Pesticide resistance has developed as a result of long‐term and extensive use of chemical pesticides. Essential oils from aromatic plants may provide a new and safe alternative to conventional insecticides. In this study, the insecticidal activities of the essential oil of Melaleuca alternifolia and their chemical constituents against Helicoverpa armigera Hubner were investigated, and the underlying mechanisms were studied. The essential oil showed distinct antifeedant (AFC50 = 8.93 mg/ml) and good contact (LD50 = 50.28 μg/larva) activities against H. armigera at 24 hr. Ten chemical components were identified using a gas chromatograph/mass spectrometer, and mainly included terpinen‐4‐ol (40.09%), γ‐terpinene (21.85%), α‐terpinene (11.34%), α‐terpineol (6.91%), α‐pinene (5.86%), terpinolene (3.24%) and 1,8‐cineole (1.83%). Among them, five components were determined and results showed that these constituents possessed obvious antifeedant activities. The activities of acetylcholinesterase and glutathione S‐transferase were notably inhibited by the essential oil, as compared with the control, with strong dose‐ and time‐dependent effects. The results provide a basis for their development and utilization in the control of insects in the future.  相似文献

Bark beetles have caused extensive damage to forests in central Sweden during the past decade, and the four‐eyed spruce bark beetle, Polygraphus poligraphus, seems to be involved. However, its role in these bark beetle outbreaks is still not clear. The purpose of this study was to develop an efficient pheromone bait for P. poligraphus, which would make it possible to study the species more carefully and thereby contribute to protect exposed forests in an environmentally friendly way. Three field studies were conducted in 2015, 2016 and 2018 in Medelpad, county of Västernorrland, Sweden. The pheromone of P. poligraphus, (?)‐terpinen‐4‐ol, was tested at different release rates and in different enantiomeric purities, to find the most attractive formulation for the beetles. It was also tested in combination with racemic frontalin, a compound which has previously been shown to produce a synergistic effect together with (?)‐terpinen‐4‐ol of low enantiomeric purity; 52% ee. Other compounds, chosen based on responses from electroantennographic studies, were also tested in an attempt to find additional attractants and repellents for P. poligraphus. The most attractive treatment tested was enantiomerically pure (?)‐terpinen‐4‐ol (99% ee). When the enantiomeric purity was lower (50% ee), the trap catches was lowered to levels comparable to the catches for unbaited control traps. A strong synergistic effect with frontalin was observed for (?)‐terpinen‐4‐ol of low enantiomeric purity (50% ee) but not for the enantiomerically pure compound (99% ee). The release rate of (?)‐terpinen‐4‐ol (99% ee) was shown to be an important factor. For the combination of frontalin and (?)‐terpinen‐4‐ol (50% ee), the attraction seemed strongest when (?)‐terpinen‐4‐ol was released at a higher rate than frontalin. An interesting and novel result was that a repellent compound, α‐terpineol, was identified in our studies. Our results from field studies and electroantennography recordings also indicate that (+)‐terpinen‐4‐ol is a repellent for P. poligraphus.  相似文献

Melaleuca quinquenervia (Cav) S.T. Blake (broadleaf paperbark) is an Australian tree that has become a serious weed in many places around the world. Two insects Oxyops vitiosa (the melaleuca weevil), and Boreioglycaspis melaleucae (the melaleuca psyllid), which were introduced to Florida as part of a biological control programme, have been very effective in reducing survival and reproduction of this weed. There are two terpene chemotypes of M. quinquenervia; one rich in the sesquiterpene E-nerolidol whereas the other is rich in viridiflorol. Viridiflorol is a strong feeding deterrent for the melaleuca weevil and retards larval development. The larvae therefore avoid the viridiflorol-rich chemotype, in contrast, female melaleuca psyllids prefer to oviposit on these leaves. To identify the molecular basis of these preferences, we isolated and characterised two terpene synthases from the viridiflorol-rich chemotype, both of which utilise farnesyl pyrophosphate and have the same product profile. Chemotypic variation in terpenes in M. quinquenervia is under strong genetic control and the reproductive potential of each chemotype is limited by a different insect. These insects could, therefore, be selective agents for the maintenance of chemotypic variation in M. quinquenervia.  相似文献

