This study evaluated one semi-direct and three indirect methods for estimating leaf area index (LAI) by comparing these estimates with direct estimates derived from litter collection. The semi-direct method uses a thin metallic needle to count a number of contacts across fresh litter layers. One indirect method is based on the penetration of diffuse global radiation measured over the course of a day. The second indirect method uses the LAI-2000 plant canopy analyser (PCA) which measures diffuse light penetration from five different sky sectors simultaneously. The third indirect method uses the Demon portable light sensor to measure the penetration of direct beam sunlight at different zenith angles over the course of half a day. The Poisson model of gap frequency was applied to estimate plant area index (PAI) from observed transmittances using the second and third methods. Litter collection from 11 temperate decidous forests gave values of LAI ranging from 1.7 to 7.5. Estimates based on the needle method showed a significant linear relationship with LAI values obtained from litter collections but were systematically lower (by 6–37%). PAI estimates using all three indirect techniques (fixed light sensor system, LAI-2000 and Demon) showed a strong linear relationship with LAI derived from litter collection. Differences, averaged over all forest stands, between PAI estimates from each of the three indirect methods and LAI from litter collections were below 2%. If we consider that LAI=PAI–WAI (wood area index) then, all three indirect methods underestimated LAI by an additional factor close to the value of WAI. One reason could be a local clumping of architectural canopy components: in particular, the spatial dispositions of branchlets and leaves are not independent, leading to a non-random relationship between the distributions of these two canopy components.  相似文献

Focal sampling is the most accurate method for measuring primate activity budgets but is sometimes impractical. An alternative is scan sampling, in which the behaviour of the group is recorded at regular intervals. The simplest technique is to record whether at least 1 animal is engaged in the behaviour of interest. By direct comparison with focal data collected simultaneously on the same population, we assess the validity of this simple group level sampling method for studying chimpanzee (Pan troglodytes schweinfurthii) feeding behaviour. In a 13-month study at Kanyawara, Kibale National Park, Uganda, group level scan sampling provided statistically similar measures of broad diet composition to those produced by focal data, despite considerable seasonal variation. Monthly means of the percentage of time spent consuming non-fig fruit calculated from group level scan sampling were highly correlated with those from focal sampling. This validates previous methodology used to identify periods of high energy availability. However, group level scans tended to overestimate the percentage of observation time spent feeding, particularly for adult males. We conclude that this method of group level scan sampling provides valuable data for characterizing broad diet choice in chimpanzees and other species, but may be of limited use for estimating individual feeding time.  相似文献

Repeated measures of periosteal and endosteal widths of the left second metacarpal from two samples of radiographs were made independently with either a dial caliper or an electronic digitizer connected to a microcomputer. In addition, periosteal and endosteal bone widths of a common sample of radiographs were measured with dial calipers and the electronic digitizer. Dial calipers and an electronic digitizer are both very reliable, but intraobserver errors produced with the use of the digitizer are significantly less than those produced with the use of dial calipers. However, measurements made with the electronic digitizer are systematically larger than corresponding widths measured with dial calipers, but this difference is very small.  相似文献

Individual organisms interact directly through behavior, and indirectly through resource consumption and environment modification. The effects of different kinds of interactions on individual growth and reproduction will differ. Freshwater snails may interact directly for food resources and indirectly through substances dissolved in water. I separated the effects of the direct behavioral interaction and indirect interaction through waters using laboratory experiments with freshwater snails Physa acuta. Behavioral direct interaction have negative effects on the growth, but indirect interaction through water environments has positive effect on the growth. The importance of distinction of different kinds of interactions were discussed.  相似文献

Summary Degradation of 10 organic chemicals by pre-acclimated microorganisms in BOD dilution water was determined directly by UV spectrophotometry and indirectly by a modified BOD method. Residual chemical concentrations were periodically measured and pseudo-first-order biodegradation rate constants (k 1) were calculated. Thek 1 spectrophotometry values ranged from 0.006/h to 0.077/h andk 1-BOD values from 0.002/h to 0.043/h for 1-methylnaphthalene and indole, respectively. The ratios ofk spectrophotometry to k1-BOD were between 1.5 for salicylic acid and 3.0 for 1-methylnaphthalene with a mean of 2.7. A significant (=0.001) linear correlation (r 2=0.854,F=46.630) existed between the two sets of rate constants. Results from this study suggest that the modified BOD method may be used to estimate chemical biodegradation rates in synthetic media.  相似文献

Carbon dioxide and oxygen exchange procedures for measuring community metabolism (two open stream methods and three chamber methods) were compared on the same reach of a third-order stream. Open stream methods were complicated by high diffusion rates and yielded net community primary productivity estimates lower than those obtained with chamber methods. Chamber methods yielded variable productivity and respiration data. However, when normalized for chlorophyll a, productivity estimates from the chamber methods were within an expected range for the system. Balances of photosynthesis and respiration from the chamber methods were similar between methods and indicated that autotrophic or heterotrophic processes could dominate the system. Considerations in applying the various procedures are discussed.  相似文献

