Preliminary data on the polysaccharide composition of mycelium of the submerged grown fungus Cunninghamella japonica (synonymous with C. echinulata) were obtained. Mild acid hydrolysis of the mycelium led to formation of glucose, mannose and galactose, whereas acid treatment under drastic conditions afforded glucosamine as the hydrolysis product of chitin and chitosan, the summary content of both glucosaminoglycans being estimated as about 35%. Sequential treatment of the mycelium with hot water, 2% aqueous NaOH and 10% AcOH gave rise to several polysaccharide fractions, which were characterized by their monosaccharide composition. The yield ofchitosan extracted by AcOH was negligible. Additional purification of the fraction obtained by the action of alkali afforded a polysaccharide preparation, which was shown to be a linear (1-->3)-alpha-D-glucopyranan according to the data of chemical methods of structural analysis and NMR spectroscopy. It was concluded that Cunninghamella japonica differs from several other known representatives of Mucorales by the presence of this alpha-D-glucan, as well as by low content of chitosan and polyuronides. 相似文献

The content of dipicolinic acid (DPA) was assayed in the spores of Bacillus anthracoides 96 during various stages of its growth. The content of DPA was ca. 10.7 per cent of the dry biomass weight in the seven-day-old culture containing 96 to 99 per cent of the spores in a "starvation" medium. The morphology of the culture was modified, and the content of DPA in the spores fell to 3.6 per cent half an hour after the inoculation into the medium favourable for the growth (MPA). During the following one to four hours of the germination, the refraction index of the spores and the content of DPA in them decreased (the content of DPA to 2 per cent). 相似文献

Exogenous ergosterol and cholesterol were found to affect the growth and lipogenesis of the oomycete fungusPythium debaryanum, which is unable to synthesize de novo steroid compounds. These sterols stimulated the growth of the fungus during its submerged
cultivation in glucose-peptone medium. This was accompanied by the shortening of the lag phase, the lengthening of the period
of active growth, and by a 3.7-or 4.3-fold increase in the maximum biomass in response to the addition of ergosterol or cholesterol,
respectively. In the presence of ergosterol, the cellular content of polyenoic fatty acids increased, and the relative content
of eicosapolyenoic fatty acids reached 31.4% of the total amount of fatty acids in cells. Conversely, cholesterol decreased
the cellular content of polyenoic acids, and the relative content of eicosapolyenoic acids fell to 19.6% of the total amount
of fatty acids. It may be inferred that exogenous sterols enhance the yield of pharmacologically active polyenoic acids because

The main stages in the production of biodiesel fuel from lipids of filamentous fungi belonging to the order Mucorales are described. Fungi of the family Cunninghamellaceae have been screened; the lipogenic activity of the examined strains has been assessed; and a producer generating up to 50% of lipids, represented by triacylglycerols, has been found. The substitution effect of a source of carbon and nitrogen with less expensive components (in particular, various industrial wastes) has been studied, as well as their influence on the quantity and major characteristics of the final product. An ecologically friendly method for extracting lipids from fungal mycelia, utilizing supercritical technologies, has been used. A correlation between the lipid content in the spore inoculum and the maximal lipid content in biomass has been discovered; this correlation is proposed for optimizing the biotechnology and increasing the yield of final products. 相似文献

The cerebrosides produced by the soil filamentous fungus Mortierella alpina strain KG-1/95 account for about 13% of the total polar lipids extractable from lyophilised cells with chloroform/methanol mixtures. By means of 1H NMR and (13)C NMR spectroscopy, matrix-assisted laser-desorption ionisation mass spectrometry, and chemical degradation experiment, they have been shown to be 1-O-beta-D-glucopyranosyl-2-N-(2'-D-hydroxyalkanoyl)-9-methylsphinga-4(E),8(E)-dienines, the fatty acid composition of which is unusual and consists of 2-hydroxytridecanoic (4%), 2-hydroxytetradecanoic (60%), 2-hydroxypentadecanoic (20%), and 2-hydroxyhexadecanoic (16%) acids. 相似文献

