The method of electron paramagnetic resonance with spin-labeled maleimide was used to study variation of the structure of Ca-ATPase of the sarcoplasmic reticulum (SR) in rabbit skeletal muscles under long-term hypercholesterolemia (HC). The rate of the maleimide spin label binding with Ca-ATPase of the SR was decreased in HC, which correlated with a lesser access of spin-labeled thiol groups for potassium ferricyanide and sodium ascorbate. HC led to a considerable reduction in the lability and to enhancement of hydrophobia of the spin-labeled fragment of the enzyme. It is concluded that the disordered function of the SR Ca-pump is a consequence of structural changes in the Ca-ATPase molecule in HC. 相似文献

Carbohydrate oxidation, the coupling of oxidation with phosphorylation and the level of nucleic acids in the brain tissue were studied as affected by variable magnetic fields of commercial frequency (50 Hz) depending on intensity and regime of generation. A repeated effect (15-seances) of the 32 kA/m continuous magnetic field decreases the content of glycogen, creatine phosphate, intensity of oxygen uptake and etherification of inorganic phosphate, glutamine and increases the DNA content. The 7.5 kA/m continuous magnetic field has no effect on the studied metabolic indices in the brain whereas the interrupted field (1s impulse duration, 2s interimpulse interval) of the same intensity and exposition lowers the amount of glycogen, glucose, creatine phosphate, the RNA and DNA exposition lowers the amount of glycogen, glucose, creatine phosphate, the RNA and DNA levels, activates the oxidation processes, intensifies their coupling with phosphorylation. 相似文献

Chemical and enzymatic syntheses of [5'-3H]adenosine, [5'-3H]guanosine, and [5'-3H]uridine have been developed. The reduction of beta-D-ribo-pentadialdo-1,4-furanosyl derivatives of corresponding bases is used in the chemical synthesis. The maximum molar activity of the labelled products was 220 TBk/mol in reactions with [3H]NaBH4 and 370-740 TBk/mol in reactions with gaseous tritium. The enzymatic synthesis was performed by the rebosylation of heterocyclic bases with nucleoside phosphorylase and [5'-3H]uridine as a ribosyl donor. Nucleoside phosphorylase is proposed to be used in the immobilized form to avoid the decrease of molar activity. Nucleosides labelled with tritium both in ribosyl and heterocyclic moieties were synthesised enzymatically. 相似文献

The recovery of a population of granulopoietic-monocytic colony-forming units of mouse bone marrow was only slightly inhibited during the first two weeks after exposure to a mixture of radiation (4 Gy) and heat (burn of 10 per cent of the skin). The method of isolation of chromatographically homogenic LDG-5 preparation from the rat liver was elaborated. The preparations obtained in the present study and LDG-5 preparations from skeletal muscles have a common component which is testified by results of the precipitation test in agar. In the rat liver there is an enzymatically non-active antigenic component, which, according to some of its physico-chemical parameters, is very similar with chemically modified forms of LDG-5 dimers. 相似文献

It was shown that intravenous injection of vasopressin in a dose of 5 pressor units per 1 kg of body weight led to changes in the ATP-ase activity of the heart and liver microsomes in one hour.