After herbivore attack, plants launch a suite of direct and indirect defense responses that must be coordinated if plants are to realize a fitness benefit from these responses. Here we characterize the volatile emissions in the native tobacco plant, Nicotiana attenuata Torr. ex Wats., that are elicited by tobacco hornworm (Manduca sexta L.) attack and are known to function as attractants for parasitoids. To provide the first ecophysiological comparison of examples of both types of defense in the same species, we characterize the elicitation and signaling mechanisms, the resources required, and the potential costs and benefits of the volatile release and compare these traits with those of the well-described induced direct defense in this species, nicotine production. The release of (E)-β-ocimene, cis-α-bergamotene and linalool is dramatically induced within 24 h by application of methyl jasmonate (MeJA), caterpillar feeding, and the treatment of mechanical wounds with larval oral secretions (OS), but not by mechanical damage alone. Plants from different geographic locations produce volatile blends that differ in composition. The most consistently released component from all genotypes, cis-α-berga-motene, is positively related to the amount of MeJA and the level of wounding if OS are applied to the wounds. The volatile release is strongly light dependent, dropping to undetectable quantities during dark periods, even when temperatures are elevated to match those of the light period. Inhibitors of wound-induced jasmonate accumulation (salicylates and auxins), which are known to inhibit wound-induced nicotine production, do not inhibit the release of volatiles. By individually inducing different leaf positions with OS and, on other plants, excising them after induction, we demonstrate that the emission is largely a systemic, whole-plant response, which is maximally triggered when the second fully expanded leaf is induced. We conclude that while both are whole-plant, systemic responses that utilize recently acquired resources for their production and are activated by the jasmonate cascade, the elicitation of the volatile release exhibits greater tissue sensitivity and utilizes additional signaling components than does nicotine production. In contrast to the large investment of fitness-limiting resources required for induced nicotine production or the resources used in benzyl acetone release from flowers for pollinator attraction, the resource requirements for the volatile release are minor. Full pyrosequencing runs of both direct-extracted (high yield, low DNA size) and indirect-extracted DNA (low yield, high DNA size) from the same prairie soil show that the sequence distribution of the majority of the metabolic functions and species detected were statistically similar. Although some microbial functions differed at the 95% confidence interval in bootstrap analyses, the overall functional diversity was the same.  相似文献

Four different measures (PETCO2, PACO2, PADCO2, and PJCO2) for indirectly estimating arterial PCO2 (PaCO2) from respired gas at the mouth have been investigated. PETCO2 was the end-tidal PCO2. PACO2 was calculated using a reconstruction of the alveolar oscillation of PCO2 obtained from the end-tidal "plateau" in PCO2. PADCO2 was calculated as for PACO2 except that the effects of dead space were incorporated. PJCO2 was calculated from an empirical relationship involving PETCO2 and tidal volume. Six subjects were studied at rest and during cycle ergometry at 50 and 100 W while breathing a variety of gas mixtures. Arterial samples were drawn for determination of true PaCO2. The differences for each method between estimated and true PaCO2 at rest and at 50 and 100 W were as follows: PETCO2, -1.35 +/- 2.64, 1.67 +/- 2.31, and 2.67 +/- 2.02 (SD) Torr; PaCO2, -2.15 +/- 2.73, -0.80 +/- 2.18, and -0.35 +/- 2.31 (SD) Torr; PADCO2, -1.55 +/- 2.54, 0.25 +/- 2.16, and 0.63 +/- 2.26 (SD) Torr; and PJCO2, -1.41 +/- 2.30, 0.12 +/- 1.79, and 0.08 +/- 1.96 (SD) Torr. It is concluded that, at rest, all methods significantly underestimate true PaCO2 and during exercise PETCO2 significantly overestimates PaCO2, but no bias was detected for any of the other methods.  相似文献

The comparative ability of different methods to assess virulence of Listeria species was investigated in ten Listeria strains. All strains were initially subjected to pulsed-field gel electrophoresis analysis to determine their relatedness. Virulence characteristics were subsequently tested for by (i) determining the presence of six virulence genes by polymerase chain reaction; (ii) testing for the production of listeriolysin O, phosphatidylcholine phospholipase C, and phosphatidylinositol-specific phospholipase C; (iii) investigating the hydrophobicity of the strains; (iv) determining the strains ability to attach to, enter, and replicate within the Caco-2 cells. Variations in most of the virulence characteristics were obvious across the strains for the range of tests performed. A wide range of anomalous results among methods were apparent. In particular, the presence of virulence genes was found to be unrelated to the production of virulence-associated proteins in vitro, while virulence protein production and hydrophobicity in Listeria monocytogenes were found to be unrelated or marginally related, respectively, to the ability to invade the Caco-2 cell line. It was concluded that the methods investigated were unable to consistently and unequivocally measure the differences in the virulence properties of the strains.  相似文献