Recently, the distribution of the Mediterranean cedar bark beetle, Phloeosinus aubei Perris (Coleoptera: Scolytidae), has expanded to Central Europe. Reported mostly on cypress in the Mediterranean area, potential host plants in the invaded range include other scale‐leafed conifers, such as cultivars of arborvitae, Thuja occidentalis L. To reveal potential kairomonal cues for P. aubei, volatiles of T. occidentalis were collected and analysed by gas chromatography with electroantennographic detection (GC‐EAD). Assignments of chemical structures of antennally active components were carried out by gas chromatography linked to mass spectrometry (GC‐MS) using authentic reference samples. Antennal responses to synthetic samples of the identified compounds were studied by electroantennography (EAG), with antennae of female and male P. aubei. GC‐EAD analysis of head space volatiles of T. occidentalis revealed 22 antennally active compounds, of which 21 were identified. The most abundant components were α‐ and β‐thujone, fenchone, camphor, terpinen‐4‐ol, bornyl acetate and α‐terpinyl acetate, all of which are oxygenated monoterpenes. When EAG activities of synthetic samples were compared, the most intensive responses from female antennae were elicited by a mixture of α‐ and β‐thujone, followed by (–)‐terpinen‐4‐ol, (+)‐camphor, cis‐4‐thujanol, (+)‐sabina ketone, (+)‐terpinen‐4‐ol, isopulegone, (–)‐fenchone, borneol, (3Z)‐hexen‐1‐ol, (–)‐1‐octen‐3‐ol and (+)‐sulcatol. Male antennae responded the most to (–)‐terpinen‐4‐ol and cis‐4‐thujanol followed by the mixture of α‐ and β‐thujone. The next highest responses were elicited by (+)‐camphor, borneol, (+)‐terpinen‐4‐ol, (+)‐sulcatol and (+)‐sabina ketone. Striking differences were found between responses to the enantiomers of fenchone, sulcatol and 1‐octen‐3‐ol, whereas responses to the enantiomers of terpinen‐4‐ol did not differ significantly from each other. Several antennally active volatiles of T. occidentalis have also been reported from cypress and various other members of the Cupressaceae, suggesting that the sensory apparatus of P. aubei may recognize the shared components, which may enable rapid adaptation to new hosts in the invaded areas.  相似文献

Aims: The aim of this study was to compare both the antimicrobial activity of terpinen‐4‐ol and tea tree oil (TTO) against clinical skin isolates of meticillin‐resistant Staphylococcus aureus (MRSA) and coagulase‐negative staphylococci (CoNS) and their toxicity against human fibroblast cells. Methods and Results: Antimicrobial activity was compared by using broth microdilution and quantitative in vitro time‐kill test methods. Terpinen‐4‐ol exhibited significantly greater bacteriostatic and bactericidal activity, as measured by minimum inhibitory and bactericidal concentrations, respectively, than TTO against both MRSA and CoNS isolates. Although not statistically significant, time‐kill studies also clearly showed that terpinen‐4‐ol exhibited greater antimicrobial activity than TTO. Comparison of the toxicity of terpinen‐4‐ol and TTO against human fibroblasts revealed that neither agent, at the concentrations tested, were toxic over the 24‐h test period. Conclusions: Terpinen‐4‐ol is a more potent antibacterial agent against MRSA and CoNS isolates than TTO with neither agent exhibiting toxicity to fibroblast cells at the concentrations tested. Significance and Impact of the Study: Terpinen‐4‐ol should be considered for inclusion as a single agent in products formulated for topical treatment of MRSA infection. However, further work would initially be required to ensure that resistance would not develop with the use of terpinen‐4‐ol as a single agent.  相似文献

