BACKGROUND: The uptake of nitrate by phytoplankton is a central issue in biological oceanography due to its importance to primary production and vertical flux of biogenic carbon. Nitrate reductase catalyzes the first step of nitrate assimilation, the reduction of NO(3) to NO(2). A cytometric protocol to detect and quantify relative changes in nitrate reductase (NR) protein content of the marine centric diatom Skeletonema costatum is presented. METHODS: Immunolabeling of NR protein was achieved with polyclonal antibodies raised against S.costatum NR. Antisera specific to a NR protein subunit and to a NR polypeptide sequence were compared, and cytometric results of NR protein abundance were related to Western analyses. Changes in cellular NR abundance and activity were followed during an upwelling simulation experiment in which S. costatum was exposed to a shift from ammonia to nitrate as major nitrogen source. RESULTS: NR protein could be detected in NO(3)-grown cells and at extremely low levels hardly discernible by Western Blot densiometry in NH(4)-grown cells. The protocol allowed observation of early stages of NR induction during an upwelling simulation. NR abundance increased after the nutrient shift to reach a new physiological "steady-state" 96 hrs later. NR activity exhibited diel variation with maxima at mid-day. NR abundance as estimated by both flow cytometry and Western analysis exhibited a hyperbolic relationship to NR activity. This pattern suggests post-translational activation of NR protein. CONCLUSIONS: The presented protocol allows the differentiation of NH(4)- versus NO(3)-grown algae as well as the monitoring of early stages in the induction of nitrate assimilatory capacities. 相似文献

The contribution of light-independent carbon fixation (LICF) to the overall carbon gain and the seasonal patterns of maximum photosynthesis (Pmax and LICF were characterized in a broad taxonomic range of macrophytes from Monterey Bay, California. Pmax and LICF rates (nmol C.g filtered seawater?1.min?1) varied among species and taxonomic groups examined, and as a function of tissue type in the phaeophyte Laminaria setchellii Silva (Phaeophyceae). On average, Pmax values were higher in the Rhodophyta, whereas LICF rates were greater in the Phaeophyceae. LICF rates were generally less than 5% of Pmax in the marine macrophytes studied and, as a consequence, cannot fully compensate for respiratory carbon losses, which usually are greater than 10% of Pmax. All species studied possessed the highest Pmax and LICF rates when irradiance levels were highest and decreased during periods of low incident irradiance. Seasonal patterns of Pmax and LICF in most of the macrophytes from the stenothermal environment of Monterey Bay were strongly correlated with photosynthetic photon flux rather than seawater temperature. The concomitant decrease of LICF and Pmax rates in all species examined argues against LICF playing a major role in carbon acquisition under light-limiting conditions as suggested previously. Rather, the strong positive correlation of Pmax and LICF indicates the direct coupling of photosynthate (e.g. 3-phosphoglyceric acid) generation with production of substrates for LICF reactions. Our results also suggest that LICF might be a useful indicator of photosynthetic metabolism in marine macrophytes. 相似文献

The present study examined the effect of UV andphotosynthetically active radiation (PAR) onphotoinhibition and recovery in the Phaeophyte Macrocystis pyrifera, the Rhodophyte Chondruscrispus and the Chlorophyte Ulva lactuca underoutdoor culture conditions. There was an increase inphotoinhibition as a consequence of high exposure toUV-B radiation in M. pyrifera, however, highlevels of PAR accounted for most of thephotoinhibition in C. crispus and U.lactuca. Photodamage by UV-A, UV-B and PAR wascompletely repaired within 5 h and effective quantumyield reached pretreatment values in the three speciesstudied. Species were less susceptible tophotoinhibition after being incubated for 5 d underhigh exposures of natural irradiance suggesting aphotoadaptive process. The recovery of the effectivequantum yield was impaired by long exposure to highlevels of UV-B in C. crispus and UV-A, UV-B andPAR in M. pyrifera. This suggests a differentkind of damage by UV-A and PAR radiation, one to thephotosynthetic apparatus and another which affects therepair mechanism of some species. There was anincrease in UV-absorption ( 330 nm) in M. pyrifera and C. crispus within four days ofthe initiation of the experiment suggesting that thesespecies photoprotect their photosynthetic system whenexposed to elevated UV and PAR levels. 相似文献

Bahía San Quintín is a coastal lagoon with large Ulva biomass and upwelling influence. Previous observations suggest that Ulva has increased recently as a result of oyster cultivation. To evaluate the possible role of Ulva as a temporary nutrient sink, biomass and tissue C, N, and P were determined seasonally. Maximum biomass was present during spring and early summer (1,413–1,160 t (dry)) covering about 400 ha. However, the biomass decreased to 35 t (dry) by winter. The mean annual percentage of Ulva C, N, and P was 28%, 2%, and 0.14%, respectively. This study shows that Ulva can store up to 28 t of N and 2 t of P in Bahía San Quintín during spring–summer. Ulva may be displacing the seagrass Zostera marina subtidal beds. A partial removal of the seaweed would reduce the risk of eutrophication and the displacement of eelgrass beds. 相似文献

Red algae have been reported to be an important source of polysaccharides with potential immunomodulatory properties. The
objective of this study was to characterize the polysaccharides from Halopithys incurva and Hypnea spinella and to evaluate their effect on the synthesis of cytokines by murine cell line RAW 264.7 macrophages. Polysaccharides were
obtained by N-cetylpyridinium bromide precipitation and characterized by Fourier transform-infrared spectroscopy. Their effect
on the activity of RAW 264.7 macrophages was examined by quantification of tumor necrosis factor (TNF)-α, interleukin (IL)-6,
and nitric oxide (NO) production using enzyme-linked immunosorbent assays. The activation of the cytokine IL-6 and NO increased
linearly as the concentration of polysaccharides from H. incurva and Hy. spinella increased. In general, the activation of IL-6 and NO was tenfold greater when macrophages were exposed to polysaccharides
from H. incurva than when exposed to polysaccharides from Hy. spinella. In contrast, TNF-α concentration did not increase when macrophages were exposed to increasing polysaccharide levels. These
results indicate that polysaccharides are strong cytokine IL-6 inducers. 相似文献

