Ferritin as a source of iron was consiered. A good iron absorption rate appears in normal rats with an in vivo absorption technique. The same absorption appears in iron-deficient animals. The effect of antioxidants applied in one step of a cryopreservation protocol by encapsulation–dehydration on recovery and genetic stability of mint shoot tips has been studied. Glutathione (0.16 or 0.24 mM), ascorbic acid (0.28 or 0.43 mM) and α-tocopherol (vitamin E) were added to the preculture medium (0.3 M sucrose). DNA was extracted from three different types of samples: leaves from shoots, callus at the base of shoots and callus. RAPD and AFLP markers were used to assess the genetic stability. The use of antioxidants did not improve recovery after cryopreservation. One of the genotypes, ‘MEN 198’, showed higher percentage of stable samples than the other one, ‘MEN 186’ (56 vs. 37?%; considering all treatments and types of explant). The use of vitamin E improved the percentage of stable samples with respect to control treatment (no antioxidant) in ‘MEN 186’. No differences in the percentages of stable samples were observed among cryopreserved and non-cryopreserved (treated similarly without immersion in liquid nitrogen) plant material. This morphometric study of the geographic variation in the Abies religiosa–hickelii–guatemalensis complex is based on samples from 15 Guatemalan and 12 Mexican populations, two populations of A. religiosa s.str. and A. hickelii s.str., and herbarium specimens of A. hickelii, A. vejarii and varieties of A. guatemalensis. The multivariate methods employed were principal components analysis, and UPGMA clustering. The multivariate and univariate analyses based on 231 operational taxonomic units imply that although morphological differences exist distinct morphospecies cannot be recognized within the A. religiosa–hickelii–guatemalensis complex. A Mantel’s test reports that taxonomic dissimilarities are significantly related to geographic distance. We suggest, therefore, that A. religiosa, A. hickelii and A. guatemalensis are merged so that A. hickelii is referred to as A. religiosa subsp. hickelii (Flous & Gaussen) U. Strandby, K.I. Chr. & M. Sørensen, comb. et stat. nov. and A. guatemalensis as A. religiosa subsp. mexicana (Martínez) U. Strandby, K.I. Chr. & M. Sørensen, comb. nov. Matrix metalloproteinase (MMP) 13 (collagenase 3) is an extracellular matrix remodeling enzyme that is induced in myofibroblasts during the earliest invasive stages of human breast carcinoma, suggesting that it is involved in tumor progression. During progression of mammary carcinomas in the polyoma virus middle T oncogene mouse model (MMTV-PyMT), Mmp13 mRNA was strongly upregulated concurrently with the transition to invasive and metastatic carcinomas. As in human tumors, Mmp13 mRNA was found in myofibroblasts of invasive grade II and III carcinomas, but not in benign grade I and II mammary intraepithelial neoplasias. To determine if MMP13 plays a role in tumor progression, we crossed MMTV-PyMT mice with Mmp13 deficient mice. The absence of MMP13 did not influence tumor growth, vascularization, progression to more advanced tumor stages, or metastasis to the lungs, and the absence of MMP13 was not compensated for by expression of other MMPs or tissue inhibitor of metalloproteinases. However, an increased fraction of thin collagen fibrils was identified in MMTV-PyMT;Mmp13(-/-) compared to MMTV-PyMT;Mmp13(+/+) tumors, showing that collagen metabolism was altered in the absence of MMP13. We conclude that the expression pattern of Mmp13 mRNA in myofibroblasts of invasive carcinomas in the MMTV-PyMT breast cancer model recapitulates the expression pattern observed in human breast cancer. Our results suggest that MMP13 is a marker of carcinoma-associated myofibroblasts of invasive carcinoma, even though it does not make a major contribution to tumor progression in the MMTV-PyMT breast cancer model. 相似文献