Abstract. 1. Plants may compensate for the effects of herbivory, especially under favourable growing conditions, limited competition, and minimal top‐down regulation. These conditions characterise many disturbed wetlands dominated by introduced plants, implying that exotic, invasive weeds in these systems should exhibit strong compensatory responses. 2. The Australian native Melaleuca quinquenervia is highly invasive in the Florida Everglades, U.S.A., where it experiences limited competition or herbivory from native species, making it a likely candidate for compensation. The introduced biological control agent Oxyops vitiosa feeds exclusively on the seasonal flushes of developing foliage at branch apices, which represents ≈15% of the total foliar biomass. 3. The hypothesis that M. quinquenervia compensates for folivory by O. vitiosa was tested in a series of field‐based experiments. Trees experiencing folivory over four consecutive years maintained similar levels of foliar biomass after attack yet possessed twice the number of leaf‐bearing terminal stems as undamaged trees. The biomass of these stems was similar among attacked and unattacked trees, indicating that herbivore‐damaged trees produce greater quantities of smaller terminal branches. However, undamaged trees were 36 times more likely to reproduce than herbivore‐damaged trees. 4. In a separate herbivore exclusion study, a single bout of herbivory on previously undamaged M. quinquenervia trees caused an 80% reduction in reproductive structures the following season. Herbivore‐damaged trees also possessed 54% fewer fruits than undamaged trees. An increase in the herbivory frequency (two bouts per year) or magnitude (100% simulated herbivory) did not result in a further reduction in fitness. 5. It has been concluded that M. quinquenervia partially compensates for herbivory by producing new stems and replacing foliage, but this compensation results in a substantial reduction in reproduction.  相似文献

The essential‐oil compositions of leaves, flowers, and rhizomes of Alpinia galanga (L.) Willd ., Alpinia calcarata Rosc ., Alpinia speciosa K. Schum. , and Alpinia allughas Rosc . were examined and compared by capillary GC and GC/MS. Monoterpenoids were the major oil constituents identified. 1,8‐Cineole, α‐terpineol, (E)‐methyl cinnamate, camphor, terpinen‐4‐ol, and α‐ and β‐pinenes were the major constituents commonly distributed in leaf and flower essential oils. The presence of endo‐fenchyl acetate, exo‐fenchyl acetate, and endo‐fenchol was the unique feature of rhizome essential oils of A. galanga, A. calcarata, and A. speciosa. On contrary, the rhizome oil of A. allughas was dominated by β‐pinene. Significant qualitative and quantitative variations were observed in essential‐oil compositions of different parts of Alpinia species growing in subtemperate and subtropical regions of Northern India. Cluster analysis was performed to find similarities and differences in essential‐oil compositions based on representative molecular skeletons. Monoterpenoids, viz., 1,8‐cineole, terpinen‐4‐ol, camphor, pinenes, (E)‐methyl cinnamate, and fenchyl derivatives, were used as chemotaxonomic markers.  相似文献

The purpose of this study was to evaluate the anaesthesia effects of eugenol and MS‐222 sedatives applied on juvenile Lateolabrax maculatus during simulated transport. In experiment 1, the juveniles were divided into two groups, with seven concentrations tested on each group (eugenol [4, 6, 8, 10, 12, 14 and 16 mg/L] and MS‐222 [20, 30, 40, 50, 60, 70 and 80 mg/L]). Induction and recovery times were recorded. The time for anaesthesia was shortened, and the time for complete recovery was prolonged as the anaesthetic concentration increased. The optimal transport concentration for each anaesthetic tested was 6 mg/L of eugenol and 30 mg/L for MS‐222. In experiment 2, the 5‐hr simulated transport test showed that the survival rate of L. maculatus juveniles with anaesthesia was 100%, and without anaesthesia, survival was 60%. After 24 hr of recovery following transport, the fish showed 100% survival for the group with added anaesthetic and 40% for the group without added anaesthetic. Compared to the non‐anaesthetized groups, the anaesthetized transport groups showed significant increases in the levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) (p < 0.05). The levels of AST, ALT and alkaline phosphatase (AKP) were significantly higher in the MS‐222 transport group than in the eugenol transport group (p < 0.05). The levels of AKP were significantly higher in the non‐anaesthetized transport group than in the anaesthetized group (p < 0.05). According to the present experiment results, eugenol was an efficient anaesthetic in L. maculatus, and we recommend eugenol instead of MS‐222 as an anaesthetic for the short‐time transport of L. maculatus.  相似文献

