Background  

When a drug is applied on the skin surface, the concentration of the drug accumulated in the skin and the amount of the drug eliminated into the blood vessel depend on the value of a parameter, r. The values of r depend on the amount of diffusion and the normalized skin-capillary clearence. It is defined as the ratio of the steady-state drug concentration at the skin-capillary boundary to that at the skin-surface in one-dimensional models. The present paper studies the effect of the parameter values, when the region of contact of the skin with the drug, is a line segment on the skin surface.  相似文献

Inertial shear force is a surface force that is generated in centrifuges especially with attached samples on flat surfaces and plays a significant role in gravitational and space research. The magnitude of this force is proportional to the radius of the centrifuge and surface area of the sample compartment. In gravitational research we want to study the impact of weight onto a system. However, the force of inertial shear is perpendicular to the gravity vector, hence, results may be obscured or even misinterpreted by this artifact.  相似文献

The versatility of neuronal electrical activity is largely conditioned by the expression of different structural and functional classes of K+ channels. More than 80 genes encoding the main K+ channel alpha subunits have been identified in the human genome. Alternative splicing, heteromultimeric assembly, post-translational modification and interaction with auxiliary regulatory subunits further increase the molecular and functional diversity of K+ channels. Mammalian two-pore domain K+ channels (K2P) make up one class of K+ channels along with the inward rectifiers and the voltage- and/or calcium-dependent K+ channels. Each K2P channel subunit is made up of four transmembrane segments and two pore-forming (P) domains, which are arranged in tandem and function as either homo- or heterodimeric channels. This novel structural arrangement is associated with unusual gating properties including “background” or “leak” K+ channel activity, in which the channels show constitutive activity at rest. In this review article, we will focus on the lipid-sensitive mechano-gated K2P channel TREK-1 and will emphasize on the polymodal function of this “unconventional” K+ channel. The aim of thepresent study was to investigate the effect of chronic long-termclenbuterol treatment (1 mg/kg subcutaneously twice a day for 12 wk) ondiaphragm morphology and function in emphysematous (EH) and normalhamsters (NH). Clenbuterol increased body weight, diaphragm weight, andskeletal muscle weight in both EH and NH to a similarextent. In the diaphragm, clenbuterol significantly increased myosin heavy chain type I, IIa, and IIx muscle fiber cross-sectional areas by ~35-55% in both EH and NH. Thisresponse to clenbuterol treatment was not significantly differentbetween EH and NH diaphragm. In EH, twitch force(Pt), maximal tetanic force, andforce-frequency curve were significantly reduced compared with NH. InEH, clenbuterol increased Pt by~10%, restoring Pt to NH level.A similar improvement was observed in the force-frequency characteristics. Clenbuterol did not alter contractile properties inNH. In conclusion, long-term clenbuterol treatment resulted in anincreased size of all diaphragm muscle fiber types in both NH and EH.Clenbuterol completely abolished the reduced force generation inducedby emphysema.
