Summary Prolactin (PRL) cells were studied electron-microscopically and morphometrically in the annual cyprinodont fish, Cynolebias whitei during its life cycle. In prehatching larvae, PRL cells possessed small secretory granules, giant mitochondria and a well-developed Golgi apparatus. During hatching, no changes were observed in the volume density of the secretory granules, indicating that no increased release of PRL occurs at hatching. A significant change in the composition of PRL cells, i.e., the volume densities per cytoplasm volume of the different organelles, occurred between one day and one week of age. Thereafter, only minor differences were observed between age groups, indicating that no major changes occur in PRL cell activity during the lifespan of C. whitei. However, the volume density per cell volume of the nucleus decreased steadily with age during the lifespan. A comparison of the PRL cells in young and adult fish reared in fresh water (FW) with siblings reared from hatching in diluted sea water (1/3 SW) did not reveal any differences with respect to the volume densities of the organelles, including the secretory granules. However, significant differences were observed with respect to the diameter, electron-dense content and affinity to anti- PRL serum of the secretory granules. These differences indicate that, despite the similar volumetric composition of the PRL cells, their secretory granules contain a substantially higher concentration of PRL in FW-reared fish than in 1/3 SW-reared fish.  相似文献

Summary A combined immunocytochemical and morphometric study on the development of the prolactin (PRL) cells of the annual cyprinodont Cynolebias whitei, transferred as newly hatched larvae to water with different salinities and/or Ca2+-concentrations, was carried out. The percentage of the pituitary volume occupied by PRL cells and the affinity of PRL cells for immunocytochemical staining were used as criteria for their activity. Exposure of the larvae for one day to salt water (260 mOsm/kg) led to a significant reduction in the pituitary volume occupied by PRL cells, indicating an osmoregulatory function of PRL shortly after hatching. In fish reared in diluted artificial seawater (70 and 260 mOsm/kg) or Na+-enriched fresh water the development of PRL cells was significantly retarded, but such an effect was not observed in fish placed in Ca2+-enriched fresh water. These experiments show that in C. whitei the development and activity of PRL cells are influenced by changes in environmental osmolarity and not by changes in ambient Ca2+-concentration.  相似文献

Summary In the annual cyprinodont Cynolebias whitei the cell types responsible for the increase of pituitary growth at the onset of maturation and for pituitary hyperplasia in old specimens were identified as gonadotropic cells and thyrotropic cells, respectively. The gonadotropic cells showed a high affinity to anti-carp -GTH serum, both at light- and electron-microscopical levels. The allometric relation of total gonadotropic cell volume to body length, determined for fish from six weeks up to six months of age, showed no inflections. Therefore pituitary growth in maturing fish may be partly a result of proliferation of gonadotropes, although gonadotropic cells do not contribute to pituitary hyperplasia in old fish. Thyrotropic cells showed a weak affinity to anti-carp -GTH serum at light-microscopical level. Under the electron microscope thyrotropic cells displayed signs of activation in maturing fish and signs of proliferation in old fish. The allometric relation of thyroid gland volume to body length paralleled that of pituitary volume to body length. Histologically the thyroid gland showed signs of inactivity in adult fish and of hyperplasia in old fish. The possibility, that gonadal maturation, pituitary thyrotropic activity, and growth of the thyroid in maturing fish are related through the inhibitory action of gonadal steroids on thyroid hormone release, is discussed. Pituitary hyperplasia in old fish is the result of proliferation of thyrotropic cells. Similar hyperplasia of pituiary and thyroid glands was observed in old Nothobranchius korthausae.  相似文献

