A set of anti-melanoma immunoconjugates were prepared which contained chlorin e6: antibody molar ratios of 18.9:1, 11.2:1, 6.8:1, and 1.7:1. All immunoconjugates retained antigen binding activity regardless of the chromophore:antibody substitution ratio that was attained. In contrast, the ground-state absorption spectra of the immunoconjugates showed features which appeared to be dependent on the chromophore:antibody molar ratio. In addition, the quantum yield of singlet oxygen generated by the conjugated chromophores was observed to be significantly less than that observed with the unbound dye. Time-resolved absorbance spectroscopy of the chromophore excited triplet state indicated that the loss of singlet oxygen quantum yield resulted from diminished chromophore triplet yield. Analysis of data obtained from in vitro photolysis of target melanoma cells, in combination with that obtained from the immunochemical and photochemical studies, indicates that the observed immunoconjugate phototoxicity can be reasonably quantitatively represented by (1) the ability of the immunoconjugate to bind SK-MEL-2 cell surface antigen, (2) the amount of chromophore localized at the target cells by immunoconjugate binding, (3) the delivered dose of light at 634 nm, and (4) the singlet oxygen quantum yield of the antibody-bound photosensitizer. Though these data argue strongly for photolysis by the cumulative dosage of singlet oxygen at the cell membrane, nonetheless, the concurrent photoinduced release of other cytotoxic agents should not be ruled out.  相似文献

The present experiment investigated whether observers' emotional and behavioral reactions to facially deformed patients could be substantially improved by surgical procedures conducted by well-trained specialists in an experienced multidisciplinary team. Also investigated was the hypothesis that emotional states mediate the effects of physical attractiveness and facial deformity on social interaction. Twenty patients between the ages of 3 months and 17 years were randomly selected from over 2000 patients' files of Kenneth E. Salyer of Dallas, Texas. Patient diagnoses included facial clefts, hypertelorism, Treacher Collins syndrome, and craniofacial dysostoses (Crouzon's and Apert's syndromes). Rigorously standardized photographs of patients taken before and after surgery were shown to 22 "naive" raters ranging in age from 18 to 54 years. Raters were asked to predict their emotional and behavioral responses to the patients. These ratings indicated that observers' behavioral reactions to facially deformed children and adolescents would be more positive following craniofacial surgery. Similarly, the ratings indicated that observers' emotional reactions to these patients would be more positive following surgery. The results are discussed in terms of current sociopsychologic theoretical models for the effects of attractiveness on social interaction. A new model is presented that implicates induced emotional states as a mediating process in explaining the effects of attractiveness and facial deformity on the quality of social interactions. Limitations of the current investigation and directions for future research are also discussed.  相似文献

Segments of briefly glycerinated muscle fibers from Rana pipiens were activated rapidly by a brief exposure to 2.5 mM free calcium followed by a solution containing calcium buffered with EGTA to produce the desired level of force. Steps to isotonic loads were made using a servomotor, usually 3-5 s after the onset of activation. The relative isotonic forces (P/P0) and velocities from contractions obtained under similar circumstances were grouped together and fitted with hyperbolic functions. Under the condition of 6 mM MgCl2 and 5 mM ATP, there was no significant difference in the relative force-velocity relations obtained at full activation compared with those obtained at partial activation when developed force was approximately 40% of its full value. Control experiments showed that a variety of factors did not alter either the relative force-velocity relations or the finding that partial activation did not change these properties. The factors investigated included the decline in force that occurs with each successive contraction of skinned fibers, the segment length (over a range of 1-3 mm), the sarcomere length (over a range of 1.9-2.2 microns), the magnesium ion concentration (26 microM and 1.4 mM were tested), the ATP concentration, the presence of free calcium, and the age of the preparation (up to 30 h). Attempts to repeat earlier experiments by others showing a dependence of shortening velocity on activation were unsuccessful because the low ionic strength used in those experiments caused the fibers to break after a few contractions. The main conclusion, that the shortening velocity is independent of the level of activation, is consistent with the hypothesis that the cross-bridges act independently and that activating calcium acts only as an all-or-none switch for individual cross-bridge attachment sites, and does not otherwise influence the kinetics of cross-bridge movement.  相似文献

