The changes in skull shape that occur in the hybrid zone between the two European subspecies of the house mouse ( Mus musculus musculus and M. m. domesticus ) were studied by relative warp analyses. Landmarks observed on the ventral view of the skulls of 269 mice sampled in 18 localities in Denmark were analysed. Each population was also characterized by a hybrid index estimated from allozymic data. Although no clear pattern of within-population variability of shape could be established across the hybrid zone, the shape changes among the 18 populations were found to be correlated with the allozymic introgression but not with geographical location. Finally, the cline obtained for skull shape seems to be steeper than that corresponding to the average allozyme hybrid index which suggests that skull development could be slightly perturbed in hybrids.  相似文献

Summary The ability to recover male gametophyte derived plants, which is necessary to get transformed haploid plants, was verified for a hybrid of maize. Using the isolated microspore culture technique, a 9 × 10–5 plant regeneration frequency was obtained. Maize microspores were bombarded with tungsten particles using a PDS He/1000 apparatus. GUS expression in the microspores was maximum with 1.1 m diameter tungsten microprojectiles for 1100 and 1350 psi helium pressures at a 6 cm distance between the launch point and the target cells. Increasing the amount of DNA coated on the microparticles from 1.66 to 4 g DNA/mg of particles allowed a two-fold and four-fold increase of the GUS-expressing microspore frequency for 1100 and 1350 psi helium pressure bombardment, respectively. Optimal concentration of solidifying agent in the bombardment support culture medium was found to be 1%. Cell density ranging from 25000 microspores/bombardment to 100000 microspores/bombardment did not affect the frequency of GUS-expressing microspores. Using these optimal conditions, the maximum frequency of GUS-expressing microspores was found to be about 9 × 10–4, while maintaining an embryo formation frequency about 5 × 10–4.Abbreviations GUS -glucuronidase - PEG polyethylene glycol  相似文献

Cells and protoplasts isolated from three different tissues of sunflower hypocotyls and cultured either in liquid or agarose medium were compared in terms of their volume, DNA content, division potential and embryoid formation. Epidermal and external cortical cells differ from other tissue cells by their small size, their weak response to plasmolysis and their low DNA content (around 1C). They contribute only very weakly to the dividing protoplast population. In contrast, protoplasts from cortical and medullar cells both have similar division potential, reaching 50%. The nuclear DNA content of these two cell types, as well as their corresponding protoplasts, has a 2C value, taking root tip cells in G0 phase as standard. The culture conditions induce the same specific response in protoplasts isolated from both tissues: exclusively loose colony formation in liquid medium, and mainly production of embryoids in agarose medium.  相似文献

Summary Protoplast diameter, a physical parameter controlling the susceptibility to electropermeation, was observed to vary between different batches of Brassica napus and Brassica campestris protoplasts. In order to control the permeation field strength associated with the protoplast size, we investigated the mathematical relationship between permeant field strength and protoplast size by measuring electroloading of the calcein dye under conditions that did not alter protoplast survival. A roughly linear relationship was obtained when the permeant field strength was plotted against the reciprocal of the radius. The plot characteristics were found to be tissue specific (mesophyll, hypocotyl and cotyledon), species specific (turnip, rapeseed) and modulated by the pulse duration. A series of amine-alkyl derivatives of 5-arylidenehydantoin 3–21 was evaluated for their ability to improve antibiotic effectiveness in two strains of Gram-negative Enterobacter aerogenes: the reference strain (ATCC-13048) and the chloramphenicol-resistant derivative over-producing the AcrAB-TolC efflux pump (CM-64). Three antibiotics, chloramphenicol, nalidixic acid and sparfloxacin were used as markers of efflux pump activity. New compounds (5–16) were obtained within 3–4 step synthesis using Knoevenagel condensation, Mitsunobu reaction and microwave aided N-alkylation. Molecular modeling based structure–activity relationship (SAR) studies were performed. The most active compounds: (Z)-5-(4-(diethylamino)benzylidene)-3-(2-hydroxy-3-(4-(2-hydroxyethyl)piperazin-1-yl)propyl)imidazolidine-2,4-dione (14) and (Z)-5-(2,4-dimethoxybenzylidene)-3-(2-hydroxy-3-(isopropylamino)propyl)imidazolidine-2,4-dione (15) induced fourfold decrease of minimal inhibition concentration (MIC) of all tested antibiotics in the strain CM-64 overexpressing the AcrAB-TolC pump.  相似文献

