An improved 13C-density-labeling method was used to study cell wall synthesis in rapidly expanding, slowly expanding and recently mature
internodes of Nitella translucens var axillaris (A.Br.) R.D.W. As cells matured, the rate of wall synthesis slowed and the deposition of cellulose microfibrils changed from
a predominantly transverse direction in the primary wall of rapidly expanding internodes to a helicoidal array in the secondary
wall of mature internodes. The secondary wall was characterized by relatively higher rates of cellulose synthesis and lower
rates of pectin synthesis than the primary wall. The synthesis of xyloglucan also decreased markedly at the transition to
secondary wall synthesis, while the synthesis of mannose-rich hemicellulose increased. Even though structural differences
were striking between the primary and secondary walls of Nitella, compositional differences between the two types of wall were quantitative rather than qualitative.

The release of high numbers of the eggparasitoid Trichogramma brassicae Bezd.(Hym. Trichogrammatidae) to control theEuropean corn borer (ECB), Ostrinianubilalis Hb. (Lep.: Crambidae) in maize hasraised concerns about potential negativeeffects on native natural enemies. The nativelarval parasitoid Lydella thompsoniHerting (Dipt.: Tachinidae) is the mostfrequent and important ECB parasitoid insouthern Switzerland and can achieve highparasitism rates. Its first generation emergestoo early to find ECB larvae and must rely onalternative hosts living in natural habitatsclose to maize fields. Inundative releases ofT. brassicae coincide with theoviposition period of the alternative hosts ofthe tachinid. T. brassicae moving out ofrelease fields may attack and diminish thepopulation of these hosts, creating abottleneck situation for L. thompsoni inthe subsequent spring. Laboratory hostspecificity tests showed that the tachinid'stwo most abundant spring hosts Archanarageminipuncta Haworth (1809) (Lep.: Noctuidae)and Chilo phragmitellus Hübner (1805)(Lep.: Crambidae) are successfully parasitisedby T. brassicae females in no-choicesituations. Our extensive field surveys,however, showed that the two tested springhosts escape parasitism since their eggs arewell hidden or not attractive. Multiple Molecular Forms of Acetylcholinesterase in the Nematode Caenorhabditis elegans 总被引:1,自引:3,他引:1
Abstract: Extracts of the nematode Caenorhabditis elegans contain five molecular forms of acetylcholinesterase (AChE) activity that can be separated by a combination of selective solubilization, velocity sedimentation, and ion-exchange chromatography. These are called form IA (5.2s), form IB (4.9.s), form II (6.7s), form III (11.3s), and form IV (13.0s). All except form III are present in significant amounts in rapidly prepared extracts and are probably native; form III is probably derived autolytically from form IV. Most of forms IA and IB can be solubilized by repeated extractions without detergent, whereas forms II, III, and IV require detergent for effective solubilization and may therefore be membrane-bound. High salt concentrations are not required for, and do not aid in, the solubilization of these forms. For all forms, molecular weights and frictional ratios have been estimated by a combination of gel permeation chromatography and velocity sedimentations in both H2O and D2O. The molecular weight estimates range from 83,000 to 357,000 and only form II shows extensive asymmetry. The separated forms have been characterized with respect to substrate affinity, substrate specificity, inhibitor sensitivity, thermal inactivation, and detergent sensitivity. Judging by these properties, C. elegans is like other invertebrates in that none of its cholinesterase forms resembles either the “true” or the “pseudo” cholinesterase of vertebrates. However, internal comparison of the C. elegans forms clearly distinguishes forms IA, III, and IV as a group from forms IB and II; the former are therefore designated “class A” forms, the latter “class B” forms. Genetic evidence indicates that separate genes control class A and class B forms, and that these two classes overlap functionally. Several factors, including kinetic properties, molecular asymmetry, molecular size, and solubility, all suggest that a molecular model of the multiple cholinesterase forms observed in vertebrate electric organs probably does not apply in C. elegans. Potential functional roles and subunit structures of the multiple AChE forms within each C. elegans class are discussed. 相似文献

The reproductive interactions of a specialized plant/pollinator system involving Asplundia uncinata Harling (Cyclanthaceae) and a derelomine flower weevil (Coleoptera: Curculionidae: Derelomini) were studied at La Selva,
Costa Rica. The inflorescences of A. uncinata exhibit a suite of cantharophilous characters including a precisely synchronized anthesis, protogyny, thermogenesis, olfactory
attraction of visitors via modified staminodes, and narrow interfloral entrances. The weevil pollinators use the inflorescences
for feeding, mating, and oviposition. The larvae are detritivorous and develop either in the detaching staminate flowers or,
at a more favorable rate, in the rotting infructescences. The rate of infructescence abortion was high and caused by low levels
of pollination. Manual pollination treatments yielded significantly higher seed counts than obtained under natural conditions,
and furthermore demonstrated the inflorescences' ability to reproduce via geitonogamy. In the longer term, the reproductive
benefits of maintaining low levels of pollination may shift away from the weevils and towards the plants via an increase in