Three strains of the aphid Myzus persicae are described which show constant differences in their ability to feed through parafilm membranes. Two of these strains fed fairly readily through the membranes, but the third fed very little. Of the two strains which fed through the membranes, one was more sensitive to the addition of neutral red and phosphamidon to the diet than the other. This sensitivity was shown by a reduction in liquid uptake which partly accounted for the high LC50 found for this strain.On the basis of the uptake methods used, sucrose was found to be phagostimulatory. An attempt at using the susceptibility of the aphids to phosphamidon as a measure of diet acceptability had a limited success.
Résumé Trois souches du puceron Myzus persicae sont décrites, qui montrent de constantes différences dans leur aptitude à se nourrir au travers de fines membranes plastiques transparentes (appelées parafilm membranes). Deux de ces souches se nourrissent assez aisément au travers des membranes, mais la troisième souche ne se nourrit que très peu. Des deux souches qui se nourrissent au travers des membranes, l'une se montra plus sensible que l'autre à l'addition à son régime de rouge neutre et de phosphamidon.Cette sensibilité fut montrée par une réduction du liquide absorbé qui, en partie, comptait pour le LC50 élevé trouvé pour cette souche.En se basant sur les méthodes d'absorption utilisées, on trouva que le saccharose était phagostimulant. Une tentative d'utiliser la sensibilité au phosphamidon des pucerons, comme mesure d'acceptation du régime, eut un succès limité.

We have developed a method to characterize fluid transport through the perialveolar interstitium using micropuncture techniques. In 10 experiments we established isolated perfused rat lung preparations. The lungs were initially isogravimetric at 10 cmH2O arterial pressure, 2 cmH2O venous pressure, and 5 cmH2O alveolar pressure. Perialveolar interstitial pressure was determined by micropuncture at alveolar junctions by use of the servo-null technique. Simultaneously a second micropipette was placed in an alveolar junction 20-40 microns away, and a bolus of albumin solution (3.5 g/100 ml) was injected. The resulting pressure transient was recorded for injection durations of 1 and 4 s in nonedematous lungs. The measurements were repeated after gross edema formation induced by elevated perfusion pressure. We model the interstitium as a homogeneous linearly poroelastic material and assume the initial pressure distribution due to the injection to be Gaussian. The pressure decay is inversely proportional to time, with time constant T, where T is a measure of the ratio of interstitial tissue stiffness to interstitial resistance to fluid flow. A linear regression was performed on the reciprocal of the pressure for the decaying portion of the transients to determine T. Comparing pressure transients in nonedematous and edematous lungs, we found that T was 4.0 +/- 1.4 and 1.4 +/- 0.6 s, respectively. We have shown that fluid transport through the pulmonary interstitium on a local level is sensitive to changes in interstitial stiffness and resistance. These results are consistent with the decreased stiffness and resistance in the perialveolar interstitium that accompany increased hydration.  相似文献

Sixteen hairy-tailed moles, Parascalops breweri, collected from the northeastern U.S.A. were examined for coccidian oocysts; all were infected with multiple species of coccidia and 3 genera were represented. Two cyclosporans, 2 eimerians, and 2 isosporans are described as new species. Sporulated oocysts of Cyclospora ashtabulensis n. sp. are subspheroid to ellipsoid, 18 X 14 (14-23 X 11-19) microns, and sporocysts are ovoid, 12 X 7 (8-14 X 5-9) microns; C. ashtabulensis was found in 7 of 16 (44%) moles. Sporulated oocysts of Cyclospora parascalopi n. sp. are spheroid, 17 X 14 (13-20 X 11-20) microns, and sporocysts are ovoid, 11 X 7 (8-14 X 5-8) microns; C. parascalopi was found in 8 of 16 (50%) moles. Sporulated oocysts of Eimeria aethiospora n. sp. are subspheroid to ellipsoid, 19 X 13 (15-24 X 10-16) microns, and sporocysts are ovoid, 11 X 6 (8-13 X 4-7) microns; E. aethiospora was found in 4 of 16 (25%) moles. Sporulated oocysts of Eimeria titthus n. sp. are subspheroid, 16 X 14 (13-19 X 11-17) microns, and sporocysts are ellipsoid, 11 X 6 (9-13 X 4-7) microns; E. titthus was found in 4 of 16 (25%) moles. Sporulated oocysts of Isospora ashtabulensis n. sp. are ellipsoid, 20 X 14 (16-24 X 10-18) microns, and sporocysts are ovoid, 10 X 7 (7-14 X 5-10) microns; I. ashtabulensis was found in 5 of 16 (31%) moles.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