The present study was conducted to identify the genetic factors controlling somatic embryogenesis in the sunflower. Two traits, the number of embryogenic explants per 40 explants plated (EE/40 E) and the number of embryos per 40 explants (E/40 E), were scored in 74 recombinant inbred lines (RILs) from a cross between ’PAC-2’ and ’RHA-266’. The experiment was designed as a randomized complete block with 76 genotypes (74 recombinant inbred lines and two parents) and three replications. Each replication consisted of three Erlenmeyer flasks with 40 epidermal layers (explants). Analyses of variance indicated the existence of highly significant differences among parental genotypes and their RILs. Heritabilities for the somatic embryogenesis traits studied, EE/40 E and E/40 E, were high (0.64 and 0.77 respectively) and the genetic gain, in percentage of the best parent for 10% of selected RILs, was significant. Four QTLs for EE/40 E (tee) and seven for E/40 E (ete) were detected using composite interval mapping and AFLP mapping. Sunflower (Helianthus annuus L.) is an economically important oil seed crop with an estimated genome size of 3000 Mb. We have constructed a bacterial artificial chromosome (BAC) library for sunflower, which represents an estimated 4- to 5-fold coverage of the genome. Nuclei isolated from young leaves were used as a source of high-molecular-weight DNA and a partial restriction endonuclease digestion protocol was used to cleave the DNA. A random sample of 60 clones indicated an average insert size of 80 kb, implying a 95% probability of recovering any specific sequence of interest. The library was screened with chloroplast DNA probes. Only 0.1% of the clones were identified to be of chloroplast origin, indicating that contamination with organellar DNAs is very low. The utility of the library was evaluated by screening for the presence of genes for putative transmembrane receptors sharing epidermal growth factor (EGF) and integrin-like domains. First, a homologous sunflower EST (HaELP1) was obtained by degenerate RT-PCR cloning, using Arabidopsis thaliana genes (AtELP) as a source of consensus sequences. Three different BACs yielded positive hybridization signals when HaELP1 was used as a probe. BAC subcloning and sequencing demonstrated the presence of two different loci putatively homologous to genes for transmembrane proteins with EGF- and integrin-like domains from sunflower. This work demonstrates the suitability of the library for homology map-based cloning of sunflower genes and physical mapping of the sunflower genome.  相似文献

The cytoskeleton is involved in major developmental events in plant cell growth and differentiation. Nucleation events play a key role in the dynamic and organization of the microtubule (Mt) cytoskeleton. Among many proteins involved in Mt nucleation, γ -tubulin has been identified as an essential component of the Mt organizing centers (MTOC). In protoplasts, somatic embryogenesis induction has been correlated with remodeling of Mt cytoskeleton. We have investigated the specific developmental expression of γ -tubulin in Helianthus annuus . Two γ -tubulin isoforms have been detected by immunoblotting, with bands at 52 and 58 kDa. The larger γ -tubulin (58 kDa) is present in all the sunflower tissues tested and is associated with the nucleus. The smaller γ -tubulin (52 kDa), differing from the former at the carboxy-terminal end, is only present in meristematic and dedifferentiated cells and is not bound to the nucleus. This first demonstration of the presence of two γ -tubulins in plant cells is discussed in terms of distinct roles in the nucleation and organization of Mts.  相似文献

The organization of actin microfilaments was studied by immunofluorescence in protoplasts isolated from sunflower hypocotyls and cultured in an agarose matrix. Removal of the cell wall completely disrupted the actin cytoskeleton, which became progressively reorganized into cortical microfilament arrays and actin cables during protoplast culture. Treatment of protoplasts with arginine-glycine-aspartic acid (Arg-Gly-Asp) motif-containing peptides, to inhibit putative cell contacts with the agarose matrix, strongly affected this repair process: microfilament elongation and cable formation were inhibited and the connectivity between the cortical network and the perinuclear basket was lost. Furthermore, embryoid formation induced by agarose embedding was reduced. Similar effects were observed with a short treatment with latrunculin B, known to disrupt actin microfilaments. These results indicate that the actin network is involved in the signalling process that leads to polarity acquisition and embryoid determination in agarose-embedded protoplasts.  相似文献

Vacuoles were isolated from Acer pseudoplatanus cell suspension culture using a one-step procedure involving the lysis of the protoplast plasmalemma through a gradient of Ficoll containing DEAE-Dextran. The vacuole suspensions were slightly contaminated by other organelles (less than 5%) and the isolated vacuoles readily accumulated neutral red. Since α-mannosidase was located exclusively in the vacuoles it was used as a convenient marker. It was shown that the number of vacuoles per protoplast decreased as the cell aged. Studies on the biochemical composition of the isolated vacuoles indicated that amino acids, organic acids and protein contents varied with the cell culture cycle, emphasizing the dynamic status of the vacuolar system in cell suspension cultures of Acer pseudoplatanus.  相似文献
