Recombinant adeno-associated virus(rAAV) has proven to be a promising gene delivery vector for human gene therapy. However, its application has been limited by difficulty in obtaining enough quantities of high-titer vector stocks. In this paper, a novel and highly efficient production system for rAAV is described. A recombinant herpes simplex virus type 1(rHSV-1) designated HSV1-rc/△UL2, which expressed adeno-associated virus type2(AAV-2) Rep and Cap proteins, was constructed previously. The data confirmed that its functions were to support rAAV replication and packaging, and the generated rAAV was infectious. Meanwhile, an rAAV proviral cell line designated BHK/SG2, which carried the green fluorescent protein(GFP) gene expression cassette, was established by transfecting BHK-21 cells with rAAV vector plasmid pSNAV-2-GFP. Infecting BHK/SG2 with HSV1-rc/△UL2 at an MOI of 0.1 resulted in the optimal yields of rAAV, reaching 250 transducing unit(TU) or 4.28×104 particles per cell. Therefore, compared with the conventional transfection method, the yield of rAAV using this "one proviral cell line, one helper virus" strategy was increased by two orders of magnitude. Large-scale production of rAAV can be easily achieved using this strategy and might meet the demands for clinical trials of rAAV-mediated gene therapy. 相似文献

Watermelon production is threatened by fusarium wilt caused by Fusarium oxysporum f.sp. niveum (FON) in continuous cultivation system. Some elements, mainly allelochemicals, released from living roots or decayed plants
might be associated with the disease. The purpose of this work was to evaluate the possible impact of coumarin, one kind of
watermelon allelochemical, on FON. Furthermore, possible new mechanisms might be investigated during the ecological interactions
of plant-microbe. Results showed that coumarin strongly inhibited growth of FON leading to a decrease in its biomass, dry
weight of mycelia of FON in a liquid culture. The dry weight was decreased by 62.9% compared with control. The hyphal growth
of FON on plates was stopped at high (>400 mg l−1) concentrations of coumarin. At 320 mg l−1, sporulation and enzyme activities of FON were also severely suppressed by coumarin. The yield of conidia, and the activities
of proteinase, cellulase, and amylase were reduced by 98.9%, 79.7%, 29.8% and 15.9% respectively. However, conidial germination
and mycotoxin (MT) production of FON were greatly stimulated, being increased by 55.7% and 14.9 fold at 320 mg l−1 respectively. We conclude that coumarin acted as an allelochemical substance to inhibit growth and pathogenic enzyme activities
of FON but to stimulate mycotoxin production and conidial germination. It was suggested that coumarin acted as a signal transduction

HrpNEa is a harpin protein from Erwinia amylovora, a bacterial pathogen that causes fire blight in rosaceous plants. Treating plants with HrpNEa stimulates ethylene and abscisic acid (ABA) to induce plant growth and drought tolerance, respectively. Herein, we report
that both growth hormones cooperate to mediate the role of HrpNEa in promoting root growth of Arabidopsis thaliana seedlings. Root growth is promoted coordinately with elevation in levels of ABA and ethylene subsequent to soaking of germinating
seeds of wild-type (WT) Arabidopsis in a solution of HrpNEa. However, these responses are arrested by inhibiting WT roots from synthesizing ethylene as well as sensing of ABA and ethylene.
The effects of HrpNEa on roots are also nullified in ethylene-insensitive etr1-1 and ein5-1 mutants and in the ABA-insensitive mutant abi2-1 of Arabidopsis. These results provide evidence for presence of a relationship between root growth enhancement and signaling by ABA and ethylene
in response to HrpNEa. Nevertheless, when HrpNEa is applied to leaves, ethylene signaling is active in the absence of ABA signaling to promote plant growth. This suggests
the presence of a different signaling mechanism in leaves from that in roots.

In this study, we have investigated the plant growth promoting effect of Bacillus mucilaginosus strain D4B1, a rhizosphere soil organism, and its transgenic strain NKTS-3 on tobacco planting. The transgenic strain contains a phytase
expression cassette that can express high active phytase extracellularly and hydrolyze phytate in the soil to liberate inorganic
phosphorus for the growth of tobacco plants. Greenhouse study and field experiments showed that both wild-type B. mucilaginosus and the transgenic strain could promote tobacco plant growth. Moreover, the transgenic strain promoted tobacco plant growth
(235% more than control in pot experiments and 125% more than control in field experiments) was higher than the wild-type
B. mucilaginosus (183% more than control in pot experiments and 108% more than control in field experiments). In addition, the inoculation
with transgenic rhizobacteria could significantly improve root acquisition of phosphorus and increase the phosphorus content

Almost 95% of the entire population of the Siberian crane (Grus leucogeranus) winter in Poyang Lake, China, where they forage on the tubers of the submerged aquatic macrophyte Vallisneria spiralis. The Three Gorges Dam on the Yangtze River may possibly affect this food source of the Siberian crane by affecting the light
intensity reaching the top of the V. spiralis canopy. In this study, the photosynthetically active radiation at the top of the V. spiralis canopy (PARtc) in Lake Dahuchi was modeled from 1998 to 2006, and the potential impacts of changes in water level and turbidity on the
underwater light climate of V. spiralis were analyzed. PARtc was calculated from incident irradiance while the losses due to reflection at the water surface, absorption, and scattering
within the water column were taken into consideration. The results indicated significant differences in PARtc between years. Six years of water level and Secchi disk depth records revealed a seasonal switching of the lake from a turbid
state at low water levels in autumn, winter, and spring to a clear state at high water levels during the monsoon in summer.
The highest PARtc occurred at intermediate water levels, which were reached when the Yangtze River forces Lake Dahuchi out of its turbid state
in early summer and the water becomes clear. The intended operation of the Three Gorges Dam, which will increase water levels
in May and June, may advance the moment when Lake Dahuchi switches from turbid to clear. We suggest that this might increase
production of V. spiralis and possibly improve the food habitat conditions for wintering Siberian crane in Poyang Lake.

The effects of nano-TiO2 (rutile) on the photochemical reaction of chloroplasts of spinach were studied. The results showed that when spinach was
treated with 0.25% nano-TiO2, the Hill reaction, such as the reduction rate of FeCy, and the rate of evolution oxygen of chloroplasts was accelerated
and noncyclic photophosphorylation (nc-PSP) activity of chloroplasts was higher than cyclic photophosphorylation (c-PSP) activity,
the chloroplast coupling was improved and activities of Mg2+-ATPase and chloroplast coupling factor I (CF1)-ATPase on the thylakoid membranes were obviously activated. It suggested that photosynthesis promoted by nano-TiO2 might be related to activation of photochemical reaction of chloroplasts of spinach. 相似文献
