The ion binding properties of horse, bovine, and tuna cytochrome c (both oxidized and reduced) have been measured using a combination of ultrafiltration, neutron activation, and ion chromatography. The ions investigated were chloride, phosphate, and Tris-cacodylate. Ion chromatography and neutron activation analysis techniques were employed to determine the concentration of free anions. Binding constants are obtained from modified Scatchard plots (in the range of 10-2000 M-1). The redox potentials for cytochrome c at different ionic strengths, pH 7.0, have been determined. In this paper we report the ionic strength and ion binding effects on the redox properties of horse, bovine, and tuna cytochrome c. Potential versus ionic strength dependence for horse, bovine, and tuna cytochrome c from the experimental data were compared with a theoretical model.  相似文献

Post-partum acyclic beef cows received continuous long-term treatment with GnRH (200 or 400 ng/kg body wt/h) or the GnRH agonist buserelin (5.5 or 11 ng/kg body wt/h) using s.c. osmotic minipumps which were designed to remain active for 28 days. All treatments increased circulating LH concentrations whereas FSH remained unchanged. Ovulation and corpus luteum (CL) formation as judged by progesterone concentrations greater than or equal to 1 ng/ml occurred in 0/5 control, 4/5 200 ng GnRH, 4/4 400 ng GnRH, 4/5 5.5 ng buserelin and 3/5 11 ng buserelin cows. The outstanding features of the progesterone profiles were the synchrony, both within and across groups, in values greater than or equal to 1 ng/ml around Day 6, and the fact that most CL were short-lived (4-6 days). Only 3 cows, one each from the 400 ng GnRH, 5.5 ng buserelin and 11 ng buserelin groups, showed evidence of extended CL function. Cows failed to show a second ovulation which was anticipated around Day 10 and this could have been due to insufficient FSH to stimulate early follicular development, or the absence of an endogenously driven LH surge. The highest LH concentrations for the respective groups were observed on Days 2 and 6 and by Day 10 LH was declining, although concentrations did remain higher than in controls up to Day 20.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

It is increasingly recognized that exposure to electrical fields can reversibly increase the electrical permeability and conductivity of the cellular membrane. It is therefore worthwhile to determine under what circumstances the critical fields capable of affecting the cellular membrane can be reached. We have evaluated the field intensities E and the gradients of E2 in the neighborhood of the more common field electrode shapes, the sphere-sphere and cylinder-cylinder electrodes, and put these in terms of the values useful for the practical electro-fusion and dielectrophoresis (DEP) of cells. The calculations were performed using the charge-image technique. From this it was observed that gross errors (underestimations of up to 1000-fold) of the field would be made if neglect of the electrode polarization were made as in simple electrostatic calculation. The dielectric forces (in terms of (E)2) show interesting spatial character.The electrical breakdown of cellular membranes affects the exchange of information and materials between the cell and its environment, and can be used to fuse cells of the same type or of differing types. It is moreover of importance in handling cells during DEP while obtaining their spectral characteristics or in cell-sorting. It is expected that the results here will aid in the more proper use of electrical fields for such ends.  相似文献