This study deals with the variation in the yield and composition of Lebanese Origanum syriacum L. essential oil (EO) according to harvesting time, drying methods used, and geographical location. Plant material was harvested twice a month all over 2013 and 2014 from Qartaba and Achkout located at high altitude and from Byblos at low altitude. EOs of the aerial parts were obtained by hydrodistillation. The highest yields were obtained at full flowering stage and slightly reduced after flowering. The GC/MS analysis revealed the presence of 50 components representing 90.49 – 99.82%, 88.79 – 100%, and 95.28 – 100% of the total oil extracted from plants harvested from Qartaba, Achkout, and Byblos, respectively. The major components in the oils were: carvacrol (2.1 – 79.8%), thymol (0.3 – 83.7%), p‐cymene (2.8 – 43.8%), thymoquinone (0.4 – 27.7%), γ‐terpinene (0.4 – 10.0%), octan‐3‐ol (0.3 – 4.9%), caryophyllene oxide (0.2 – 4.7%), oct‐1‐en‐3‐ol (0.3 – 3.7%), β‐caryophyllene (0.7 – 3.2%), cis‐sabinene hydrate (0.1 – 2.8%), terpinen‐4‐ol (0.1 – 2.8%), and α‐terpinene (0.2 – 2.2%). Independent components analysis (ICA) revealed that two groups were discriminated, reflecting compositional differences in the EOs profiles of the Lebanese oregano samples: O. syriacum grown in Qartaba and Achkout belongs to carvacrol chemotype, while O. syriacum grown in Byblos belongs to thymol chemotype. The flowering phase was the most productive period in terms of yield, bringing marked changes in the EO composition by increasing the amounts of carvacrol or thymol, and decreasing those of thymoquinone and p‐cymene.  相似文献

Abstract

Melaleuca styphelioides is considered as medicinal plant. This study was carried out to evaluate for the first time the phytochemical composition and to compare the antifungal activities of essential oil (EOs), methanol and aqueous extracts of M. styphelioides Sm. leaves against three fungi (Aspergillus niger, Rhizopus nigricans and Penicillium digitatum). A total of 10 components of the EO were identified, with the principal compound being methyl eugenol (87.2%). Results of the phytochemical analysis of leaves extract exhibited the presence of different phytoconstituents (phenolic compounds, flavonoids, tannins and anthocyanins). Volatile and non-volatile extracts were found to express dose-dependent inhibition against all tested fungi. Indeed, the EO oil showed significant inhibition of fungal growth and the IC50 was 2.08?µL/mL for A. niger indicating that M. styphelioides leaf EO was particularly effective against this pathogen. The most susceptible species for the aqueous extract was P. digitatum (IC50= 9.54?mg/mL) whereas R. nigricans was found to be more susceptible to the methanolic extract (IC50= 8.31?mg/mL). Thus, the EO and aqueous as well as methanol extracts of M. styphelioides leaves possess antifungal activity and hence, it can be suggested for use in the food or pharmaceutical industries as an alternative to chemical preservatives.  相似文献

The present study was focused on exploring the presence of active compounds in Origanum majorana essential oil (OmEO), and its various knock‐down effects against the rice moth, Corcyra cephalonica. GC–MS analysis detected the existence of major compounds such as monoterpenes, cis‐β‐terpineol and terpinen‐4‐ol with the total proportion of 52.16%. Fumigant toxicity against adult and larvae was calculated with an LC50 value of 11.31 and 49.83 μL/L air, respectively. The contact toxicity against adult, pupa, larvae and eggs was observed with LC50 value 2.54, 0.95, 2.78, and 0.49 μL/L, respectively. Furthermore, the influential repellent behavior against adults has been observed. Acetylesterase (AChE) inhibition activity of OmEO was observed against adult and larvae of C. cephalonica with an IC50 value of 35.89 and 118.54 μL/mL, respectively. Moreover, computational docking study revealed the binding affinity of Cis‐β‐terpineol and terpinen‐4‐ol towards the active binding sites of AChE. On the other hand, Fluorescence‐assisted cytometry and comet assay confirmed the cytotoxic and genotoxic effect of OmEO at various concentrations on C. cephalonica. Altogether, the results showed the knock‐down effect of OmEO against C. cephalonica, and it could be a potential biocontrol measure against the stored product pest.  相似文献

Aims

To determine the herd prevalence of Enterobacteriaceae producing CTX‐M‐type extended‐spectrum β‐lactamases (ESBLs) among 381 dairy farms in Japan.

