Cockroach ileum has a high capability to concentrate mercury compared with other tissues. Part of the mercury contained in the soluble phase of this organ is bound to metallothionein. Polysomes prepared from frozen rat brain powder were fractionated by centrifugation in a sucrose gradient. Individual fractions were used to program a reticulocyte lysate in a run-off reaction. The products of cell-free synthesis were assayed for the brain-specific enolase (14.3.2 protein) and S100 protein by immunoprecipitation with specific antisera and for tubulin by two-dimensional electrophoresis in polyacrylamide slab gels. The relative synthesis of these proteins by unfractionated free brain polysomes were 0.1 per cent, 0.05 per cent and 0.7 per cent respectively. After centrifugation in a sucrose gradient polysomes synthesizing S100 protein were separated from those synthesizing the other two markers. There was a threefold enrichment in the specific messenger RNA activity for each of the three proteins studied in their respective peak fractions of polysomes. 相似文献

A single injection of a massive dose of Al-gluconate (4 mg.kg-1) into the saphenous vein of the rat did not provoke any ultrastructural damage in the liver cells, from 2 min. to 8 days after the injection. Al hepatocytes overload appeared only in nuclei and not in nuclei and not in lysosomes, contrarily to chronic intoxications. Nuclear Al concentration concerned only a small fraction of the quantity injected; another part was sequestered by the macrophage system after precipitation in the blood as phosphates chemically transformed during their incorporation in lysosomes. This effective detoxication mechanism which functioned probably after a first absorption by the hepatocyte, was likely to depend upon the form of gluconate and would explain the resistance of liver cells. 相似文献

Histochemical methods applied to the fat body ofFormica rufa queens show during the postembryonnic development, the following results:
- Synthesis of lipids, glycogen and protein, successively occur in the cytoplasm of larval adipose cells. At the time of cocoon spinning, the energetic reserves (glycogen and mainly lipids) regress. During the nymphosis, the proteins are used for the histogenesis of the imaginal organs; yet, part of the proteins synthetised by the prenymph is transformed by the nymph into lipids, or constitutes the protein reserves of the swarming imago.
- The nitrogenous products, which are copious for the larva, undergo transformations during the nymphosis, but reappear in microscristallin form at the hatching.

Summary The metabolic fate of translation initiation factor after inhibition of protein synthesis by different means has been investigated. We have found a decay in initiation factor activity when protein synthesis is blocked by chloramphenicol but not during arginine starvation of PA1 (Rel–) or PA2 (Rel+) strains or during puromycin incubation. These results suggest that inactivation of certain initiation factors occurs when the regeneration of ribosomal subunits from polysomes is inhibited in the cells.Complementation experiments indicate that IF3 factor activity is preferentially affected during chloramphenicol treatment.Same preferential inhibition of IF3 activity seems to occur during in vitro incubation of crude IF. 70S ribosomes or 30S subunits protect this factor against the inactivation. Preliminary results seem tosuggest that ATP is implicated in this in vitro inactivation process. 相似文献

Résumé Les cellules du mésentéron des ouvrières, des reines et des mâles de Formica polyctena F. possèdent un certain nombre de particularités cytologiques dont l'évolution a été suivie au cours du développement post-embryonnaire et du cycle annuel.A l'apex des cellules de régénération les microvillosités se différencient avant l'élimination des cellules caduques larvaires ou nymphales. A partir de la nymphose une activité sécrétoire apocrine se manifeste dans la partie dorsale de l'épithélium du mésentéron, l'ensemble des cellules assurant par ailleurs la fonction absorbante de l'organe. Il existe deux sortes d'inclusions cytoplasmiques, des polysaccharides et des concrétions minérales. Les polysaccharides sont surtout abondants chez les larves et les nymphes: le glycogène, polysaccharide de réserve, est utilisé au cours de l'histogénèse; des mucopolysaccharides acides, d'origine golgienne, représentent une sécrétion muqueuse. Les sphérocristaux sont constitués de strates concentriques de phosphates et chlorures de calcium et d'une matrice de mucopolysaccharides. La cristallisation des éléments minéraux s'effectue, à partir de la nymphose seulement, dans les citernes ergastoplasmiques. Cette accumulation d'ions pourrait être en relation avec le régime alimentaire de l'insecte ou représenter une voie d'excrétion.

Histophysiological studies on the postembryonic development and the annual cycle of Formica II. Histochemical and ultrastructural characteristics of the midgut of F. polyctena
Summary The midgut cells of workers, queens and males of the ant Formica polyctena show cytological characteristics which were studied in the course of postembryonic development and annual cycle. The microvilli of the regenerating cells appear before the elimination of the regressing larval and pupal cells. At the time of pupation, an active phase of apocrine secretion begins in the dorsal part of the midgut epithelium, while the absorptive function is carried out by all cells of the organ.Two types of cytoplasmic inclusions coexist: polysaccharides and mineral concretions. The polysaccharides are particulary abundant in larvae and pupae. Glycogen is metabolized during histogenesis; acid mucopolysaccharides, elaborated in the Golgi apparatus, represent a mucous secretion. The spherites are composed of concentric strata of calcium phosphate and chloride and a matrix of mucopolysaccharides. These minerals form in the ergastoplasmic cisternae of pupal cells only. Their accumulation could be related to the insect's diet, or it could reflect a process of excretion.

Two different approaches were used to study the bioavailability of sediment-bound lead. In vitro techniques simulating the potential metal desorption under conditions prevailing in the digestive tract were assayed on a contaminated sediment. An experimental model of a food chain was designed to determine the retention of lead in the soft tissues of oysters according to the environmental source of the metal (water or sediment). Neither enzymatic action nor leaching at low pH (both aspects of digestion) induce the release of important lead amounts from particles. Therefore, after 3 weeks of exposure, the retention of lead from the trophic source is lower (1%) compared with direct contamination (5%). Lysosomes are the major intracellular structures responsible for lead storage in the gills, digestive tract and digestive gland. The abundance of lysosomes and their lead amount vary according to the gross concentrations in the soft tissues. The cytopathological data are in agreement with the results about lead accumulation: in oysters exposed to sediment-bound lead, impairments are not so marked as in individuals contaminated directly from water but the same organelles are concerned. Mitochondrial impairments may be related to the effect of lead on cellular respiration processes and changes involving the granular endoplasmic reticulum may have an effect on the level of protein synthesis. Cellular extrusions carrying away numerous lysosomes loaded with lead could account for the balancing of lead incorporation between 2 and 3 weeks of exposure. 相似文献

Liver tubulin was analyzed and characterized during the development of the mouse, from embryonic stages to adult. Separation and identification of isotubulins were achieved by isoelectric focusing, two-dimensional gel electrophoresis and peptide mapping. Liver tubulin is a heterogeneous population composed of 7 isoforms, 4 α and 3 β. By comparison with the events occurring at the tubulin level during brain development, we observed that 1) tubulin heterogeneity is less important in liver than in brain 2) no appearance of new isotubulins takes place during development 3) the only developmental change detectable in our gels is an accumulation of an α tubulin subunit which is absent from brain isotubulin population. The relationship between this α isotubulin accumulation and liver secretion during development is discussed. 相似文献
