Summary
Chrysomela confluens produces a salicylaldehyde-based defensive secretion which is very effective against generalist predators and apparently produced at no cost. If no cost defenses are common, then one of the basic assumptions in the plant-herbivore literature, i.e. tradeoffs among defense, reproduction, and growth, must be reconsidered. We examined the effectiveness of this defense by exposing defended larvae and larvae whose secretion had been removed to a generalist predator. Larvae which had their secretions intact were attacked by only 7% of the ants which encountered them, and none of these larvae suffered serious damage. In contrast, those which had been milked of their secretions immediately prior to exposure were attacked in 48% of such encounters, and two-thirds of the larvae were killed. Larvae which had been milked 24 or 72 h before exposure, then allowed to regenerate their defenses, were attacked at rates indistinguishable from larvae that had not been milked. Thus regenerated defenses are just as effective as original defenses. We also tested the hypothesis that the cost of defense production and maintainence would be reflected in reductions in developmental rates and final adult mass and increases in leaf consumption rate. We found that larvae which were milked daily of their secretions manifested no measurable cost of recharging reservoirs. Milked larvae grew and fed at the same rates as their control sibs, and became adults of equal or slightly larger size. The liberation of glucose from salicin, a precursor present in leaves of salicaceous hosts, during the production of salicylaldehyde apparently provides enough of an energetic benefit to offset the cost of maintaining an effective defense. Consistent with this hypothesis, we did not find that milked larvae compensated for increased nutritional or salicin demands by increasing their feeding rates. Although this patterns is familiar to chemical ecologists it is generally unappreciated in the plant-herbivore literature. It is likely that many arthropod herbivore defensive systems come at little or no cost, given the intimacy of association between herbivores and their food plants. Sequestration of host plant defensive chemicals which eliminates the cost of synthesis is common in arthropods. The de novo synthesis of chemical defenses may be less costly than expected if it is integrated into other parts of an insects metabolism. Calculations based on the bond energies or molecular constitution of the compounds will not yield a complete perception of cost. Tests over the life of the herbivore, coupled with an understanding of the herbivore's metabolism, are necessary. 相似文献

Prostate cancer (CaP) is the second leading malignancy in men. The role of epithelial cell adhesion molecule (EpCAM), also known as CD326, in CaP progression and therapeutic resistance is still uncertain. Here, we aimed to investigate the roles of EpCAM in CaP metastasis and chemo/radioresistance. Expression of EpCAM in CaP cell lines and human CaP tissues was assessed using immunofluorescence and immunohistochemistry, respectively. EpCAM was knocked down (KD) in PC-3, DU145 and LNCaP-C4-2B cells using small interfering RNA (siRNA), and KD results were confirmed by confocal microscope, Western blotting and quantitative real time polymerase chain reaction (qRT-PCR). Cell growth was evaluated by proliferation and colony formation assays. The invasive potential was assessed using a matrigel chamber assay. Tumorigenesis potential was measured by a sphere formation assay. Chemo-/radiosensitivity were measured using a colony formation assay. Over-expression of EpCAM was found in primary CaP tissues and lymph node metastases including cancer cells and surrounding stromal cells. KD of EpCAM suppressed CaP proliferation and invasive ability, reduced sphere formation, enhanced chemo-/radiosensitivity, and down-regulated E-cadherin, p-Akt, p-mTOR, p-4EBP1 and p-S6K expression in CaP cells. Our findings suggest that EpCAM plays an important role in CaP proliferation, invasion, metastasis and chemo-/radioresistance associated with the activation of the PI3K/Akt/mTOR signaling pathway and is a novel therapeutic target to sensitize CaP cells to chemo-/radiotherapy. 相似文献

Current methods for aligning biological sequences are based on dynamic programming algorithms. If large numbers of sequences or a number of long sequences are to be aligned, the required computations are expensive in memory and central processing unit (CPU) time. In an attempt to bring the tools of large-scale linear programming (LP) methods to bear on this problem, we formulate the alignment process as a controlled Markov chain and construct a suggested alignment based on policies that minimise the expected total cost of the alignment. We discuss the LP associated with the total expected discounted cost and show the results of a solution of the problem based on a primal-dual interior point method. Model parameters, estimated from aligned sequences, along with cost function parameters are used to construct the objective and constraint conditions of the LP problem. This article concludes with a discussion of some alignments obtained from the LP solutions of problems with various cost function parameter values. 相似文献