Three widely used methods for measuring total soil CO2 evolution are evaluated, including the dynamic CO2 absorption method, the static CO2 absorption method and the closed chamber method. The study covers laboratory experiments. numerical experiments with a simulation model and field measurements. The results are used to perform an error analysis. The aim of this error analysis is to indicate the impact of each method on the CO2 dynamics during the measurement, and to select the most suitable method for frequent field usage.Laboratory experiments and simulation results show that the dynamic CO2 absorption method has the potential to absorb all CO2 evolving at the soil surface. The results also prove that the method has only a minor impact on the CO2 concentration-depth gradient and the CO2 efflux. The static CO2 absorption method underestimates the soil CO2 evolution, because the absorption velocity is too low, due to slow diffusion processes. Measurements with the closed-chamber method are based on an increasing concentration with time under a closed cover. However, the accumulation of gas alters the concentration gradient in the soil profile and thus causes a rapidly decreasing efflux during the measurement. A commonly used mathematical procedure, which corrects for the altered concentration gradient, does not yield the exact surface efflux, because the effect of increasing storage in the soil profile is not incorporated. Field measurements of CO2 evolution, using the closed-chamber method and the dynamic CO2 absorption method confirm the trends that have been predicted by the simulation model. The results of this study indicate that the dynamic CO2 absorption method is accurate. As it is cheap and simple, it is suitable for the study of temporal and spatial dynamics of CO2 evolution from the soil.  相似文献

This article outlines a number of methods for the determination of inhibitory and bactericidal activity against H. pylori. Direct methods rely on the ability of bacteria to divide and multiply and ultimately form visible colonies after subjection to antibiotic treatment. Indirect methods rely on the measurement of metabolic activity as a viability marker and are much more rapid, especially taking into account the slow growth and fastidious nature of the organism. Inhibitory concentration measurement does not indicate the bactericidal ability of a drug; inhibition of growth does not necessarily correlate with cell death. Theoretical generation of viable but nonculturable bacteria could bring in to question the validity of direct measurements based on the colony forming ability of an organism posttreatment.  相似文献

This paper describes a new method for the measurement of cuticularconductance (go;) using a leaf disc sealed in a specially-designedenvelope. Conductances for astomatous (adaxial) and stomatous(abaxial) surfaces of beech {Fagus sylvatica L.) were determinedfrom measurements of water flux. Leaf discs were punched outfrom attached leaves and placed inside individual envelopesthat provided a water supply. Water flux from an exposed epidermalsurface of the leaf discs was measured gravimetrically. Allmeasurements were made under darkness. Conductance of the adaxialsurface was referred to as gc, whereas conductance of the abaxialsurface was considered as a minimum leaf surface conductance()- The main advantage of this method is that it enables measurement of gc and from leaf samples with intact cuticles and a highrelative water content [RWC) for periods of up to 12 d. Conductancesof leaf discs in envelopes were compared with those of wholeleaves and leaf discs without envelopes. The membrane potential, ΔΨ, of various yeasts estimated from the distribution of tetraphenylphosphonium cations ranged from ?50 to ?120 mV, depending on species, incubation conditions and technique of measurement. A re-evaluation was made of the efficiency with which some of the commonly used assays would detect intestinal antibodies. The data indicate that the most sensitive assays for the detection of intestinal antibodies are the baby mouse protection test and the radioimmunoassay. Experiments were carried out to compare the direct approach for estimating crop N uptake from 15N labelled organic inputs, to two indirect approaches, 15N isotope dilution and A value. In the first experiment soils received 25, 50, 75, or 100 mg N kg soil−1 in the form of Casuarina equisitifolia residues in addition to ammonium sulphate fertiliser, to give a total of 100 mg N kg soil−1 added. This was a cross labelling design, thus two matching sets of treatments, were set up, identical in all but the position of the 15N label. Maize (Zea mays L.) plants were grown in the soils amended with residues for 11 weeks and N derived from residues (Ndfr) estimated using the A-value or the direct approach. The A-value approach appeared to significantly overestimate %Ndfr compared to the direct method. In the second experiment contrasting residues were added to soil, fababean (Vicia faba L. var. minor), alfalfa (Medicago sativa L.), soyabean fixing, (Glycine max (L.) Merrill), soyabean non-fixing, barley (Hordeum vulgare L.) and maize. This was also cross-labelling design, labelled and unlabelled residues were used. Maize plants were grown in these soils for 11 weeks and %Ndfr in the maize plants estimated using 15 N isotope dilution and the direct approach. The 15 N isotope dilution approach also overestimated %Ndfr compared to the direct method in this experiment. Pool substitution appeared to be responsible for the discrepancy between the direct and indirect techniques. It was concluded that 15N isotope dilution and A-value approaches as used in these experiments (i.e where residues and 15N label are added simultaneously) were not appropriate techniques for estimating N derived from organic residues in soils.
