Electromyography (EMG) is commonly used to determine the electrical activity of skeletal muscle during contraction. To date, independent verification of the relationship between muscle use and EMG has not been provided. It has recently been shown that relaxation- (e.g., T2) weighted magnetic resonance images (MRI) of skeletal muscle demonstrate exercise-induced contrast enhancement that is graded with exercise intensity. This study was conducted to test the hypothesis that exercise-induced magnetic resonance (MR) contrast shifts would relate to EMG amplitude if both measures reflect muscle use during exercise. Both MRI and EMG data were collected for separate eccentric (ECC) and concentric (CON) exercise of increasing intensity to take advantage of the fact that the rate of increase and amplitude of EMG activity are markedly greater for CON muscle actions. Seven subjects 30 +/- 2 (SE) yr old performed five sets of 10 CON or ECC arm curls with each of four resistances representing 40, 60, 80, and 100% of their 10 repetition maximum for CON curls. There was 1.5 min between sets and 30 min between bouts (5 sets of 10 actions at each relative resistance). Multiple echo, transaxial T2-weighted MR images (1.5 T, TR/TE 2,000/30) were collected from a 7-cm region in the middle of the arm before exercise and immediately after each bout. Surface EMG signals were collected from both heads of the biceps brachii and the long head of the triceps brachii muscles. Hepatic metabolites and enzymes in the marine fish, scup or porgy (Stenotomus chrysops), were determined in freeze-clamped tissue taken either within a day of removing fish from their natural habitat or after scup were held in captivity for 6-8 months. The same determinations were made for liver from fed or 48 hr-starved rats (Mus norvegicus albinus). Compared with rat liver, both groups of fish had, per gram of liver, higher contents of AMP, inorganic phosphate, glucose, glucose-6-phosphate, malate, glutamate and NH4+. ATP was lower in fish liver, and ADP, lactate and pyruvate contents were similar in rats and fish. Fish held in captivity had significantly lower pyruvate, alpha-ketoglutarate, and cytosolic free NAD+/NADH and higher cytosolic free NADPH/NADP+. These decreases were similar to those seen when starved rats were compared with fed ones. In scup liver, glucose-6-phosphate dehydrogenase was 3-8 times, malic enzyme about 2 times, and alanine aminotransferase 2-4 times higher than those activities in rat liver. Those results and a higher cytosolic free NADPH/NADP+ are consistent with the liver being the major site of lipogenesis in fish. 相似文献

To ascertain the health risks that may be posed by the land application of sewage sludges, a scheme was devised to determine the types and numbers of pathogenic and potentially pathogenic bacteria present in sludges. A processing treatment was adapted to sludge to give a homogenate which yielded the greatest numbers of viable bacteria. Conventional methods were successful in enumerating Klebsiella, Staphylococcus, gram-negative enteric bacteria, and commonly used indicator organisms. Modifications of conventional methods improved the enumeration of Salmonella, Mycobacterium sp., fluorescent Pseudomonas sp., and Clostridium perfringens. However, Shigella methodology yielded only one isolate. Utilizing the proposed scheme, the population densities of these organisms were estimated in three domestic wastewater sludges. In light of these results, the potential impact of land application of sewage sludges is discussed. 相似文献

This study evaluated the sensitivity of mitochondrial respiratory control as a function of tissue oxidative capacity. The mitochondrial content of rat skeletal muscle was increased by exercise training or decreased by hypothyroidism. Muscles of the lower hindlimb were stimulated to tetanically contract in situ for 3 min at one of four frequencies to elicit a 30-fold range of oxygen consumption rates. Freeze-clamped sections of fast-twitch red gastrocnemius muscle were extracted and analyzed for metabolite levels. The sensitivity of respiratory control was examined for three models of cytosolic respiratory control (ADPf, ATP/ADPf, and ATP/(ADPf X Pi]; for each proposed model, sensitivity went up as mitochondrial content increased. Thus, a smaller change in cytosolic modulator (e.g., ADPf) is required as oxidative capacity increases. Increases in the sensitivity of cytosolic respiratory control resulted in lower flux through the near-equilibrium energy exchange reactions of creatine kinase and myokinase such that calculated free concentrations of ADP and AMP were less. Other energetically important reactions/pathways were also affected. Accumulation of lactate and the deamination of AMP to IMP were lower in tissues with higher mitochondrial content. In summary, changes in oxidative capacity directly influence the sensitivity of cytosolic respiratory control and this, in turn, has important consequences for maintenance of cellular energy balance. 相似文献

Summary Endophagous folivores, which are concealed inside leaf tissue for much of their life cycle, or which live externally but feed internally, should be more successful on heavily defended plants than ectophagous species. This is because endophages are more facile at feeding selectively and can manipulate tissue development to avoid physical and chemical defenses and to enhance nutrition. As a result, endophage-ectophage ratios should increase on more heavily defended hosts. This pattern will likely be strengthened by negative asymmetrical interactions with ectophages and pathogens, which may displace endophages from lightly defended hosts. The hypothesis predicts that endophages should be particularly abundant in resource-poor habitats which seem to support a preponderance of heavily defended hosts. Although data do not yet exist for a rigorous test, several observed cases where endophage distributions seem biased toward heavily defended hosts are at least consistent with the hypothesis. Plant defense levels may have little influence on the total number of herbivores associated with a host, but I suggest that guild structure can be profoundly altered. 相似文献

We characterized the surface antigen and mRNA expression for the CD11c (alpha X, p150) subunit of the human leukocyte adherence receptor family during hematopoietic cell differentiation. The CD11c subunit antigen and mRNA are constitutively expressed in undifferentiated HL-60 promyelocytic leukemia cells, and levels increase markedly with differentiation along the monocyte/macrophage pathway using phorbol myristate acetate. Human monocyte-derived macrophages and human alveolar macrophages express elevated levels of the CD11c subunit antigen and mRNA, indicating that the changes observed in vitro are present in vivo. Dot blot analysis of immature and mature lymphoid and myeloid cells and cell lines demonstrate equivalent levels of CD11c mRNA expression. We conclude that CD11c gene expression is selectively increased during hematopoietic cell differentiation along the monocyte/macrophage pathway. 相似文献