The outer armour of fossil jawless fishes (Heterostraci) is, predominantly, a bone with a superficial ornament of dentine tubercles surrounded by pores leading to flask-shaped crypts (ampullae). However, despite the extensive bone present in these early dermal skeletons, damage was repaired almost exclusively with dentine. Consolidation of bone, by dentine invading and filling the vascular spaces, was previously recognized in Psammolepis and other heterostracans but was associated with ageing and dermal shield wear (reparative). Here, we describe wound repair by deposition of dentine directly onto a bony scaffold of fragmented bone. An extensive wound response occurred from massive deposition of dentine (reactionary), traced from tubercle pulp cavities and surrounding ampullae. These structures may provide the cells to make reparative and reactionary dentine, as in mammalian teeth today in response to stimuli (functional wear or damage). We suggest in Psammolepis, repair involved mobilization of these cells in response to a local stimulatory mechanism, for example, predator damage. By comparison, almost no new bone is detected in repair of the Psammolepis shield. Dentine infilling bone vascular tissue spaces of both abraded dentine and wounded bone suggests that recruitment of this process has been evolutionarily conserved over 380 Myr and precedes osteogenic skeletal repair. 相似文献

Lianas are poorly characterized for central African forests. We quantify variation in liana composition, diversity and community structure in different forest types in the Yangambi Man and Biosphere Reserve, Democratic Republic of Congo. These attributes of liana assemblages were examined in 12 1-ha plots, randomly demarcated within regrowth forest, old growth monodominant forest, old growth mixed forest and old growth edge forest. Using a combination of multivariate and univariate community analyses, we visualize the patterns of these liana assemblage attributes and/or test for their significant differences across forest types. The combined 12 1-ha area contains 2,638 lianas (≥2 cm diameter) representing 105 species, 49 genera and 22 families. Liana species composition differed significantly across forest types. Taxonomic diversity was higher in old growth mixed forests compared to old growth monodominant and regrowth forests. Trait diversity was higher than expected in the regrowth forest as opposed to the rest of forest types. Similarly, the regrowth forest differed from the rest of forest types in the pattern of liana species ecological traits and diameter frequency distribution. The regrowth forest was also less densely populated in lianas and had lower liana total basal area than the rest of forest types. We speculate that the mechanism of liana competitive exclusion by dominant tree species is mainly responsible for the lower liana species diversity in monodominant compared to mixed forests. We attribute variation in liana community structure between regrowth and old growth forests mostly to short development time of size hierarchies. 相似文献

CD44 and CD147 are associated with cancer metastasis and progression. Our purpose in the study was to investigate the effects of down-regulation of CD44 or CD147 on the metastatic ability of prostate cancer (CaP) cells, their docetaxel (DTX) responsiveness and potential mechanisms involved in vitro and in vivo. CD44 and CD147 were knocked down (KD) in PC-3M-luc CaP cells using short hairpin RNA (shRNA). Expression of CD44, CD147, MRP2 (multi-drug resistance protein-2) and MCT4 (monocarboxylate tranporter-4) was evaluated using immunofluorescence and Western blotting. The DTX dose-response and proliferation was measured by MTT and colony assays, respectively. The invasive potential was assessed using a matrigel chamber assay. Signal transduction proteins in PI3K/Akt and MAPK/Erk pathways were assessed by Western blotting. An in vivo subcutaneous (s.c.) xenograft model was established to assess CaP tumorigenecity, lymph node metastases and DTX response. Our results indicated that KD of CD44 or CD147 decreased MCT4 and MRP2 expression, reduced CaP proliferation and invasive potential and enhanced DTX sensitivity; and KD of CD44 or CD147 down-regulated p-Akt and p-Erk, the main signal modulators associated with cell growth and survival. In vivo, CD44 or CD147-KD PC-3M-luc xenografts displayed suppressed tumor growth with increased DTX responsiveness compared to control xenografts. Both CD44 and CD147 enhance metastatic capacity and chemoresistance of CaP cells, potentially mediated by activation of the PI3K and MAPK pathways. Selective targeting of CD44/CD147 alone or combined with DTX may limit CaP metastasis and increase chemosensitivity, with promise for future CaP treatment. 相似文献

This paper presents a practical numerical method for separatingand estimating growth and mortality coefficients in stage- orsize-structured populations using only observations of the relativeor absolute abundance of each stage. The method involves writinga system of linear ordinary differential equations (ODEs) modellingthe rate of change of abundance. The solution of the differentialsystem can be numerically approximated using standard (e.g.sixth-order Runge-Kutta-Felhberg) methods. An optimization problemwhose solutions yield optimal coefficients fora given model is formulated. The ODE numerical integration techniquecan then be employed to furnish required function and gradientinformation to the optimization algorithm. The data-fittingsoftware package ODRPACK is then successfully employed to estimateoptimal coefficients for the ODE population model. The contribution of the legume community to the nitrogen cycle during natural forest regeneration remains poorly understood. We systematically assessed the changes in abundance and nodulation status of all legumes, across taxa and plant types, in a forest succession gradient in the Equateur Province of the Democratic Republic of the Congo.