The strength and direction of phenological responses to changes in climate have been shown to vary significantly both among
species and among populations of a species, with the overall patterns not fully resolved. Here, we studied the temporal and
spatial variability associated with the response of several insect species to recent global warming. We use hierarchical models
within a model comparison framework to analyze phenological data gathered over 40 years by the Japan Meteorological Agency
on the emergence dates of 14 insect species at sites across Japan. Contrary to what has been predicted with global warming,
temporal trends of annual emergence showed a later emergence day for some species and sites over time, even though temperatures
are warming. However, when emergence data were analyzed as a function of temperature and precipitation, the overall response
pointed out an earlier emergence day with warmer conditions. The apparent contradiction between the response to temperature
and trends over time indicates that other factors, such as declining populations, may be affecting the date phenological events
are being recorded. Overall, the responses by insects were weaker than those found for plants in previous work over the same
time period in these ecosystems, suggesting the potential for ecological mismatches with deleterious effects for both suites
of species. And although temperature may be the major driver of species phenology, we should be cautious when analyzing phenological
datasets as many other factors may also be contributing to the variability in phenology. 相似文献

An integrated process for the indoor production of 13C labelled PUFA from Phaeodactylum tricornutum is presented. The core of the process is a bubble column photobioreactor from which the exhaust gas from the reactor is returned to the culture by a low pressure compressor. To avoid accumulation of dissolved oxygen in the culture medium, the exhaust gas is bubbled through a sodium sulphite solution before returning it to the reactor. Carbon is removed from the medium before inoculating the alga, then labelled 13CO2 is injected for pH control and carbon supply. The reactor has been operated in semicontinuous mode at a dilution rate of 0.01 h–1, a biomass productivity of 0.1 g L–1 d–1 being obtained. Under this conditions both pH and dissolved oxygen were correctly controlled and the adequacy of the system for autotrophic production of labelled biomass was demonstrated. Analysis by GC-MS revealed that the fatty acids content of the biomass obtained was 10% d.wt., the content of eicosapentaenoic acid was 2.5% d.wt. All the fatty acids were labelled, more that 90% of the carbon present in these fatty acids was 13C. Element analysis of biomass and supernatant showed that 59.5% of injected carbon was assimilated into the biomass whereas 33% remained in the supernatant, and 7.5% remained undetected. Due to the high cost of 13CO2 different strategies for the optimisation of labelled carbon use are proposed. 相似文献

Growth and Differentiation Factor 1 (GDF-1) has been implicated in left-right patterning of the mouse embryo but has no other known function. Here, we demonstrate a genetic interaction between Gdf1 and Nodal during anterior axis development. Gdf1-/-;Nodal+/- mutants displayed several abnormalities that were not present in either Gdf1-/- or Nodal+/- single mutants, including absence of notochord and prechordal plate, and malformation of the foregut; organizing centers implicated in the development of the anterior head and branchial arches, respectively. Consistent with these deficits, Gdf1-/-;Nodal+/- mutant embryos displayed a number of axial midline abnormalities, including holoprosencephaly, anterior head truncation, cleft lip, fused nasal cavity, and lack of jaws and tongue. The absence of these defects in single mutants indicated a synergistic interaction between Nodal and GDF-1 in the node, from which the axial mesendoderm that gives rise to the notochord, prechordal plate, and foregut endoderm originates, and where the two factors are co-expressed. This notion was supported by a severe downregulation of FoxA2 and goosecoid in the anterior primitive streak of double mutant embryos. Unlike that in the lateral plate mesoderm, Nodal expression in the node was independent of GDF-1, indicating that both factors act in parallel to control the development of mesendodermal precursors. Receptor reconstitution experiments indicated that GDF-1, like Nodal, can signal through the type I receptors ALK4 and ALK7. However, analysis of compound mutants indicated that ALK4, but not ALK7, was responsible for the effects of GDF-1 and Nodal during anterior axis development. These results indicate that GDF-1 and Nodal converge on ALK4 in the anterior primitive streak to control the formation of organizing centers that are necessary for normal forebrain and branchial arch development. 相似文献

