one strategy
to reduce the chance for serious photoinhibition in desiccated L. pulmonaria thalli during high light exposures. 相似文献

Ultrathin sectioning of submerged mycelium of Claviceps purpurea Tul. producing clavine alkaloids revealed yeast-like budding resulting in asexual sporesblastospores. These deciduous spores were born by extended hyphal cells and retained the same ultrastructure of cell organelles. Both the extended hyphae and the blastospores resembled the cells of ergot sclerotial tissue. A surface culture of C. purpurea Tul. producing no alkaloids was used as a reference. 相似文献

The most widely known theoretical basis for the hypothesis of terminal investment is the classic model by George C. Williams (1966). Although this model predicts that reproductive effort (i.e. the proportion of available resources devoted to reproduction) increases with decreasing reproductive value, it implies that reproductive allocation in absolute terms should remain stable. This contrasts with the empirical evidence on terminal investment reported to date: the vast majority of positive case studies report an increase in some aspect of reproductive allocation in absolute terms. Also, a substantial number of studies have failed to record terminal investment, despite expectations. Here, I present a simple conceptual model which explains such results. I argue that to explain terminal investment, an organism’s reproductive capacity must not be considered as a common pool of resources (often described by the term ‘reproductive value’), but as a set of different resources which are not easily convertible to each other, and should be exhausted in balance. Thus, if one resource accidentally decreases, in response, the others must be expended at higher rate. To test this model, each reproductive allocation should be measured in a more specific currency (or currencies) than traditional ‘reproductive effort’. Human erythrocyte membranes which had been thoroughly extracted with organic solvents contained 20 nmol of fatty acids/mg dry wt. The major fatty acids were palmitic and stearic with their monoethenoic derivatives as minor constituents. No other fatty acids were detected. When solvent-extracted membranes were digested with Pronase about 90% of the original content of fatty acids was retained in the insoluble residue. Fatty acids were linked to membrane proteins through alkali-labile bonds of which 30% were of a thiol ester and the remainder of an O-ester type. This conclusion is based on differential liberation of fatty acids by hydroxylamine at pH 7.0 and pH 11.0. Two extracts of membranes enriched in peripheral proteins (bands 1, 2, 5 and 2.1, 4.1, 4.2, 6) were prepared and extracted with organic solvents but each contained about six times less fatty acids than the parent solvent-extracted membranes. Glycophorin A contains little if any covalently bound fatty acids. Anion transporter (band 3) contains about 1 mol of thiol ester of fatty acid. This accounts for about half of the thiol ester-linked fatty acids in the parent solvent-extracted membranes. Developmental processes are closely connected to certain states of epigenetic information which, among others, rely on methylation
of chromatin. S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH) are key cofactors of enzymes catalyzing DNA and histone methylation. To study the consequences
of altered SAH/SAM levels on plant development we applied 9-(S)-(2,3-dihydroxypropyl)-adenine (DHPA), an inhibitor of SAH-hydrolase, on tobacco seeds during a short phase of germination
period (6 days). The transient drug treatment induced: (1) dosage-dependent global DNA hypomethylation mitotically transmitted
to adult plants; (2) pleiotropic developmental defects including decreased apical dominance, altered leaf and flower symmetry,
flower whorl malformations and reduced fertility; (3) dramatic upregulation of floral organ identity genes NTDEF, NTGLO and NAG1 in leaves. We conclude that temporal SAH-hydrolase inhibition deregulated floral genes expression probably via chromatin
methylation changes. The data further show that plants might be particularly sensitive to accurate setting of SAH/SAM levels