I tested the utility of Seyfarth's (1977) model of rank-related attractiveness to explain the distribution of allogrooming behavior among captive bonobos (Pan paniscus). Adult female bonobos generally have high social status and may be dominant over males. As predicted by the model, I found that high-ranking adult females received most allogrooming within each of the four investigated groups. Among adult female-adult female dyads, however, allogrooming was not clearly associated with dominance rank. Contradictory to predictions of the model, the highest-ranking females were responsible for most displacements over allogrooming, and grooming competition is positively correlated with dominance rank. In the second part of this study, I investigated the social significance of allogrooming body site preferences. Bonobos direct significantly most allogrooming to the face of conspecifics, and high- and low-ranking individuals, as well as males and females, differ significantly in their preferences for certain allogrooming sites. Subordinates and males tended to avoid facial grooming and preferred the back and anogenital region, while high-ranking individuals and females directed most allogrooming to the face and head of grooming partners. Data from this study support the hypothesis that high-ranking females are the most attractive grooming partners within a female-centered bonobo society. The handicap principle (HP) stipulates that signal reliability can be maintained if signals are costly to produce. Yet empirical biologists are typically unable to directly measure evolutionary costs, and instead appeal to expenditure (the time, energy and resources associated with signaling behavior) as a sensible proxy. However the link between expenditure and cost is not always as straightforward as proponents of HP assume. We consider signaling interactions where whether the expenditure associated with signaling is converted into an evolutionary cost is in some sense dependent on the behavior of the intended recipient of the signal. We illustrate this with a few empirical examples and demonstrate that on this alternative expenditure to cost mapping the traditional predictions of HP no longer hold. Instead of full information transfer, a partially informative communication system like those uncovered by Wagner (Games 4(2):163–181, 2013) and Zollman et al. The Hfq protein mediates gene regulation by small RNAs (sRNAs) in about 50% of all bacteria. Depending on the species, phenotypic defects of an hfq mutant range from mild to severe. Here, we document that the purified Hfq protein of the plant pathogen and natural genetic engineer Agrobacterium tumefaciens binds to the previously described sRNA AbcR1 and its target mRNA atu2422, which codes for the substrate binding protein of an ABC transporter taking up proline and γ-aminobutyric acid (GABA). Several other ABC transporter components were overproduced in an hfq mutant compared to their levels in the parental strain, suggesting that Hfq plays a major role in controlling the uptake systems and metabolic versatility of A. tumefaciens. The hfq mutant showed delayed growth, altered cell morphology, and reduced motility. Although the DNA-transferring type IV secretion system was produced, tumor formation by the mutant strain was attenuated, demonstrating an important contribution of Hfq to plant transformation by A. tumefaciens. 相似文献

1. Animal search patterns reflect sensory perception ranges combined with memory and knowledge of the surrounding environment. 2. Random walks are used when the locations of resources are unknown, whereas directed walks should be optimal when the location of favourable habitats is known. However, directed walks have been quantified for very few species. 3. We re-analysed tracking data from three shark species to determine whether they were using directed walks, and if so, over which spatial scales. Fractal analysis was used to quantify how movement structure varied with spatial scale and determine whether the sharks were using patches. 4. Tiger sharks performed directed walks at large spatial scales (at least 6-8 km). Thresher sharks also showed directed movement (at scales of 400-1900 m), and adult threshers were able to orient at greater scales than juveniles, which may suggest that learning improves the ability to perform directed walks. Blacktip reef sharks had small home ranges, high site fidelity and showed no evidence of oriented movements at large scales. 5. There were inter- and intraspecific differences in path structure and patch size, although most individuals showed scale-dependent movements. Furthermore, some individuals of each species performed movements similar to a correlated random walk. 6. Sharks can perform directed walks over large spatial scales, with scales of movements reflecting site fidelity and home range size. In the cold-shock protein CspB from Bacillus subtilis three exposed Phe residues (F15, F17, and F27) are essential for its function in binding to single-stranded nucleic acids. Usually, the hydrophobic Phe side chains are buried in folded proteins. We asked here whether the exposition of the essential Phe residues could be a cause for the very low conformational stability of CspB. Urea-induced and heat-induced equilibrium unfolding transitions were measured for three mutants of CspB, where Phe 15, Phe 17, and Phe 27 were individually replaced by alanine. Unexpectedly, all three mutations strongly destabilized CspB. The aromatic side chains of Phe 15, Phe 17, and Phe 27 in the active site are thus important for both binding to nucleic acids and conformational stability. There is no compromise between function and stability in the active site. Model calculations indicate that, although they are partially exposed to solvent, all three Phe residues nevertheless lose accessible surface upon folding, and this should favor the native state. A different result is obtained with the F38A variant. Phe 38 is hyperexposed in native CspB, and its substitution by Ala is in fact stabilizing. Since steroids are only slightly soluble in the aqueous solutions in which enzymatic reactions take place, it is difficult to obtain high effective concentrations per unit reactor volume when enzymes are used to catalyze steroid reactions. In order to obtain high effective concentrations in the present work, we have used small particles of a hydrophobic polymer, poly (dimethyl siloxane), as a reservoir for the steroid substrate and product. The activity of a bacterial hydroxysteroid dehydrogenase in a buffer solution declines much more slowly in the presence of those polymer particles than in the presence of a comparable amount of butyl acetate or ethyl acetate, the organic solvents used as steroid reservoirs in previous work with steroid transforming enzymes. When another substrate of the hydroxysteroid dehydrogenase is loaded into the polymer particles and the particles are suspended in an aqueous solution containing the enzyme and its cofactor, more product is formed that when a similar solution is emulsified with butyl acetate. 相似文献