Rhodoliths are bed forming, unattached coralline algae that incorporate large quantities of carbonate into their thalli in oceans worldwide. Lithophyllum margaritae and Neogoniolithon tricotomun are common Rhodoliths from the Gulf of California, however, little is known about their biogeographic distribution, and their ecological and physiological characteristics. As a consequence, the objective of this study was to define the temporal population dynamics, growth rates and photosynthetic capabilities of L. margaritae and N. tricotomun from the Gulf of California. The Gulf of California is characterized by annual temperature fluctuations that can exceed 20° C. Rhodolith beds exceeding one km in length were observed from the intertidal zone to a depth of approximately 20 m at the southern Gulf. Percent cover estimates for spherical unattached individuals of L. margaritae and N. tricotomun ranged from 10 to 100%. Growth rates were determined by staining the rhodoliths with Alizarine red and evaluating the linear increment of calcium carbonate after an incubation period in the field. Annual growth rates averaged 1.8 mm year−1 in L. margaritae and 3.4 mm year−1 in N. tricotomun , however, growth rates varied seasonally. The photosynthetic response of both species as a function of temperature was evaluated in the laboratory from 10° to 30° C. Maximum net photosynthetic rates peaked at approximately 25 to 30° C in both species, suggesting that maximum growth rates occur during the warmest months of the year. These results lead to a better understanding of the biogeographic ranges of subtidal coralline algae. 相似文献

BACKGROUND AND AIMS: Seagrasses are important facilitator species in shallow, soft-bottom marine environments worldwide and, in many places, are threatened by coastal development and eutrophication. One narrow-leaved species (Zostera marina) and one wide-leaved species, variously designated as Z. marina, Z. pacifica or Z. asiatica, are found off the California Channel Islands and adjacent California-Mexico coast. The aim of the present study was to confirm species identification genetically and to link patterns of genetic diversity, connectivity and hybridization among and within the populations with historical sea levels (Ice Age) or the contemporary environment. METHODS: Samples (n = 11-100) were collected from 28 sites off five California Channel Islands and six sites off the adjacent coast of southern California and Baja California, Mexico. DNA polymorphisms of the rDNA-ITS (internal transcribed spacer) cistron (nuclear), the matK intron (chloroplast) and nine microsatellite loci (nuclear) were examined in a population genetic and phylogeographic context. KEY RESULTS: All wide-leaved individuals were Z. pacifica, whereas narrow-leaved forms were Z. marina. Microsatellite genotypes were consistent with hybridization between the two species in three populations. The present distribution of Z. pacifica follows a glacial age land mass rather than present oceanographic regimes, but no link was observed between the present distribution of Z. marina and past or present environments. Island populations of Z. marina often were clonal and characterized by low genotypic diversity compared with populations along the Baja California coast. The high level of clonal connectivity around Santa Catalina Island indicated the importance of dispersal and subsequent re-establishment of vegetative fragments. CONCLUSIONS: The pristine environmental conditions of offshore islands do not guarantee maximum genetic diversity. Future restoration and transplantation efforts of seagrasses must recognize cryptic species and consider the degree of both genetic and genotypic variation in candidate donor populations. 相似文献

The marine flora of Panama remains poorly described to date, having been described from a few sporadic and species poor (<50 species) collections in the early half of the century and a couple of dedicated surveys in the latter half. With the exception of a few studies that have focused on particular genera or species, only a single marine floral survey has been published in the past three decades. In 1999 we collected marine algae from nearly 100 different collection sites along both coasts of Panama over a 10 month period of time. Over 1500 specimens have been curated, representing 250 Caribbean species and 117 Pacific species. This is an increase in algal diversity of approximately 100 and 50% for the Caribbean and Pacific flora, respectively, relative to Earle's compilation of 1972. We estimate that algal diversity in Panama may approach 450 species as the remaining 30% of the collection is identified and previous records are incorporated. Aspects of biogeography will be discussed in light of such extraordinary diversity over a relatively confined region. 相似文献

The Baja California peninsula represents a biogeographical boundary contributing to regional differentiation among populations of marine animals. We investigated the genetic characteristics of perennial and annual populations of the marine angiosperm, Zostera marina, along the Pacific coast of Baja California and in the Gulf of California, respectively. Populations of Z. marina from five coastal lagoons along the Pacific coast and four sites in the Gulf of California were studied using nine microsatellite loci. Analyses of variance revealed significant interregional differentiation, but no subregional differentiation. Significant spatial differentiation, assessed using θST values, was observed among all populations within the two regions. Z. marina populations along the Pacific coast are separated by more than 220 km and had the greatest θST (0.13–0.28) values, suggesting restricted gene flow. In contrast, lower but still significant genetic differentiation was observed among populations within the Gulf of California (θST = 0.04–0.18), even though populations are separated by more than 250 km. This suggests higher levels of gene flow among Gulf of California populations relative to Pacific coast populations. Direction of gene flow was predominantly southward among Pacific coast populations, whereas no dominant polarity in the Gulf of California populations was observed. The test for isolation by distance (IBD) showed a significant correlation between genetic and geographical distances in Gulf of California populations, but not in Pacific coast populations, perhaps because of shifts in currents during El Niño Southern Oscillation (ENSO) events along the Pacific coast. 相似文献