Methods and Results

Between 2007 and 2009, we screened 897 faecal samples using BTB lactose agar plates containing cefotaxime (2 μg ml?1). Positive isolates were tested using ESBL confirmatory tests, PCR and sequencing for CTX‐M, AmpC, TEM and SHV. The incidence of Enterobacteriaceae producing CTX‐M‐15 (= 7), CTX‐M‐2 (= 12), CTX‐M‐14 (= 3), CMY‐2 (= 2) or CTX‐M‐15/2/14 and CMY‐2 (= 4) in bovine faeces was 28/897 (3·1%) faecal samples. These genes had spread to Escherichia coli (= 23) and three genera of Enterobacteriaceae (= 5). Herd prevalence was found to be 20/381 (5·2%) dairy farms. The 23 E. coli isolates showed clonal diversity, as assessed by multilocus sequence typing and pulsed‐field gel electrophoresis. The pandemic E. coli strain ST131 producing CTX‐M‐15 or CTX‐M‐27 was not detected.

Conclusions

Three clusters of CTX‐M (CTX‐M‐15, CTX‐M‐2, CTX‐M‐14) had spread among Japanese dairy farms.

Significance and Impact of the Study

This is the first report on the prevalence of multidrug‐resistant CTX‐M‐15–producing E. coli among Japanese dairy farms.  相似文献

Essential oil extracted from nutmeg seeds (Myristica fragrans Houtt .) by hydrodistillation was subjected to GC/MS and GC analysis. A total of 27 constituents were identified, of which eugenol (19.9%), methylisoeugenol (16.8%), methyleugenol (16.7%), sabinene (11.8%), and terpinen‐4‐ol (8.5%) were the major components. The essential oil was tested against Lasioderma serricorne for insecticidal and repellent activity, the LD50 value at the end of 24 h exposure period was 19.3 μg/adult. Six active compounds were isolated by bioassay‐guided fractionation. They were identified as eugenol ( 1 ), methyleugenol ( 2 ), methylisoeugenol ( 3 ), elemicin ( 4 ), myristicin ( 5 ), and safrole ( 6 ). Among these isolates, 4 showed the strongest contact toxicity against L. serricorne adults with an LD50 value of 9.8 μg/adult. Repellency of crude oil and active compounds were also determined. Compounds 1, 2, 4 , and 5 were strongly repellent against the cigarette beetle and exhibited the same level of repellency compared with the positive control, DEET. The results indicate that the essential oil of M. fragrans and its active constituents have potential for development as natural insecticides and repellents to control L. serricorne.  相似文献

The aerial parts of Lantana camara L. were collected from three different geographical locations: Artemisa (Cuba), Biratnagar (Nepal), and Sana'a (Yemen). The essential oils were obtained by hydrodistillation and analyzed by gas chromatography/mass spectrometry. A cluster analysis of 39 L. camara essential oil compositions revealed eight major chemotypes: β‐caryophyllene, germacrene D, ar‐curcumene/zingiberene, γ‐curcumen‐15‐al/epiβ‐bisabolol, (E)‐nerolidol, davanone, eugenol/alloaromadendrene, and carvone. The sample from Cuba falls into the group dominated by (E)‐nerolidol, the sample from Nepal is a davanone chemotype, and the sample from Yemen belongs to the β‐caryophyllene chemotype. The chemical composition of L. camara oil plays a role in the biological activity; the β‐caryophyllene and (E)‐nerolidol chemotypes showed antimicrobial and cytotoxic activities.  相似文献