Summary Changes in the prolactin cells of the euryhaline marine teleosts Gillichthys mirabilis and Platichthys stellatus were studied by light and electron microscopy after transfer from sea water to fresh water. In seawater fish the secretory granules were smaller and the cellular organelles poorly developed. Within 3 hours after transfer to fresh water, the prolactin cells of Gillichthys exhibited definite functional activation: exocytosis of granules and development of rough endoplasmic reticulum (RER), Golgi systems and mitochondria. Concurrently, plasma sodium fell from about 172 meq/l to about 133 meq/l.As adaptation to fresh water progressed, prolactin cells of Gillichthys showed greater prominence of cellular organelles but granule storage was not detected even 10 days after transfer. Platichthys adapted to fresh water for 10 days showed RER in an expanded state containing irregular electron-dense material which was not seen in Gillichthys. Plasma sodium levels were much lower than in the controls. These results were in contrast to those obtained from euryhaline freshwater fishes such as Poecilia and Oryzias. Although the prolactin cells of euryhaline marine fish exhibited intense secretory activity when transferred into fresh water, the secreted prolactin per se appeared to be insufficient to maintain plasma sodium at seawater levels.We thank Dr. R. The present study examined the effect of salinity and temperature on the rate of oxygen consumption and total body osmolality of the triclad turbellarian Procerodes littoralis, a common marine flatworm normally found in areas where freshwater streams run out over intertidal areas. Extremes in environmental factors encountered by P. littoralis were recorded at the study site. These were salinity (0-44 psu), temperature (2.7-24.9 °C) and oxygen concentration (2.8-16.1 mg l−1). Respirometry experiments showed minimal oxygen consumption rates at the salinity extremes encountered by the study species (0 and 40 psu). Further experiments showed relatively constant oxygen consumption rates over the temperature range 5-20 °C and elevated consumption rates at temperatures above 25 °C. Total body osmolality of P. littoralis increased with increasing salinity. The study illustrates how a marine flatworm uses integrated physiological and behavioural mechanisms to successfully inhabit an environment that is predominantly freshwater for up to 75% of the tidal cycle.  相似文献

Hypophysectomy resulted in a loss of ability of the euryhaline salmonid, Salmo trutta to survive in fresh water. The mean survival time was 4–5 days. Maintenance in a medium containing 5 mM calcium increased the mean survival time to 8 days while 10 mM decreased it. Injection of 0–21.U./gm prolactin enabled hypophysectomized fish to survive the 2-week duration of the experiment.
High environmental calcium, or pre-adaptation to a medium of high calcium, increased salinity tolerance of the brown trout probably by promoting a quick return of plasma electrolyte concentration to normal after transfer to sea water.  相似文献

Mechanosensitive (MS) ion channels are activated by mechanical stress and then transduce this information into electrical signals. These channels are involved in the growth, development and response to environmental stress in higher plants. Detailed analyses of the electrophysiology in higher plants are difficult because such plants are composed of complex tissues. The large cells of the charophytes facilitate electrophysiological measurements and allow us to study MS ion channels at the level of single cells. We draw parallels between the process of touch-perception in freshwater Chara, and the turgor-regulating response to osmotic shock in salt-tolerant Lamprothamnium. In terms of electrophysiology, these responses can be considered in three stages: (1) stimulus perception, (2) signal transmission and (3) induction of response. In Chara the first stage is due to the receptor potential (RPD), a transient depolarization with a critical threshold that triggers action potentials, which are responsible for stages (2) and (3). Receptor potentials are generated by MS ion channels. Action potentials involve a transient influx of Ca(2+) to the cytoplasm, effluxes of K(+) and Cl(-) and a temporary decrease of turgor pressure. Reducing cell turgor increases sensitivity to mechanical stimulation. In Lamprothamnium, a hypotonic shock produces an extended depolarization that resembles an extended RPD and is responsive to osmotic rather than ionic changes. Like the action potential, a critical threshold depolarization triggers Ca(2+) influx, opening of Ca(2+)-sensitive Cl(-) channels and K(+) channels; effluxes that last over an hour and result in turgor regulation. These processes show us, in primal form and at the level of single cells, how mechanoperception occurs in higher plants. Recent progress in research into the role of MS ion channels in the freshwater and salt-tolerant Characeae is reviewed and the relevance of these findings to plants in general is considered.  相似文献

