Water-Stress Induced Changes in Concentrations of Proline and Other Solutes in Growing Regions of Young Barley Leaves

Young, intact barley Hordeum vulgare L. cv. ‘Arivat’seedlings stressed with PEG or NaCl were measured for waterstatus and levels of proline, sugars and inorganic cations ingrowing (basal 1.0 cm) and other areas of leaves. In growingregions, reductions in _n were found within 1 h in seedlingsstressed with PEG (–0.8 M Pa) but 4 h were required beforeproline increased and proline accounted for less than 5% ofthe osmotic adjustment after 24 h. Increases in proline occurredto a lesser extent in expanded mid-blade regions where osmoticadjustment was less. After one or more days stress, prolinelevels in basal tissues were 10 x higher than in mid-blade tissues,and proline increases in these and intervening tissues occurredwithout loss of turgor. When seedlings were stressed for extendedperiods with mild to moderate concentrations of PEG, prolineelevations in basal to mid-blade areas were proportional toreductions in tissue and _n NaCl-stress led to similar responsesexcept prohne increases per unit decrease in _x, were 2–3x greater than in PEG-stressed tissues. Time-course studiesshowed levels of inorganic cations were not altered in the growingareas during the first few h of PEG-induced stress, but glucoseincreased as _x, decreased. The results showed that proline increaseis not the cause but could be a consequence of osmotic adjustment,and its increase is not due to cell death. They also providedindependent evidence that stress responses of growing tissuesdiffer substantially from expanded areas in young barley leaves. Key words: Proline, water stress, growing regions     

In Vitro Regression of Tadpole Tail by Thyroid Hormone

DERBY successfully maintained the tail of tadpole ( Rana pipiens) in vitro over a period of 2 weeks in a physiological salt solution (1). When we tried to apply DERBY'S methods of the tissue culture to tadpoles of bullfrog, Rana catesbeiana , it was found that the tissue regressed spontanously without stimulation of thyroid hormone. Several different media were examined in order to select a better culture medium for the bullfrog tadpole tissues. RPMI-1640 medium supplemented with insulin and transferrin was found to be satisfactory for this aim. With this improved medium, the interaction between the epidermis and the mesenchyme has been investigated during the hormone-induced tadpole tail regression and the epidermal dependence of the mesenchyme regression was demonstrated by the following three experiments. (i) Some of surgically prepared mesenchymes regressed in responce to thyroid hormone. In these cases the mesenchymes were revealed to be contaminated with the remaining epidermal cells. (ii) Complete removal of the epidermis was accomplished by the chemical treatment. The mesenchyme thus obtained ("nude tail fin") was insensible to thyroid hormone. (iii) "Skin conditioned medium" (SCM) was prepared by culturing the skin in the presence and absence of thyroid hormone. Nude tail fin regressed when cultured in the SCM containing thyroid hormone.

WE have shown^1,2 that cytochrome c, when orally administered, is absorbed intestinally. The effects of parenterally administered cytochrome c on the liver have been studied by Drabkin³ and Murano⁴. We have also studied the effects of orally administered cytochrome c on experimental liver injury⁵.