Novel benzoxazole derivatives were synthesized, and their antitubercular activity against sensitive and drug‐resistant Mycobacterium tuberculosis strains (M. tuberculosis H37Rv, M. tuberculosis sp. 210, M. tuberculosis sp. 192, Mycobacterium scrofulaceum, Mycobacterium intracellulare, Mycobacterium fortuitum, Mycobacterium avium, and Mycobacterium kansasii) was evaluated. The chemical step included preparation of ketones, alcohols, and esters bearing benzoxazole moiety. All racemic mixtures of alcohols and esters were separated in Novozyme SP 435‐catalyzed transesterification and hydrolysis, respectively. The transesterification reactions were carried out in various organic solvents (tert‐butyl methyl ether, toluene, diethyl ether, and diisopropyl ether), and depending on the solvent, the enantioselectivity of the reactions ranged from 4 to >100. The enzymatic hydrolysis of esters was performed in 2 phase tert‐butyl methyl ether/phosphate buffer (pH = 7.2) system and provided also enantiomerically enriched products (ee 88‐99%). The antitubercular activity assay has shown that synthesized compounds exhibit an interesting antitubercular activity. Racemic mixtures of alcohols, (±)‐4‐(1,3‐benzoxazol‐2‐ylsulfanyl)butan‐2‐ol ((±)‐ 3a ), (±)‐4‐[(5‐bromo‐1,3‐benzoxazol‐2‐yl)sulfanyl]butan‐2‐ol ((±)‐ 3b ), and (±)‐4‐[(5,7‐dibromo‐1,3‐benzoxazol‐2‐yl)sulfanyl]butan‐2‐ol ((±)‐ 3c ), displayed as high activity against M. scrofulaceum, M. intracellulare, M. fortuitum, and M. kansasii as commercially available antituberculosis drug‐Isoniazid. Moreover, these compounds exhibited twice higher activity toward M. avium (MIC 12.5) compared with Isoniazid (MIC 50).  相似文献

Forty‐two essential oil samples were isolated from leaves of Xylopia rubescens harvested in three forests of Southern Ivory Coast. All the samples have been submitted to GC‐FID and the retention indices (RIs) of individual components have been measured on two capillary columns of different polarity. In addition, 20 oil samples, selected on the basis of their chromatographic profile, were also analyzed by 13C‐NMR and 24 components (78.0 – 92.4% of the whole compositions) have been identified. The content of the main components varied drastically from sample to sample: furanoguaia‐1,4‐diene (5.7 – 54.1%), furanoguaia‐1,3‐diene (1.1 – 10.5%), (8Z,11Z,14Z)‐heptadeca‐8,11,14‐trien‐2‐one (4.3 – 16.0%), and (E)‐β‐caryophyllene (1.7 – 17.3%). Hierarchical cluster and principal components analysis of the 42 oil compositions allowed the distinction of two well‐differentiated groups of unequal importance within the oil samples. Oil samples of the main group (Group II) contained mainly furanoguaia‐1,4‐diene (mean [M] = 43.1%; standard deviation [SD] = 3.2%) while furanoguaia‐1,3‐diene (M = 8.4%; SD = 0.9%) and (8Z,11Z,14Z)‐heptadeca‐8,11,14‐trien‐2‐one (M = 7.1%; SD = 1.5%) were present at appreciable contents. The composition of Group I was dominated by furanoguaia‐1,4‐diene (M = 17.0%; SD = 8.5%), (8Z,11Z,14Z)‐heptadeca‐8,11,14‐trien‐2‐one (M = 10.2%; SD = 2.4%) and (E)‐β‐caryophyllene (M = 9.5%; SD = 5.3%).  相似文献

An examination of the leaf oils of Melaleuca quinquenervia over its geographical range in Australia and Papua New Guinea has shown wide variation in chemical composition but only two major chemotypes. Chemotype 1 is comprised of E-nerolidol (74–95%) and linalool (14–30%) and is found from Sydney, north along the east coast of Australia to Selection Flat, New South Wales, with an isolated occurrence near Maryborough, Queensland. Two divisions occur in this chemotype which are based on the presence or absence of significant proportions of linalool (14–40%). Chemotype 2 contains 1,8-cineole (10–75%), viridiflorol (13–66%), α-terpineol (0.5–14%) and β-caryophyllene (0.5–28%) in varying proportions and order of dominance in the oils. It is found throughout the distribution of the species, from Sydney to Papua New Guinea and New Caledonia. Within chemotype 2 there appears to be a continuous spread of oil composition without formation of any further discrete divisions as in chemotype 1.Analyses have shown that M. quinquenervia trees that occur at latitudes south of 25°S have high oil yields (1–3% w/w%, fresh leaves) and comprise chemotypes 1 and 2. North of 25°S, however, chemotype 1 does not occur and oil yields amongst the Australian populations are uniformly low (0.1–0.2%).  相似文献
