The accumulation of polyols by Aspergillus niger (van Tiegh) strain S 1 and Penicillium chrysogenum (Thom) strain S 30 was followed during growth in media of different concentrations of NaCl. The major polyols found were glycerol, erythritol and mannitol. The total polyol pool increased in both organisms in response to raised salinity, and the proportion of glycerol and erythritol was markedly enhanced at high salinity.  相似文献

The Z‐molecule is a small, engineered IgG‐binding affinity protein derived from the immunoglobulin‐binding domain B of Staphylococcus aureus protein A. The Z‐domain consists of 58 amino acids forming a well‐defined antiparallel three‐helix structure. Two of the three helices are involved in ligand binding, whereas the third helix provides structural support to the three‐helix bundle. The small size and the stable three‐helix structure are two attractive properties comprised in the Z‐domain, but a further reduction in size of the protein is valuable for several reasons. Reduction in size facilitates synthetic production of any protein‐based molecule, which is beneficial from an economical viewpoint. In addition, a smaller protein is easier to manipulate through chemical modifications. By omitting the third stabilizing helix from the Z‐domain and joining the N‐ and C‐termini by a native peptide bond, the affinity protein obtains the advantageous properties of a smaller scaffold and in addition becomes resistant to exoproteases. We here demonstrate the synthesis and evaluation of a novel cyclic two‐helix Z‐domain. The molecule has retained affinity for its target protein, is resistant to heat treatment, and lacks both N‐ and C‐termini. The leucocyte-specific phosphatase CD45 is present in two main isoforms: the large CD45RA and the short CD45RO. We have recently shown that distinctive expression of these isoforms distinguishes natural killer (NK) populations. For example, co-expression of both isoforms identifies in vivo the anti tumor NK cells in hematological cancer patients. Here we show that low CD45 expression associates with less mature, CD56bright, NK cells. Most NK cells in healthy human donors are CD45RA+CD45RO-. The CD45RA-RO+ phenotype, CD45RO cells, is extremely uncommon in B or NK cells, in contrast to T cells. However, healthy donors possess CD45RAdimRO- (CD45RAdim cells), which show immature markers and are largely expanded in hematopoietic stem cell transplant patients. Blood borne cancer patients also have more CD45RAdim cells that carry several features of immature NK cells. However, and in opposition to their association to NK cell progenitors, they do not proliferate and show low expression of the transferrin receptor protein 1/CD71, suggesting low metabolic activity. Moreover, CD45RAdim cells properly respond to in vitro encounter with target cells by degranulating or gaining CD69 expression. In summary, they are quiescent NK cells, with low metabolic status that can, however, respond after encounter with target cells.  相似文献

The survival of motor neurons (SMN) protein is the product of the disease-determining gene of the neurodegenerative disorder spinal muscular atrophy (SMA). SMN is part of a stable multiprotein complex that is found in all metazoan cells in the cytoplasm and in nuclear Gems. The SMN complex contains, in addition to SMN, at least six other proteins, named Gemins2-7, and plays an essential role in the assembly of the spliceosomal small nuclear ribonucleoproteins (snRNPs). Through its binding to specific sequences in the snRNAs, the SMN complex surveys the correct identity of the target RNAs and facilitates snRNP assembly. Based on its ability to interact with several other protein and RNA components of cellular RNPs, it is likely that the SMN complex functions as an assemblyosome in the formation of diverse RNP particles, some of which may be of particular importance to the motor neuron. A detailed understanding of the cellular roles of the SMN complex may help the development of therapeutic strategies for this neurodegenerative disease.  相似文献

The egg laying pattern of the spotted stalk borer on sorghum and maize, Chilo partellus (Swinhoe) (Lepidoptera: Pyralidae) was studied during the first three nights of oviposition under laboratory conditions. More than 50% of both eggs and egg batches are laid during the first night of oviposition. Batch size decreases with time, whereas mean egg weight remains constant.Larger females lay both larger eggs and egg batches than smaller ones. They are also more fecund but it is only the second night of oviposition that contributes significantly to this difference.The reproductive effort decreases much with time. Allocation of reproductive reserves to early eggs seems more important than having eggs evenly distributed over time. Larger females use proportionally more resources to the production of late eggs. Large eggs are likely to contain more yolk which could be of importance for the ballooning behaviour of newly hatched larvae.
Résumé La ponte de C. partellus Swinhoe a été étudiée au laboratoire pendant les trois premières nuits de la ponte. Plus de 50% des ooplaques et des oefs ont été pondus pendant la première nuit. La taille des ooplaques diminue avec le temps, tandis que le poids moyen des oeufs reste constant.Les grosses femelles pondent des ooplaques et des oeufs plus grands que les petites. Elles sont aussi plus fécondes, mais c'est seulement à la seconde nuit de ponte qu'est due cette différence.Le reproduction diminue beaucoup avec le temps. L'affectation aux premiers oeufs des réserves utilizées pour la reprodution semble plus importante que l'émission d'oeufs régulièrement répartis dans le temps. Les grosses femelles utilisent proportionnellement plus de ressources à la production d'oeufstardifs. Les gros oeufs contiennent vraisemblablement plus de vitellus que pourrait être nécessaire pour le comportement aérostatique des chenilles néonates.