The “cost‐benefit” hypothesis states that avian body organs show mass changes consistent with the trade‐off between their functional importance and maintenance cost, which may vary throughout the annual cycle. Flightless moulting common scoter Melanitta nigra in Danish marine waters select rich undisturbed offshore feeding areas lacking predators, suggesting active feeding during moult. We tested four predictions relating to organ size during flightlessness in moulting male common scoter under this hypothesis. Namely that (i) pectoral muscles would show atrophy followed by hypertrophy, but that there would be no change in (ii) leg muscles and heart (the locomotory architecture required to sustain diving for food), (iii) digestive organs and liver (required to process food), or (iv) fat deposits (because birds could fulfil daily energy requirements from locally abundant food resources). Dissection of scoters collected at different stages during wing moult south of the Danish island of Læsø provided data on organ size that were consistent with these predictions. Pectoral muscle mass showed a c.23% atrophy during the middle of the flightless period relative to that at the end of moult. There was no significant loss in leg muscle, heart, digestive organs (except gizzard mass), liver, fat reserves or body mass with remigial growth. These findings are consistent with the hypothesis that common scoter moult in a rich feeding area, and rely on their diet to meet the nutritional requirements of remigial moult. These results differ in detail from those of a similar study of terrestrial feeding moulting greylag geese Anser anser, but because of the widely differing ecology of the species concerned, both sets of findings provide strong support for the hypothesis that variations in phenotypic plasticity in size of fat stores, locomotor and digestive organs can be interpreted as evolutionary adaptations to meet the conflicting needs (feather growth, nutritional challenges and predator avoidance) of the flightless moult period in different Anatidae species. 相似文献

Deep‐sea octopuses of the genus Muusoctopus are thought to have originated in the Pacific Northern Hemisphere and then diversified throughout the Pacific and into the rest of the World Ocean. However, this hypothesis was inferred only from molecular divergence times. Here, the ancestral distribution and dispersal routes are estimated by Bayesian analysis based on a new phylogeny including 38 specimens from the south‐eastern Pacific Ocean. Morphological data and molecular sequences of three mitochondrial genes (16S rRNA, COI and COIII) are presented. The morphological data confirm that specimens newly acquired from off the coast of Chile comprise two species: Muusoctopus longibrachus and the poorly described species, Muusoctopus eicomar. The latter is here redescribed and is clearly distinguished from M. longibrachus and other closely related species in the region. A gene tree was built using Bayesian analysis to infer the phylogenetic position of these species within the species group, revealing that a large genetic distance separates the two sympatric Chilean species. M. longibrachus is confirmed as the sister species of Muusooctopus eureka from the Falkland Islands; while M. eicomar is a sister species of Muusoctopus yaquinae from the North Pacific, most closely related to the amphi‐Atlantic species Muusoctopus januarii. Molecular divergence times and ancestral distribution analyses suggest that genus Muusoctopus may have originated in the North Atlantic: one lineage dispersed directly southward to the Magellan region and another dispersed southward along the Eastern Pacific to the Southern Ocean and Antarctica. The Muusoctopus species in the Southern Hemisphere have different phylogenetic origins and represent independent invasions of this region. 相似文献

Nerve growth factor (NGF), like many other growth factors and hormones, binds to two different receptor molecules on responsive cells. The product of the proto-oncogene trk, p140trk, is a tyrosine kinase receptor that has been identified as a signal-transducing receptor for NGF, while the role of the low affinity NGF receptor, p75NGFR, in signal transduction is less clear. The crystal structure of NGF has recently been determined, although structures involved in receptor binding and biological activity are unknown. Here we show that Lys-32, Lys-34, and Lys-95 form a positively charged interface involved in binding to p75NGFR. Simultaneous modification of Lys-32 with either of the two other lysines resulted in loss of binding to p75NGFR. Despite the lack of binding to p75NGFR, these mutants retained binding to p140trk and biological activity, demonstrating a functional dissociation between the two NGF receptors. 相似文献