The relationship of brain and body weights can be expressed in log-log regression: log (brain weight) = log (A) + B log (body weight). To investigate further the weights' similarity, gestation length and brain and body weights were determined from the literature for 46 primate genera. The results of allometric and path regression analyses suggest that the relationship between brain and body weights may not be mainly pleiotropic in the order Primates. The correlation between brain and body weights appears to be due to epigenetic factors in hyperplastic growth related to time constraint by gestation length and to energy utilization limitations imposed by metabolic rate.  相似文献

A selected Glycine max (L.) salt-tolerant calluscell line (R100) was significantly more tolerant to salt than a salt-sensitiveline (S100) during exposure to salt stress. Growth (Fresh and Dry weights) ofthe R100 cell line declined significantly at NaCl concentrations greater than 75mM, while growth of the S100 cell line was already impaired at 25mM NaCl. Levels of Na+ and Cl inthe callus were elevated as the salt concentration increased, whileK+, Ca2+ and Mg2+ levels weremarkedly reduced. The lower s reduction and Na+accumulation found in the S100 callus corresponded with the higher callusdehydration during salinity. Calli grown on Miller's basal medium weresupplied with 100 mM NaCl for 12 days and then supplied with mediumwithout NaCl to relieve salinity stress. The Na+ andCl content decreased in both R100 and S100 cell lines duringthe first 24 h and reached normal levels four days after transferto the normal medium. This lower concentration was maintained until the end ofthe experiment. Concurrently, the K+ content andK+/Na+ ratio increased sharply and reached theirhighest levels within 24 h in both salt-sensitive and salt-tolerantcell lines. These data suggest that the inhibitory effects of salinization ongrowth and accumulation of potentially toxic ions (Na+,Cl) can be readily reversed when salinity is relieved.  相似文献

The length of the posterolateral spines in the rotifer, Brachionus calyciflorus, is variable. The effect of the length of these spines on the clearance rates of B. calyciflorus was examined under laboratory conditions using radioactively-labelled yeast (Rhodotorula) and bacteria (Aerobacter). The hypothesis that individuals with longer spines have higher clearance rates than those of similarly-sized individuals with shorter spines was rejected. The effect of body length (L) on the clearance rates (C) of short-spined and long-spined animals was also examined. Statistically significant linear regressions of the form log10 C = aL + b, were derived for both types. The slopes of both regressions were similar and suggest that clearance rate in B. calyciflorus increases as a cube function of body length.  相似文献

A population of bullhead Cottus gobio inhabiting a mountainous river network, was examined for the influence of maternal condition (total length, L T, and age class in years) and environmental factors (water temperature and fish density) on fecundity, egg size and net annual reproductive effort ( E R, defined as the ratio of total egg production mass to body mass) within several stocks. Regardless of location within the drainage network, bullhead reproduction was synchronous, with only a single batch and fecundity ranging from 69 to 896 eggs per female. As with L T fecundity increased with age, although the difference between age classes 5 and 6 years was smaller than between other age classes. Egg diameter also increased with age from age classes 2 to 4 years, but then tended to decrease for age classes 5–6 years. Multiple stepwise regression was used to examine the effect of female L T, water temperature and fish densities on bullhead egg production for each of the four age classes (2, 3, 4 and 5–6 years) separately. When statistically significant, water temperature always had a positive effect and fish densities a negative effect, on fecundity and egg size. The L T always had a positive effect on fecundity for all the age classes. In contrast, L T had a positive effect on egg size and E R only for female reaching maturity (2 years), whereas intermediate aged females (3–4 years) were insensitive and the oldest fish showed a negative response.  相似文献

