Irreversible inhibition of the human placental NADP-linked 15-hydroxyprostaglandin dehydrogenase/9-ketoprostaglandin reductase by glutathione thiosulfonate

Oxidation of glutathione disulfide by a mixture of performic and hydrochloric acids leads to the formation of several compounds that are stronger inhibitors than glutathione disulfide of the placental enzyme that possess both NADP-linked 15-hydroxyprostaglandin dehydrogenase and 9-ketoprostaglandin reductase activities. The only one of these inhibitors that has been identified is glutathione thiosulfonate. The others are unstable and may include glutathione sulfinyl sulfone and glutathione disulfone. Since the enzyme appears to have a glutathione binding site in close proximity to its active site and glutathione thiosulfonate reacts with free sulfhydryl groups, the effects of this thiosulfonate on the enzyme were examined in more detail. Glutathione thiosulfonate and methyl methanethiosulfonate cause a time-dependent irreversible inhibition of both the hydroxyprostaglandin dehydrogenase and the ketoprostaglandin reductase activities, presumably by reacting with a free sulfhydryl at the prostaglandin binding site. Experiments with PGA1-glutathione show that this sulfhydryl is not necessary for the catalytic activity of the enzyme as long as the substrate can bind at the glutathione site.     

Differential reactivities of lysines in calmodulin complexed to phosphatase

Calmodulin and calmodulin complexed with calcineurin phosphatase were trace labeled with [3H]acetic anhydride and the incorporation of [3H]acetate into each epsilon-amino lysine of calmodulin was measured. The relative reactivities of calmodulin lysines were higher in the presence of Ca2+ than in the presence of EGTA, and the order was: Lys-75 greater than Lys-94 greater than Lys-148 greater than or equal to Lys-77 greater than Lys-13 greater than or equal to Lys-21 greater than Lys-30. The changes in relative reactivity implied a change in conformation. When calmodulin was complexed with the phosphatase, Lys-21, Lys-77, and Lys-148 were most protected, implying that these residues are at or near the interaction sites or are conformationally perturbed by the interaction. Lys-30 and Lys-75 were slightly protected, lysine 13 showed no change, while lysine 94 significantly increased in reactivity. Comparison with results obtained from myosin light chain kinase using a similar technique (Jackson, A. E., Carraway, K. L., III, Puett, D., and Brew, K. (1986) J. Biol. Chem. 261, 12226-12232) reveals that calmodulin may interact with each of the two enzymes similarly at or near Lys-21, Lys-75, and Lys-148; one difference with phosphatase is that complex formation also involved Lys-77. These findings suggest that calmodulin interacts differently with its target enzymes.     

Cultivation of trematodes in chick embryos

The chick embryo can be used as a substitute for adult animals in the cultivation of a variety of parasites. Bernard Fried describes its use in cultivating trematodes, and suggests that the system might be useful as a preliminary chemotherapeutic screen.     

Hemolytic and coagulation properties of Sphaeridiotrema globulus (Trematoda)

Metacercariae of Sphaeridiotrema globulus (Trematoda) were obtained from naturally infected Goniobasis virginica (Pleuroceridae). Excysted metacercariae were placed individually on blood-agar plates. After 8 hr of incubation at 42 C the plates exhibited beta-hemolysis. The reaction zone was approximately 1.5 mm in diameter. Laboratory-reared mallard ducks (Anas platyrhynchos) were infected with S. globulus. Mallards developed fatal sphaeridiotremiasis and demonstrated increased prothrombin time on days 3 and 6 postinfection.     

The effects of crowding on adults of Echinostoma revolutum (Digenea: Echinostomatidae) in experimentally infected golden hamsters, Mesocricetus auratus

All 30 female golden hamsters, Mesocricetus auratus, fed either 125 +/- 50 (group A), 300 +/- 50 (group B), or 500 +/- 50 (group C) metacercarial cysts of Echinostoma revolutum were infected 7-35 days postexposure. The mean number of worms in A, B, and C were 62, 96, and 212, respectively. Most of the worms in A were in the jejunum, but in C worms were about equally distributed in the duodenum, jejunum, and ileum, and some were in the cecum. The body area and wet and dry weights of worms from C were significantly less than that of A or B at 2, 4, and 5 wk postinfection. Echinostoma revolutum eggs were in the feces of 100% of the hamsters by days 12, 13, and 14 in A, B, and C, respectively.