The effect of exogenously added vitamin E and its synthetic analogues (the hydrophilic form of vitamin E and chromans C13and C1) at a concentration of 9.86 × 10–5M on the growth, lipogenic activity, and the fatty acid composition of the eicosapolyenoic acid–synthesizing oomycete Pythium debaryanumwas studied. The effect was found to depend on the molecular structure of particular compounds. For instance, vitamin E and chroman C13stimulated fungal growth, whereas chroman C1inhibited it. The hydrophilic form of vitamin E enhanced the lipogenic activity of the oomycete. The studied compounds, which possess antioxidant activity, did not exert any noticeable effect on the content of eicosapolyenoic acids and the degree of the unsaturation of fungal lipids. 相似文献

The blocking potency of P- and L-selectin was studied for certain small molecule mannosides and their polyacrylamide (PAA, 30 kDa) conjugates in comparison to SiaLe(x) and fucoidan. Two experimental systems were used: (1) solid phase static assay based on recombinant selectins, and (2) P-selectin dependent rat peritoneal inflammation. betaMan-SC6H4NO2- p was four times more potent P-selectin inhibitor as compared to SiaLe(x). Docking of this molecule onto the P-selectin carbohydrate-binding site demonstrated that a nitro group enabled an electrostatic interaction with residue Lys 84, while the phenyl ring and the CH2 at C-6 contacted the CH2 groups of the same Lys residue. In vivo, betaMan-SC6H4NO2- p blocked experimental inflammation better than SiaLe(x), but significantly lower than fucoidan. In vitro Man-polyacrylic acid conjugates appeared to be very potent inhibitors comparable to fucoidan, uncharged Man-PAA proved rather active, comparable to SiaLe(x)-PAA both in vitro, and in vivo, whereas mannan did not display any P-selectin blocking effect. 相似文献

Lipogenic activity and fatty acid composition of two strains of Geomyces pannorum were studied in the course of fungal growth. The strains were isolated from an Arctic cryopeg lens (VKM FW-2241) and from Central Russia (VKM F-3808). The adaptive reactions in both strains towards the temperature decreasing to 2°C involved intensification of the fatty acid desaturation. The degree of lipid unsaturation increased mainly due to a higher amount of α-linolenic acid (α-C18:3) especially in the case of strain VKM FW-2241. Elevated NaCl concentration in the medium enhanced the level of linoleic acid (C18:2) which apparently played a specific role in osmoprotection. Strain VKM FW-2241 was more tolerant to increased salinity than strain VKM F-3808. Almost complete inhibition of the growth of strains VKM F-3808 and VKM FW-2241 occurred at salinity of 10 and 20%, respectively; however, the viability of the strains was not affected. Under the combined effect of high salinity and hypothermia, the ratio between C18:2 and α-C18:3 acids was intermediate, indicating that these acids were involved in two adaptation mechanisms. The inhibition of fungal growth under stress was found to result in lipid overproduction. An increased pool of energy-rich lipids in fungi possibly contributes to their strategy of cell survival. 相似文献

The cell lipids of the zygomycete Absidia corymbifera F-965 extracted with isopropanol and CHCl3-MeOH mixtures at the exponential growth phase comprise 20+/-2% of mycelium dry wt. The lipids consist of: triacylglycerols (51% of the total lipids extracted), diacylglycerols (9%), monoacylglycerols (3%), ergosterol (5%), ergosterol peroxide (5alpha,8alpha-epidioxyergosta-6,22-diene-3beta-ol) (3%), fatty-acid esters of ergosterol (less than 0.5%), free fatty acids (4%), glucocerebroside (3%), and glycerophospholipids (22%). The main phospholipids are phosphatidylethanolamine (39% of the total phospholipids), phosphatidyl-myo-inositol (17%), diphosphatidylglycerol (12%), phosphatidic acid (7%), phosphatidylcholine (6%), phosphatidylglycerol (3%), and two unusual phospholipids reported earlier, N-acetylphosphatidylethanolamine (7%) and N-ethoxycarbonyl phosphatidylethanolamine (9%). In addition, two unknown acidic phospholipids are present in traces. Saturated fatty acids of the lipids are dominated by n-hexadecanoic acid and unsaturated ones by octadecenoic acid; octadecadienoic and octadecatrienoic acids are present in lesser amounts. Ergosterol peroxide as well as the above glucocerebroside which contains 9-methylsphinga-4(E),9(E)-dienine have first been revealed in zygomycetes. 相似文献
