The temperature dependence of the circular dichroism of the DNA-ethidium bromide complex at elevated temperatures provides evidence that the optical activity of the complex near 307 nm originates from interactions between intercalated dye molecules while the optical activity near 515 nm results from singly intercalated ethidium bromide molecules. The behavior of the circular dichroism of the complex at elevated temperatures also explains the higher ellipticities near 307 nm which characterize complexes formed between ethidium bromide and denaturated DNA. Finally the circular dichroism data indicate that the melting of the complex takes place in a stepwise manner with some DNA regions, probably AT-rich regions, dissociating first. The implications of these findings regarding the inhibiting effect of ethidium bromide on the function of DNA polymerase are examined.  相似文献

Abstract. The phylogenetic relationships among the “archaeogastropod” clades Patellogastropoda, Vetigastropoda, Neritimorpha, and Neomphalina are uncertain; the phylogenetic placement of these clades varies across different analyses, and particularly among those using morphological characteristics and those relying on molecular data. This study explores the relationships among these groups using a combined analysis with seven molecular loci (18S rRNA, 28S rRNA, histone H3, 16S rRNA, cytochrome c oxidase subunit I [COI], myosin heavy-chain type II, and elongation factor-1α [EF-1α]) sequenced for 31 ingroup taxa and eight outgroup taxa. The deep evolutionary splits among these groups have made resolution of stable relationships difficult, and so EF-1α and myosin are used in an attempt to re-examine these ancient radiation events. Three phylogenetic analyses were performed utilizing all seven genes: a single-step direct optimization analysis using parsimony, and two-step approaches using parsimony and maximum likelihood. A single-step direct optimization parsimony analysis was also performed using only five molecular loci (18S rRNA, 28S rRNA, histone H3, 16S rRNA, and COI) in order to determine the utility of EF-1α and myosin in resolving deep relationships. In the likelihood and POY optimal phylogenetic analyses, Gastropoda, Caenogastropoda, Neritimorpha, Neomphalina, and Patellogastropoda were monophyletic. Additionally, Neomphalina and Pleurotomariidae fell outside the remaining vetigastropods, indicating the need for further investigation into the relationship of these groups with other gastropods.  相似文献

The inhibition of the RNA polymerase-catalyzed synthesis of RNA by daunomycin was examined. Saturation binding of daunomycin to the template leads, as expected, to complete inhibition of RNA synthesis as a result of daunomycin interference with enzyme-template interactions. However at concentrations of the inhibitor below saturation formation of the enzyme-template complex remains remarkably undisturbed, while both the transformation of this complex to an elongating complex and the elongation of the nacsent RNA chains are substantially inhibited. Clearly, daunomycin interferes with a number of different substeps of RNA synthesis and inhibits the synthesis by different mechanisms depending on the amount of inhibitor bound to the template. Elucidation of the mechanism of inhibition at low daunomycin concentrations may be a prerequisite for a better understanding of the mechanism of the pharmacological action of the drug.  相似文献

We use agent-based modeling to study osotua, a gift giving system used by the Maasai of East Africa. Osotua’s literal meaning is “umbilical cord,” but it is used metaphorically to refer to a specific type of gift-giving relationship. Osotua relationships are characterized by respect, responsibility and restraint. Osotua partners ask each other for help only if they are in need and provide help only when asked and only if they are able. We hypothesize that under the ecologically volatile conditions in which Maasai pastoralists have traditionally lived, such a system is particularly suited to risk pooling. Here we explore whether osotua increases the viability of herds by comparing herd survivorship and stability under osotua rules to a) no exchange and b) probabilistic rules for requesting and giving livestock. Results from this model suggest that this gift-giving system can dramatically increase herd longevity through a limited pooling of risk.  相似文献

Models of the evolution of cooperation suggest that an important characteristic of successful strategies is the ability to respond contingently to the social environment. A number of mechanisms by which this can be accomplished have been suggested, some of which require relatively complex information processing systems. This research explores relaxing the requirements on information processing while preserving the evolvability of a cooperative strategy. The agent-based computer simulations reported here show that 'Walk Away,' a behavioral rule of extremely limited complexity (move after partner defects), can outperform more complex strategies under a number of conditions. Previous simulations of exit strategies have not examined the effect of implicit and explicit movement costs, different error rates, or the simultaneous presence of TFT and PAVLOV. The simulations reported here establish that the Walk Away strategy resists invasion and can invade a population of defectors at a lower initial frequency than any other strategy. The Walk Away strategy was successful, despite its simplicity, because it exploited aspects of the physical and social environment.  相似文献

Cancer therapy selects for cancer cells resistant to treatment, a process that is fundamentally evolutionary. To what extent, however, is the evolutionary perspective employed in research on therapeutic resistance and relapse? We analyzed 6,228 papers on therapeutic resistance and/or relapse in cancers and found that the use of evolution terms in abstracts has remained at about 1% since the 1980s. However, detailed coding of 22 recent papers revealed a higher proportion of papers using evolutionary methods or evolutionary theory, although this number is still less than 10%. Despite the fact that relapse and therapeutic resistance is essentially an evolutionary process, it appears that this framework has not permeated research. This represents an unrealized opportunity for advances in research on therapeutic resistance.  相似文献