This paper describes the analysis of over 15,000 weights of Mallard caught for ringing at Borough Fen Decoy, Northamptonshire, during a 13 year period. Weights are corrected to give condition indices by using wing length to give an indication of body size. The weight of pectoral muscles varies with body weight but as the fat stores increase in weight the proportion of body weight made up of flight muscles decreases. Body weight corrected for size gives a better indication of the bird's condition (defined as its fitness for its present and future needs) than the relative weight of the pectoral muscles or the shape of the breast.
The relationships between food availability and weather, and condition are examined. Only occasionally can a drop in condition following hard weather be detected; the birds normally emigrate to the coast after a few days of adverse conditions. There is a relationship between the condition of Mallard in autumn and the amount of grain left on their stubble feeding grounds after harvest. The implications of variations in winter condition on survival and breeding success are briefly discussed.  相似文献

There is assumption about active role of immune modulators in cell death process. The involvement of interferon-alpha and cycloferon in apoptosis regulation of hypothalamic neurons of mice during stress and aging was studied. We determined the expression of apoptosis markers (Bcl-2, Mcl-1, Bax) in comparison with apoptosis level. We have found that immune modulators suppress activity of nonapeptidergic neurons. Thus, interferon-alpha treatment reduces synthesis of Bcl-2; cycloferon treatment inhibits expression of Bax and Bcl-2. So the role of immune modulators in neuron apoptosis depends on the stage of ontogenesis and type of immune modulator. Cycloferon is able to reduce the level of age-dependent apoptosis of neurons in aging, but under stress condition both interferon-alpha and cycloferon act as protectors of cell death.  相似文献

Prolactin (PRL) cell activity was investigated in eels kept in fresh water (FW), deionized water (DW) supplemented or not with Ca (2 mM), in Ca-enriched FW (10 mM), in normal (Ca 3.4 mM) or Ca-free 1/3 sea water (SW), and in SW (Ca 10.2 mM) or Ca-free SW (Ca 0.15 mM). Light-microscopic studies, including measurement of the nuclear area and cell height, showed that PRL cell activity, reduced in DW, is not affected by Ca supplementation. Activity is reduced in Ca-enriched FW, in 1/3 SW and in SW, conditions inducing an increase in the plasma sodium level. The lack of calcium in saline environments partly suppresses the nuclear atrophy occurring in SW. There is no significant correlation between external or total plasma calcium concentration and PRL cell activity. In artificial Ca-free SW, eels show a rapid increase in plasma osmolarity and sodium levels; there is a significant negative correlation between these two plasma values and the nuclear area or cell height of PRL cells. As in some other teleosts, plasma osmolarity and plasma sodium seem to play a more important role than external or internal calcium in controlling PRL secretion. This correlation is not apparent in eels kept in SW, having unstimulated PRL cells but active calcium-sensitive (Ca-s) cells in the pars intermedia.  相似文献

The effect of thermal environment on condition factor was examined for six different size-classes of Atlantic cod Gadus morhua fed to satiation. A weight–length relationship for 8 to 1303 g fish reared at 4–20°C indicated an allometric growth ( W  =  aL b , a  = 0.0045, b  = 3.257) of cod. Changes in relative condition factor ( K rel) with temperature were described with a second order polynomial. The most pronounced effect of temperature on body condition was found in the smallest size-classes, but the curves flattened with increased size. Temperature had size-dependent effects on the relative condition factor obtained from an overall weight–length relationship for all fish in the experiment, i.e. K rel increased with weight at 4°C, but decreased with weight at 16 and 20°C. K rel remained high for most size-classes at 8 and 12°C. The slopes ( b -values) of the weight–length relationships decreased linearly with temperatures from 4 to 16°C.  相似文献

