Summary The effect of selection intensity and population size on the response to selection for percent oil in the grain of maize (Zea mays L.) was evaluated in a replicated experiment over ten cycles of selection. An open-pollinated variety, Armel's Reid Yellow Dent, was divided into subpopulations of 6,10 and 50 plants. Selection proportions of 17% and 5% were imposed upon each subpopulation. Selection was based on the percentage of oil in individual kernels as determined by wide-line nuclear magnetic resonance spectroscopy. As expected, total response to selection increased with larger population sizes and selection intensities. The concave shape of the response curves suggested that an appreciable part of the genetic variance can be attributed to additive genes at high initial frequencies, dominance genes at low initial frequencies, or to the generation of negative linkage disequilibrium due to selection. The consistently greater loss of vigor experienced by the more intensely selected populations reflects the enhancement of inbreeding due to artificial selection, an effect that increases with the intensity of selection. The results indicate that combined selection, based on kernels and using within- and amongfamily information, will be more efficient than other conventional selection procedures, including the normal combined scheme where selection is based on plants.Deceased 相似文献

The formation of arthrospores in Mucor rouxii was studied by transmission and scanning electron microscopy and light microscopy. The arthrospores formed in a random manner in terminal and internal regions of the hyphae. The earliest appearance of the arthrospores was seen by scanning electron microscopy as compartments delineated by double ridges. These ridges probably corresponded to the site of septal wall formation. The elongated compartments varied considerably in size. As the arthrospores matured, they tended to separate as a result of a gradual change in the shape of the arthrospores to a nearly spherical form and also as the result of simultaneous degradation of the outermost cell wall layer. The mature arthrospores were surrounded by a complex cell wall consisting of at least three distinct layers in addition to the original hyphal cell wall. Crystal-like structures were seen in the cytoplasm of some of the arthrospores in addition to the usual organelles such as mitochondria, nuclei, and ribosomes. Septum formation by centripetal cell wall growth from the lateral hyphal wall was documented by transmission electron microscopy. However, evidence was also found which suggested that not all internal cell wall development in the fungal hyphae during arthrosporogenesis necessarily led to the formation of mature arthrospores. 相似文献

Biomass, litterfall, litter standing crop, and decomposition was studied in a native subtropical alluvial forest locally known as Selva Marginal (SM) and an exotic Ligustrum lucidum forest (LF) at the Reserva Integral de Punta Lara, Buenos Aires Province, 34°47S and 58°1W. The alluvial forest site was at the southern limit of distribution of subtropical forests in South America. The Ligustrum forest was invading disturbed areas. Total biomass was 147.7 Mg/ha (86% aboveground and 14% belowground) in the SM, and 71.4 Mg/ha (93% and 7%, respectively) in the LF. Litterfall was 10.3 Mg/ha·yr and 13.8 Mg/ha·yr respectively. Annual leaf decomposition rate was greater for Ligustrum (k=4.07) than for SM species (k=1.48). The mean residence time of aboveground biomass was 12 yr for the SM and 5 yr for the LF. The k1 values (litterfall/standing crop) were 1.9 and 2.0 for SM and LF respectively. The influence of coastal road and wall in the hydroperiod, native forested wetland ecosystem survival and exotic forest invasion is discussed. 相似文献

Biocatalytic transesterification of methylmethacrylate is possible in many different solvents. The reaction rate is readily controlled by variation in solvent physical properties. The reaction proceeds better in hydrophobic solvents, and activity can be restored in hydrophilic solvents by the addition of water. We have now demonstrated that supercritical carbon dioxide is not a good solvent for the reaction between 2-ethlhexanol and methylmethacrylate. It apperars that the supercritical carbon dioxide may either alter the pH of the microaqueous environment associated with the protein or reversibly form covalent complexes with free amine groups on the surface of the enzyme. Although supercritical carbon dioxide is a poor solvent for acrylate transesterification, many other supercritical fluids (ethane, ethylene, sulfur hexafluoride, and fluoroform) are better than most conventional solvents. In supercritical ethane it is possible to control the activity of the enzyme by changing pressure, and the enzyme appears to follow Michaelis-Menten Kinetics. We find that sulfur hexafluoride, the first anhydrous inorganic solvent in which biocatalytic activity has been reported, is a better solvent than any conventional or supercritical organic fluid tested. 相似文献

The regulation of amino acid transport in L6 muscle cells by amino acid deprivation was investigated. Proline uptake was Na+-dependent, saturable and concentrative, and was predominantly through system A. Proline uptake was inhibited by alanine, α-amino isobutyric acid (AIB), and by α-methylamino isobutyric acid, but not by lysine or valine. At 25°C, Km of proline uptake was 0.5 mM. Amino acid-deprivation resulted in a progressive increase in the rate of proline uptake, reaching up to 6-fold stimulation after 6 hours. The basal and stimulated transport were equally Na+-dependent, and both were inhibited by competition with the same amino acids. Kinetic analysis showed that Km decreased by a factor of 2.4 and Vmax increased 1.9-fold in deprived cells. Amino acid-deprivation did not stimulate amino acid uptake through systems other than system A. This suggests that the higher Km in proline-supplemented cells is not due to release of intracellular amino acids into unstirred layers surrounding the cells. The presence of amino acids which are substrates of system A (including AIB) during proline-deprivation, prevented stimulation of proline uptake, whereas those transported by systems Ly+ or L exclusively were ineffective. The stimulation of the transport-rate in deprived cells could be reversed by subsequent exposure to proline or other substrates of system A. L6 cells, deprived of proline for 6 hours, retained the stimulation of transport after detachment from the monolayers with trypsin. Uptake rates were comparable in suspended and attached cells in monolayer culture. Thus, amino acid-depreivation of L6 cells results in an adaptive increase in proline uptake, which is not due to unstirred layers but appears to be mediated by other mechanisms of selective transport regulation. 相似文献