Social stressors such as depressed maternal care and family conflict are robust challenges which can have long-term physiological and behavioral effects on offspring and future generations. The current study investigates the transgenerational effects of an ethologically relevant chronic social stress on the behavior and endocrinology of juvenile and adult rats. Exposure to chronic social stress during lactation impairs maternal care in F0 lactating dams and the maternal care of the F1 offspring of those stressed F0 dams. The overall hypothesis was that the male and female F2 offspring of stressed F1 dams would display decreased social behavior as both juveniles and adults and that these behavioral effects would be accompanied by changes in plasma corticosterone, prolactin, and oxytocin. Both the female and male F2 offspring of dams exposed to chronic social stress displayed decreased social behavior as juveniles and adults, and these behavioral effects were accompanied by decreases in basal concentrations of corticosterone in both sexes, as well as elevated juvenile oxytocin and decreased adult prolactin in the female offspring. The data support the conclusion that social stress has transgenerational effects on the social behavior of the female and male offspring which are mediated by changes in the hypothalamic–pituitary–adrenal axis and hypothalamic–pituitary–gonadal axis. Social stress models are valuable resources in the study of the transgenerational effects of stress on the behavioral endocrinology of disorders such as depression, anxiety, autism, and other disorders involving disrupted social behavior.  相似文献

Previous investigations have shown that mammalian prolactin has a pronounced effect on lipid deposition in teleost fishes (e.g. Meier et al., 1971; de Vlaming & Sage, 1972). Prolactin can be either lipogenic or lipolytic depending on when during the day it is injected. Studies were initiated to determine whether the diurnal effects of prolactin on lipid metabolism in Fundulus similis vary with season or photoperiod acclimation. Data presented here indicate that the diurnal rhythm of response to prolactin persists in the pre-spawning and post-spawning seasons if F. similis is maintained on a long photoperiod. When animals are acclimated to a short photoperiod, treatment with mammalian prolactin either early or late in the photoperiod causes lipid depletion. Homogenates of the portion of the pituitary containing only prolactin cells cause lipid depletion in F. similis when injected early during a long photoperiod, but stimulate lipogenesis if injected late in the photoperiod; these results imply that fish prolactin may be involved in lipid metabolism. Furthermore, prolactin activity in the pituitary of F. similis varies diurnally. In vitro studies of lipid metabolism in Notemigonous crysoleucas and Ictalurus natalis suggest that the liver may be one of the target organs for prolactin.  相似文献

A single injection of 2.5 mg perphenazine (PH)/kg body wt to rats on the day of estrus (day 0) did not result in increased serum progesterone 24 hr later. Continued daily injections, however, resulted in a 2.5-fold increase in serum progesterone between days 1 and 3 and a 1.6-fold increase between days 3 and 5 to a final concentration of 58 plus or minus 4 ng/ml on day 5 in serially anesthetized and bled rats. Neither daily administration of 5.0 nor 10.0 mg PH/kg body wt to rats subjected to the stressful conditions of this regimen resulted in further increases in serum progesterone, but the 5.0 mg dose of PH in unstressed rats bled only on day 5 resulted in a highly significant increase in serum progesterone to 110 plus or minus 7 ng/ml. In unstressed rats the increase in serum progesterone over control values after five daily injections of 2.5 mg PH/kg body wt could be attributed to decreased 20alpha-reduction of progesterone, but when the dose of PH was increased to 5.0 mg/kg, a highly significant increase in both progesterone and total progestins occurred indicating that prolactin can increase steroidogenesis as well as reduce 20alpha-hydroxysteroid dehydrogenase activity. After inhibition of ovulation, the 5.0 mg daily dose of PH resulted in serum progesterone of only 25 plus or minus 8 ng/ml on day 5 in unstressed rats. Thus, serum progesterone in ovulating rats treated with PH originated primarily in the corpora lutea. Perphenazine, 5.0 mg/kg, administered only on estrus and the first day of diestrus was sufficient to induce pseudopregnancy of 14.5 plus or minus 1.6 days. No evidence for gonadotropin stimulation of the ovaries of any rats was observed. The effect of stress on the progesterone response was not mimicked by administration of cortisol acetate and is assumed to be medicated by suppression of prolactin secretion.  相似文献
